Melanoma is an aggressive form of skin cancer known for developing into metastatic disease. Current clinical diagnostics, including medical imaging and tissue biopsy, provide a poor prognosis since the cancer is in the late stages of disease progression. In recent years, photoacoustic flow cytometry has allowed for the detection of circulating melanoma cells within patient blood samples in vitro. Although this method exploits the naturally-produced melanin within the cells, it has only successfully detected highly-pigmented melanoma cell lines. Approximately 1 in 50 Americans develop melanoma in their lifetime. Early detection of melanoma is pivotal to the survival of the patient, with a 99% survival rate for 5 or more years after an early diagnosis. Metastasis, or the spread of cancer, increases the chances of cancerous growth in other parts of the body. In this study, we propose the use of photoacoustic flow cytometry as a diagnostic of patient disease state, by closely monitoring the amount of circulating melanoma cells (CMCs) in a patient’s blood before and after chemotherapy treatments. Metastatic melanoma is the deadliest form of skin cancer, which is in part, attributed to its rapid aggression and lack of response to typical treatment methods. There are far too often cases where a lymph node biopsy does not detect the severity of the cancer, which in turn causes a lack of diagnosis until a mass can be visually detected on a scan, such as a PET, CT, or MRI. “Milia-like cysts (MLCs) are dermoscopic structures frequently observed in seborrheic keratoses(SKs), which are the most common type of skin lesions. Diverse appearances of these skin lesions make them difficult to differentiate from melanoma, a deadly type of skin cancer. Classified by size into two main groups, starry MLCs and cloudy MLCs, the presence of these structures in a skin lesion has been known to help differentiate benign lesions from melanoma. “Earlier detection of malignant melanoma can save lives. Automatic in-vivo methods of melanoma detection including smart-phone applications are now available, but specificity is low, i.e. too many benign lesions are detected as possible melanomas. Polypoids, elevated and rounded lesions are one class of benign lesions. These raised lesions with piecewise circular borders, predominantly intradermal nevi and cutaneous polyps (IDNs), are always benign. Variegated coloring present in polypoid lesions can lead physicians to perform an unneeded biopsy. This thesis discusses the research, experimental methods, and data gathered for the investigation of a novel method for the diagnosis of melanoma skin cancer. First, a background about human skin tissue is presented. Then, a detailed description of melanoma along with current diagnosis techniques and treatment options are presented. In the experimental methods, the electrical properties of several types of tissue were analyzed, the purpose of which was to discover if a tissue type can be distinguished by its electrical properties alone.
