Expression Optimization Of The Gst-Gfp Fusion Protein Through The Alteration Of Induction Conditions, Matthew J. Vaccaro

This research sought to determine which induction condition resulted in the greatest GST-GFP fusion protein expression. It will hopefully serve as a guide for future researchers trying to produce their own recombinant protein containing GST and GFP-tags. The CDNB Enzyme Assay was used to determine the quantity of GST-GFP fusion protein present and tested three variables: IPTG concentration, duration, and temperature of induction. The findings showed that IPTG concentration, temperature, and induction duration all had a significant impact on protein expression. Induction temperatures of 20 °C and 25 °C showed better protein expression at IPTG concentrations of 1.0 mM IPTG …

Advanced engineering materials are designed to display tensile-compressive asymmetry (TCA) and anisotropy to provide unique attributes to critical components necessary in the hot section of turbines. The never-ending chase for higher efficiencies, and with them, higher temperature gradients, intrinsically leads to more and more of these complex materials, like single crystal turbine blades, embedded within the turbine environment. This thesis focuses on the development, parameterization and optimization of a novel meso-scale pattern used to induce auxetic behavior, i.e., negative Poisson's ratio, at the bulk scale. Currently, the majority of auxetic structures are too porous to be utilized in conventional load-bearing applications. For others, manufacturing methods have yet to realize the meso-scale pattern. Consequently, new auxetic structures must be developed in order to confer superior thermo-mechanical responses to structures at high temperature. In the combustion zone of industrial- and aero- gas turbines, thermomechanical fatigue (TMF) is the dominant damage mechanism. Thermomechanical fatigue is a coupling of independent creep, fatigue, and oxidation damage mechanisms that interact and accelerate microstructural degradation. A mixture of intergranular cracking due to creep, transgranular cracking due to fatigue, and surface embrittlement due to oxidation is often observed in gas turbine components removed from service. The current maintenance scheme for gas turbines is to remove components from service when any criteria (elongation, stress-rupture, crack length, etc.) exceed the designed maximum allowable. A method for solving the inverse geometrical problem is presented by reconstructing the unknown subsurface cavity geometry using boundary element methods, a genetic algorithm, and Nelder-Mead non-linear simplex optimization. The heat conduction problem is solved utilizing the boundary element method, which calculates the difference between the measured temperature at the exposed surface and the computed temperature under the current update of the unknown subsurface flaws and cavities. This dissertation reports on research related to ultrafiltration (UF) membranes in drinking water applications. A pilot-scale investigation identified seasonal surface water quality impacts on UF performance and resulted in the development of a dynamic chemically enhanced backwash protocol for fouling management. Subsequent analysis of UF process data revealed limitations with the use of specific flux, transmembrane pressure (TMP), and other normalization techniques for assessing UF process fouling. A new TMP balance approach is presented that identifies the pressure contribution of membrane fouling and structural changes, enables direct process performance comparisons at different operating fluxes, and distinguishes between physically and chemically …

The modern view of optimization is that optimization algorithms are not designed in a vacuum, but can make use of information regarding the broad class of objective functions from which a problem instance is drawn. The goal of this thesis is to design and fabricate a DC-to-DC converter for use in high-voltage capacitor charging applications. The primary objectives include increasing the efficiency and reducing the cost of traditional methods used for this application. Traditional methods were not designed specifically for high-voltage capacitor charging and were thus very primitive and exhibited lower efficiency. Recently, a highly resolved, finite element mesh was developed for the purpose of performing hydrodynamic calculations in the Western North Atlantic Tidal (WNAT) model domain. The WNAT model domain consists of the Gulf of Mexico, the Caribbean Sea, and the entire portion of the North Atlantic Ocean found west of the 60° W meridian. This high resolution mesh (333K) employs 332,582 computational nodes and 647,018 triangular elements to provide approximately 1.0 to 25 km node spacing.
