In skeletal muscle (SM), the microcirculation distributes blood flow to capillary networks to meet local oxygen (O2) demand. Experiments using intra-vital video microscopy (IVVM) have quantified blood flow and O2 supply in capillary networks, and using an O2 exchange chamber have measured local changes in blood flow believed due to O2-dependent regulation. However, IVVM is unable to simultaneously measure capillary O2 supply and SM O2 content, implying the need for theoretical models to understand how flow regulation and SM oxygenation interact. During the summer of 2021, I was employed as an intern at Dynatek Labs, an implantable prosthetics testing company located in southern Missouri. My primary project was to modernize an abandoned pulse duplicator device and protocol to allow the company to re-enter a market area which they left nearly 15 years prior: heart valve testing. The company halted this initiative due to changes in international testing standards in previous years. The International Standards Organization Cardiac Valves Working Group released the latest version of standards concerning artificial heart valve testing (ISO 5840) in January of 2021. Cardiovascular diseases (CVDs) are the leading cause of death worldwide in males (XY) and females (XX). Prior to menopause, females have a relative protection against serious cardiac pathologies compared to age-matched males. This phenomenon is widely attributed to the ovarian hormone estrogen. Unfortunately, hormone replacement therapy to maintain estrogen levels in postmenopausal females has overall adverse effects, and it is not recommended for long-term use or as a preventative measure for eCVDs. The headgear design research community has focused on developing accurate computational and experimental analysis techniques to better assess the ability of headgear technology to attenuate impacts and protect athletes from TBI. This study developed an analytical tool for understanding spine tissues’ behavior in response to vertebral kinematics and spine pathology over a range of body postures. It proposed a novel pipeline of computational models based on predicting individual vertebral kinematics from measurable body-level motions, using musculoskeletal dynamics simulations to drive the vertebrae in corresponding spine FEMs.
A reformulated elastic surface node (ESN) lumbar model was developed for use in MSD simulations. The ESN model modifies the lumbar spine within an existing MSD model by removing non-physiological kinematic constraints and including elastic IVD behavior. Introduction
Lateral dislocation of the patella is a common injury in active adolescents and young adults. Patients who are ultimately managed surgically have a significantly lower risk of recurrent dislocation. However, determining the optimal surgical treatment remains a challenge, with patients sometimes undergoing multiple surgeries prior to successful stabilization. The aim of this study is to computationally evaluate patients that have undergone multiple surgeries to correct for recurrent lateral patellar dislocation and predict their clinical outcome.
Methods

Hydrogel adhesives are a new class of materials with excellent biocompatibility, which makes them very attractive for biomaterial applications. It has been previously shown that Tetronic T1107, a four-arm poly (propylene oxide)-poly (ethylene oxide) (PPO-PEO) block copolymer, is useful as a chemical crosslinking thermo-responsive hydrogel for bioadhesive applications. The end groups of this polymer are modified with acrylate and N-hydroxysuccinimide (NHS) functional groups. The acrylate end group gives the polymer cohesive properties with long-range chemical crosslinking using dithiothreitol (DTT), while the NHS end group gives the polymer adhesive properties through bonding with amines found in organic tissue. The motivation for this Ph.D. dissertation is to provide football equipment managers, coaches, parents, athletes, and relevant industry personnel with an understanding of the implication a chosen football facemask design will have on the safety of the athlete. As athletes have increased their capacity for speed, size, and strength, so too has the head injury risk increased in American football. In the Unites States, approximately 1.5 million people currently have a spinal cord injury and suffer permanent sensory and motor loss due to the disruption of the spinal cord. Due to the significant morbidity, it is vital to understand the functional impact of disrupting neural descending pathways that modulate spinal neurons involved in intermuscular coordination critical for gait behaviors. Finite element models can predict subject-specific chondrolabral stresses and help to elucidate the effect of under-coverage and incongruency of the hip joint in patients with dysplasia. However, complex stress patterns are difficult to generalize and evaluate statistically. With an established correspondence across shapes from statistical shape modeling (SSM), statistical parametric mapping (SPM) allows for evaluation of local variability while preserving model subject-specificity. Herein, we evaluated the combined application of SSM and SPM to compare cartilage contact stress between control subjects and patients with dysplasia.

Developmental dysplasia of the hip (DDH) is described as under-coverage of the femoral head by the acetabulum, resulting in mechanical instability. Though DDH is often diagnosed using plain film radiographs, these images cannot adequately capture 3D joint coverage. Herein, we applied a 3D statistical shape model (SSM) to the femur and hemi-pelvis of patients with DDH to objectively measure shape variation and evaluated whether SSM outputs could predict measurements of joint coverage.

