In 2016, Dr. Ahmad Nooristani identified a need for a personal EKG that is cheaper and simpler than hospital grade systems, but more accurate than personal devices currently on the market. Two previous teams did significant background research, prototyping, focusing on ease of use, portability, and physical specifications. However, neither team was able to produce a device with electrodes that could successfully collect an EKG signal and transmit it to a phone or tablet, one of the main requirements from the Dr. Nooristani. The goal of this study is to create an optimized Blalock-Taussig shunt used to temporarily repair pulmonary vascular blockages allowing a child time to grow so a more permanent surgical repair of the heart and vasculature can be performed. Blalock-Taussig or BT shunts are a surgical procedure performed on infants suffering from cyanosis or “Blue Baby Syndrome.” A BT shunt is an artificial vessel placed between the right ventricle and the pulmonary artery to increase blood flow in the lung and blood oxygen saturation levels. Advances in imaging technology have led to a variety of available clinical and investigational systems. In this collection of studies, we tested the relevance of morphological image feature quantification on several imaging systems and epithelial tissues. Quantification carries the benefit of creating numerical baselines and thresholds of healthy and abnormal tissues, to potentially aid clinicians in determining a diagnosis, as well as providing researchers with standardized, unbiased results for future dissemination and comparison.

Tibiotalar arthrodesis is a surgical procedure, used for the treatment of end-stage ankle arthrosis and instability. There are dozens of described procedures in the literature, all with varying rates of success. Two of the most common reasons for reoperation in tibiotalar arthrodesis are nonunion and infection; few studies have established any associations between patient/surgical factors and reoperation for nonunion and infection.

Mitral Valve Disease (MVD) describes a variety of pathologies that result in regurgitation of blood during the systolic phase of the cardiac cycle. Decisions in valvular disease management rely heavily on non-invasive imaging. Transesophageal echocardiography (TEE) is widely recognized as the key evaluation technique where backflow of high velocity blood can be visualized under Doppler. This dissertation describes fabrication of devices and other tools for biomedical applications through the integration of acoustofluidic systems with bio separation assays, instrumentation components, and soft materials interfaces. For example, we engineer a new class of transparent acoustic flow chambers ideal for optical interrogation. We demonstrate efficacy of these devices by enhancing the signal for high throughput acoustic flow cytometry, capable of robust particle focusing across multiple parallel flowing streams. A lack of adherence to prescribed physical therapy regimens in improper healing results in poor outcomes for those affected by musculoskeletal disorders (MSDs) of the upper limb. Societal and psychological barriers to proper adherence can be addressed through the system presented in this work consisting of the following components: an ambulatory biosignal acquisition sleeve, an electromyography (EMG) based motion repetition detection algorithm, and the design of a compatible capacitive EMG acquisition module.

The goal of this study is to design and manufacture a microfluidic device capable of measuring changes in impedance valuesof microfluidic cell cultures. Tocharacterize this, an interdigitated array of electrodes was patterned over glass, where it was then bonded to a series of fluidic networks created in PDMS via soft lithography. The device measured ethanol impedance initially to show that values remain consistent over time. Impedance values of water and 1% wt. saltwater were compared to show that the device is able to detect changes in impedance, with up to a 60% reduction in electrical impedance in saltwater. Musculoskeletal injuries can severely inhibit performance of activities of daily living. In order to regain function, rehabilitation is often required. Assistive devices for use in rehabilitation are an avenue explored to increase arm mobility by guiding therapeutic exercises or assisting with motion. Electromyography (EMG), which are the muscle activity signals, may be able to provide an intuitive interface between the patient and the device if appropriate classification models allow smart systems to relate these signals to the desired device motion.

The size of its floating mass transducer (FMT) can preclude implantation in certain anatomies, necessitating comprehensive surgical planning. Determining normal and abnormal fetal function in utero in order to better predict which fetuses are at risk for adverse outcome is critical. However, the medical imaging tools that could assist with diagnosis are very expensive and rarely available in the developing world. In this study, we developed a prototype audio-motio-tachograph (AMTG), which measures fetal movements through the recording of abdominal wall deformations and tested it in Rwanda. First, we showed that AMTG detected fetal signals and that fetuses respond to complex acoustic stimuli. The objective of this dissertation was to study the effect of locally delivered bioactive modulators on macrophage activation at the implantation site of different biomaterials in rats. Iloprost, a prostacyclin analog, was tested for its ability to direct macrophages to their pro-wound healing phenotype after the implantation of microdialysis probe in the subcutaneous space of male Sprague Dawley rats. Background: Continuous-flow (CF) left ventricular assist devices (LVADs) have gained widespread clinical acceptance as a treatment option for advanced heart failure (HF); however, they have also been associated with an increased risk of adverse events, including gastrointestinal bleeding, aortic insufficiency, and hemorrhagic stroke. It has been hypothesized that the increase in adverse event incidence may be due in part to diminished vascular pulsatility and high shear stress when CF-LVADs are operated at fixed speeds. Prior work at Marquette University developed the Marquette Prosthesis, an active transtibial prosthesis that utilized a torsional spring and a four-bar mechanism. The controls for the Marquette Prosthesis implemented a finite state control algorithm to determine the state of gait of the amputee along with two lower level controllers, a PI moment controller to control the moment during stance and a PID position controller to control the position during stance. The Marquette Prosthesis was successful in mimicking the gait profile presented by Winter. Despite the emergence of stemless humeral implants that utilize short fixation features to gain purchase solely in the metaphysis, the literature contains little information regardingthe morphology and mechanical properties of the humerus’ proximal trabecular-canal, and how stemless implants impact bone response. The present work employs in-silicotools, including CT-based and Finite Element (FE) methods, to define parameters that may influence stemless implant design.

The density and morphology of the proximal humerus were assessed using CT-derived point clouds of the trabecular-canal. Bone density was found to diminish 15-20mm beneath the humeral head resection and was greater peripherally. The depth, path and bounding …

This report documents all the research, ideation, and mockups used to determine right ventricle motion and develop a system capable of reproducing that motion on a tissue sample. The model is intended for evaluating anchoring systems being developed by Edwards Lifesciences for use with tricuspid valve therapies. Several design solutions were considered for the primary functions of recreating motion of the right ventricle and attaching tissue to the device. In this thesis, a prototype ECG monitor was developed that is integrated into an elastic shirt and takes a 3-lead ECG for over 5 days. The high-quality measurements can be used to identify markers indicative of various detrimental heart conditions. Measurements recorded by the device are encrypted and stored onto a micro-SD card. Current Holter monitors are expensive and have functional lives less than 48 hours; however, extended duration monitoring has been proven more useful in diagnosis. The device designed demonstrates that ECG measurements can be taken over longer durations without sacrificing quality, comfort, or device cost.

Some unilateral transfemoral amputees (TFAs) run in a prosthesis with an articulating prosthetic knee, others choose to run without a knee (prosthetic socket and foot are linked via a straight, non-articulating pylon) to increase stability. Research regarding unilateral TFA running with an unlocked versus locked prosthetic knee (approximately equivalent to the no-knee condition) with respect to energy cost and temporal metrics is limited; no studies have investigated the impact of knee condition on kinematic metrics.
