One of the biggest challenges the clinical research industry currently faces is the accurate forecasting of patient enrollment (namely if and when a clinical trial will achieve full enrollment), as the stochastic behavior of enrollment can significantly contribute to delays in the development of new drugs, increases in duration and costs of clinical trials, and the over- or under- estimation of clinical supply. After amputation, muscles in the residual limb are detached from their insertion points and no longer span the missing joints. Our objective was to quantify the effect of amputation-induced disuse on residual muscle structure, an indirect indicator of muscle force-generating capacity. One hind paw was surgically removed at the ankle joint of ten rabbits. At two weeks (n=5) and 4 weeks (n=5) post-amputation and for select muscles (gastrocnemius, soleus, tibialis cranialis, extensor digitorum, and flexor digitorum superficialis), we measured and computed several muscle structure properties. This research focuses on the development of a low dose radiotherapy (LD-RT) device for treatment of acute respiratory distress syndrome (ARDS), a respiratory illness that affects millions of people a year. The use of orthovoltage (200 – 500 keV) X-ray energy provides many advantages over traditional radiotherapy delivery with linear accelerators, such as low cost and greater accessibility. In addition, X-ray tubes have been shown throughout history to provide good treatment outcomes for pneumonia, and research has shown LD-RT to be just as effective with ARDS. Positron Emission Tomography (PET) data suffers from low image quality and quantitative accuracy due to different kinds of motion of patients during imaging. Hardware-based motion correction is currently the standard; however, is limited by several constraints, the most important of which is retroactive data correction. Data-driven techniques to perform motion correction in this regard are active areas of research. This thesis presents the use of diblock copolymers, poly(butadiene)-b-poly(ethylene oxide) (PBm PEOn) and poly(isoprene)-b-poly(ethylene oxide) (PImPEOn), as amphiphilic molecular building blocks for the formation of synthetic polymer bilayer membranes using the droplet interface bilayer (DIB) technique. The DIB technique makes use of the self-assembly of amphiphilic macromolecules along oil-water droplet interfaces that can then be physically connected for the construction of liquid supported macromolecular bilayers at the droplet interface. These bilayer membranes are capable of hosting both naturally occurring and synthetic protein channels. This technique has been used to form synthetic bilayer membranes …

According to the National Highway Traffic Safety Administration (NHTSA), motor vehicle crashes are the leading cause of death among people aged 1-54 in the United Sates. In 2015, the number of motor-vehicle deaths on U.S. roadways totaled 35,092, a 7% increase from 2014 (32,744). Though lower gas prices and increased vehicle mileage combined with risky driving behaviors (e.g. speeding, driving while texting) account for the increased fatality rate, seatbelt non-use has been a significant contributory factor. It is estimated that nearly half (48%) of passenger vehicle occupants involved in fatal crashes each year are unrestrained.

Since almost 70% of the U.S. population over 20 years old is overweight and 30% is obese, with much of the western world following suit, many patients that will potentially be administered circulating nanoparticles designed to localize to tumors and avoid non-target areas will have significant amounts of white adipose tissue (WAT), enlarged livers, and additional metabolic complications such as type 2 diabetes. Preventable symptoms continue to reduce the quality of life for chronic asthma sufferers and place an unnecessary burden on emergency services and the health care system at large. Over 1.7 million hospital-associated infections (HAIs), resulting in 99,000 deaths, occur each year in the United States. HAIs are defined as infections that occur within 48 hours of hospital admission without evidence of the infection being present or incubating at the time of admission. HAIs are a major concern to the medical community due to the potential loss of life and high costs. Cerebrospinal fluid (CSF) shunts are fully implantable medical devices that are used to treat patients suffering from conditions characterized by elevated intracranial pressure, such as hydrocephalus. In cases of shunt failure or malfunction, patients are often required to endure one or more revision surgeries to replace all or part of the shunt. This thesis describes the development of a portable, smart-phone connected system for continuous cardiac monitoring. Effects of varus knee alignment on the internal knee abduction moment (KAM) in walking has been widely studied. KAM has been shown to be closely associated with the development of medial knee osteoarthritis (OA). Photo-crosslinkable polymeric biomaterials have emerged in the field of biomedical research to promote tissue regeneration. For example, scaffolds that can be crosslinked and hardened in situ have been known to make suitable implant alternatives. Since injectable and photo-crosslinkable biomaterials offer the advantage of being minimally invasive, they have emerged to compete with autografts, a current highly invasive method to repair diseased tissue. A series of novel photo-crosslinkable, injectable, and biodegradable nano-hybrid polymers consisting of poly(ε-caprolactone fumarate) (PCLF) and polyhedral oligomeric silsesquioxane (POSS) has been synthesized in our laboratory via polycondensation. Balance is among the most challenging tasks for patients with movement disorders. Study and treatment of these disorders could greatly benefit from combined software tools that offer better insights into neuromuscular biomechanics, and predictive capabilities for optimal surgical and rehabilitation treatment planning. A platform was created to combine musculoskeletal modeling, closed-loop forward dynamic simulation, optimization techniques, and neuromuscular control system design. Spinal (stretch-reflex) and supraspinal (operational space task-based) controllers were developed to test simulation-based hypotheses related to balance recovery and movement control. Total knee replacement (TKR) is commonly used to correct end stage knee osteoarthritis (OA) of the knee joint. Unfortunately, difficulty with stair climbing has been seen to exist, prolonging the challenges of TKR patents. Complete understanding of loading at the knee is of great interest in order to aid patient populations, implant manufacturers, rehabilitation, and future research. The outcome of a TKR is intended to reestablish normal motion and loading of the knee. Musculoskeletal modeling provides a means to accurately approximate joint loading and the corresponding muscle contributions during a movement.

This dissertation concentrates on the introduction of Predictive Adaptive Radiation Therapy (PART) as a potential method to improve cancer treatment. PART is a novel technique that utilizes volumetric image-guided radiation therapy treatment (IGRT) data to actively predict the tumor response to therapy and estimate clinical outcomes during the course of treatment. To implement PART, a patient database containing IGRT image data for 40 lesions obtained from patients who were imaged and treated with helical tomotherapy was constructed. The specific-locus mutation frequency resulting from 300 R of acute X irradiation has been determined for the germ cells present in male mice at 0, 2, 4, 6, 8, 10, 14, 21, 28, and 35 days of age and also for female mice at 0 days of age, Sample size was much larger for the males irradiated on day 0 than for other age groups but in all groups it was large enough to insure that an extremely high rate would be noticed.