In order to study the human brain in a non-invasive way, the vestibular and oculomotor systems have been analyzed simultaneously. To achieve this, an eye movement and balance assessment was conducted for 29 (23 male; 21.6 ± 3.8 years) healthy participants with no history of concussions, brain injuries, vertigo, migraines, brain surgeries, eye surgeries, or amblyopia. The tasks consisted of eyes open, eyes closed, horizontal saccades, vertical saccades, vergence jumps, horizontal vestibulo-ocular reflex (VOR), and vertical VOR. Sustained local administration of active agents has been proposed to cure bacterial vaginosis in the female reproductive tract and restore the resident bacterial fauna. Bioprinting has shown promise for the development of systems for local agent delivery. In contrast to oral ingestion, agent release kinetics can be fine-tuned by bioprinting specialized scaffold designs tailored for particular treatments while enhancing dosage effectiveness via localized sustained release. Venous valve failure allows for the retrograde, or backward, flow of blood into the lower extremities, which leads to Chronic Venous Insufficiency (CVI). CVI infringes upon quality of life through ulceration and can result in death due to Deep Vein Thrombosis (DVT), or blood clots, causing pulmonary embolism. A successful treatment of CVI restores valve function and prevents retrograde blood flow; however, current bioprosthetic venous valves exhibit low patency and high calcification. Cardiovascular diseases are the leading cause of death in the United States. Atherosclerosis in peripheral arteries is a major contributing factor. Autologous saphenous vein grafts are the most common bypass grafts for treating peripheral artery diseases. A quarter of these grafts fail within a year, and around half have failed within 10 years of the initial surgery. Graft failure is attributed to the development of intimal hyperplasia indicated by the migration and proliferation of vascular smooth muscle cells, fibroblasts, and the deposition of extracellular proteins.

Falls are a prevalent problem within elderly populations. Falls also increase the cost of healthcare, since they cause severe injuries frequently and negatively affect quality of life. Lack of postural stability is the consequence of falls, and postural stability is defined as the correct biomechanical execution based on sensory feedback. The types of sensory feedback are vision, vestibular (ear), proprioceptive (body awareness), and somatosensory (sensation of the environment). Utilizing resistance methods for sprinters is a common approach to their training. In this study, six athletes from the University of Nebraska-Lincoln Women’s Track and Field Team ran a series of sprints using a resistance machine to collect data regarding the change in power output, stride length, level of trunk tilt with respect to the ground, and acceleration throughout a distance of ten meters when different amounts of resistant forces were applied to the athlete. The meshless method is an incredibly powerful technique for solving a variety of problems with unparalleled accuracy and efficiency. The pharmacokinetic problem of transdermal drug delivery (TDDD) is one such topic and is of significant complexity. The locally collocated meshless method (LCMM) is developed in solution to this topic. First, the meshless method is formulated to model this transport phenomenon and is then validated against an analytical solution of a pharmacokinetic problem set, to demonstrate this accuracy and efficiency. Hemiarthroplasty, where one side of a joint is replaced, is a minimally invasive procedure. It allows for the preservation of native tissue, though a significant ramification is accelerated cartilage wear when articulating with high stiffness materials that do not mimic the mechanical stiffness of the native tissue. An implant that employs a lattice design can significantly lower the stiffness of a solid structure whilst maintaining strength. Research development involving large scale joint mechanics and biomechanical adaptations is growing. However, interest in smaller scale joints, such as the fingers, is limited. Thus, the present work describes the enhancement and clinical application of a previously designed in-vitro active finger motion simulator in measuring and assessing intrinsic joint kinematics and tissue biomechanics including load transfer and strains induced tissues within the finger. Accuracy of electromagnetic tracking (EM) systems were evaluated compared to the standard optical tracking systems and used to develop motion derived finger joint coordinate systems. Breast cancer most frequently metastasizes to the skeleton. Bone metastatic cancer is incurable and induces wide-spread bone osteolysis, resulting in significant patient morbidity and mortality. Mechanical stimuli in the skeleton are an important microenvironmental parameter that modulates tumor formation, osteolysis, and tumor cell-bone cell signaling, but which mechanical signals are the most beneficial and the corresponding molecular mechanisms are unknown. In most everyday activities, we head towards a specific goal by updating our choices for a more direct path. However, there are specific clinical tasks where taking the direct path is more challenging. Clinical investigations of optimizing a prosthesis involve the assessment of multiple parameter settings through trial and error rather than goal-directed optimization. We investigate if a human-in-the-loop optimization algorithm can guide manual alterations to a prosthesis-simulating device to reduce the ground reaction force on the contralateral limb. Background: Patients with peripheral artery disease (PAD) have significantly reduced lower extremity muscle strength compared with healthy individuals as measured during isolated, single plane joint motion by isometric and isokinetic strength dynamometers. The objective of this study was to understand the contribution of the ankle muscles during walking in patients with PAD and compared to healthy older individuals.
Methods: A total of 12 patients diagnosed with Fontaine stage II PAD and 10 healthy older controls were recruited for the study. Each subject walked across a 10-meter pathway with reflective markers placed on specific anatomical locations on lower limbs while the …
