Reverse engineering the whole-genome networks of complex multicellular organisms continues to remain a challenge. While simpler models easily scale to large number of genes and gene expression datasets, more accurate models are compute intensive limiting their scale of applicability. To enable fast and accurate reconstruction of large networks, we developed Tool for Inferring Network of Genes (TINGe), a parallel mutual information (MI)-based program. Building energy simulation is of considerable interest and benefit for architects, engineers, and urban planners. Only recently has it become possible to develop integrated energy models for clusters of buildings in urban areas. Simulating energy consumption of the built environment on a relatively large scale (e.g., such as a neighborhood) will be necessary to obtain more reliable results, since building energy parameters are influenced by characteristics of the nearby environment. This report contains the information and background on top-down cost estimating using artificial neural networks (ANN)_to enhance the accuracy of MDT early estimates of construction costs. Upon conducting an extensive review of MDT’s budgeting and cost estimating efforts, and following a survey of agency experts on the identification of the most salient project attributes with the dual-objectives of low effort and high accuracy, a rational method for top-down variable selection is proposed. Selected variables were further tested in their explanatory power of construction costs through the application of two cost estimating methodologies—multiple regression and artificial neural network methodologies. Many large infrastructure projects are subject to considerable uncertainty over their revenue streams which makes their valuations particularly challenging. When project scalability can be altered as new information is revealed over the course of a project’s lifecycle, the presence of uncertainty can, in fact, provide option values that are ignored with static Net Present Value (NPV) analyses. To achieve these benefits, however, a better understanding of volatility is of the essence. After being the standard plant propagation protocol for decades, cultures of Arabidopsis thaliana sealed with Parafilm remain common today out of practicality, habit, or necessity (as in co-cultures with microorganisms). Regardless of concerns over the aeration of these cultures, no investigation has explored the CO2 transport inside these cultures and its effect on the plants. Segmentation of vegetation is a critical step in using machine vision for field automation tasks. A new method called reduced-dimension clustering (RDC) was developed based on theoretical considerations about the color distribution of field images. RDC performed unsupervised classification of pixels in field images into vegetation and background classes. Bayes classifiers were then trained and used for vegetation segmentation. The performance of the classifiers trained using the RDC method was compared with that of other segmentation methods. The RDC method produced segmentation performance that was consistently high, with average segmentation success rates of 89.6% and 91.9% across both cloudy and …

Genes encoding early signaling events in pathogen defense often are identified only by their phenotype. Such genes involved in barley-powdery mildew interactions include Mla, specifying race-specific resistance; Rar1 (Required for Mla12-specified resistance1), and Rom1 (Restoration of Mla-specified resistance1). The HSP90-SGT1-RAR1 complex appears to function as chaperone in MLA-specified resistance, however, much remains to be discovered regarding the precise signaling underlying plant immunity. Genetic analyses of fast-neutron mutants derived from CI 16151 (Mla6) uncovered a novel locus, designated Rar3 (Required for Mla6-specified resistance3). Host cell proteins (HCP) are a problematic set of impurities in downstream processing (DSP) as they behave most similarly to the target protein during separation. Approaching DSP with the knowledge of HCP separation behavior would be beneficial for the production of high purity recombinant biologics. Therefore, this work was aimed at characterizing the separation behavior of complex mixtures of HCP during a commonly used method: anion-exchange chromatography (AEX). The accuracy of machine learning tasks critically depends on high quality ground truth data. Therefore, in many cases, producing good ground truth data typically involves trained professionals; however, this can be costly in time, effort, and money. Here we explore the use of crowdsourcing to generate a large number of training data of good quality. We explore an image analysis task involving the segmentation of corn tassels from images taken in a field setting. Project ExCEL (Extended Classroom for Enhanced Learning) brings the capabilities of scanning electron microscopy (SEM) into classrooms. University and industry personnel, working together, have developed a web-based interface to allow schools to control a modern SEM. The interface allows a user control of the operating parameters of the microscope, stage movement, and chemical analysis. Such total control is not available on any other system. Since Iowa State University (ISU) pioneered the idea of remote SEM for education, researchers have learned that providing teachers access to sophisticated equipment does not ensure that it will be used. The goal of Project ExCEL, the Extended Classroom for Enhanced Learning, is to bring the capabilities of Scanning Electron Microscopy (SEM) into elementary and secondary classrooms. We have developed an entirely web-based interface to allow schools to control a modern SEM. This paper evaluates key functional data parameters that must be considered for suborbital spaceflight participants wearing pressurized suits for intravehicular activity (IVA). Data parameters of an analog spacesuit worn in an analog flight environment were obtained from 40 civilian participants using the Suborbital Space Flight Simulator (SSFS) at Embry-Riddle Aeronautical University (ERAU) while donning Final Frontier Design’s (FFD) fully pressurized third-generation spacesuit as part of their training for Project PoSSUM (the Polar Suborbital Science in the Upper Mesosphere Project). The SUIT Lab at Embry-Riddle Aeronautical University is a joint student-faculty project utilizing multiple high altitude pressure garments to investigate suited crew capabilities within a spacecraft during simulated spaceflight missions. The testing environment within the SUIT lab includes the use of suits in a lowfidelity capsule cabin mockup with a horizontally situated launch-positioned chair simulator. Standard videography and analytical video software are used to determine levels of achievement in ergonomic range of motion and comfort design across multiple spacesuits. Comparative analysis and testing provide data supporting the requirement for the use of particular spacesuits inside proposed commercial launch vehicles. Results …

Conceptual designs for a space suit Personal Life Support Subsystem (PLSS) were developed and assessed to determine if upgrading the system using new, emerging, or projected technologies to fulfill basic functions would result in mass, volume, or performance improvements. Technologies were identified to satisfy each of the functions of the PLSS in three environments (zero-g, Lunar, and Martian) and in three time frames (2006, 2010, and 2020). The viability of candidate technologies was evaluated using evaluation criteria such as safety, technology readiness, and reliability.
