Background: A family history of Type 2 Diabetes (FH) is considered a risk factor for development of type 2 diabetes (T2D). Exercise training is an effective strategy to improve insulin resistance. Recently, micro-RNAs (miRNAs) have emerged as mediators of various intracellular processes, including insulin signaling, and may act as signaling molecules when released into circulation. Molecular techniques for guiding plant breeding have successfully used wild progenitors of domestic crops as sources of genetic variants conveying desirable traits. However, epigenetic variation, in particular DNA methylation, is a significant source of phenotypic variation and epigenetic effects of plant domestication are poorly understood. Described herein are the first single-base pair resolution methylomes of the highly valued crop iceberg lettuce (Lactuca sativa cv. Salinas) and its close relative, and ubiquitous weed, L. serriola. Pancreatic ductal adenocarcinoma (PDAC) is a notoriously lethal disease characterized by prominent stromal involvement, which plays complex roles in regulating tumor growth and therapeutic response. The extracellular matrix (ECM)-rich stroma has been implicated as a barrier to drug penetration, although stromal depletion strategies have had mixed clinical success. It remains less clear how biophysical interactions with the ECM regulate invasive progression and susceptibilities to specific therapies. Very few vertebrates survive extended periods of time without oxygen. Entry into metabolic depression is central to surviving anoxia, which is supported by overall suppression of protein synthesis, yet requires increased expression of specific proteins. Interleukin-1β (IL-1β), classically considered as a pro-inflammatory cytokine, has proven essential to cellular defense in nearly all tissues (Kaur et al., 2014, Ren and Torres, 2009). In brain, studies suggest that IL-1β has pleiotrophic effects. It acts as a neuromodulator, has been implicated in the pathogenic processes associated with a number of CNS diseases, but has also been shown to provide protection to the injured CNS. With respect to the latter, IL-1β signalling appears to mitigate pathology and motor symptoms in a mouse model of Huntington’s disease (HD), a progressive neurodegenerative condition that targets the striatum. The model of basal ganglia function proposed by Albin, Young and Penney (1989) describes two anatomically independent motor pathways, the direct and indirect. However, under normal conditions striatal dopamine (DA) is required for the expression of motor behavior, and DAergic control of the two pathways (via D1 and D2 receptors, respectively) is dependent on co-activation. We tested for a possible breakdown of D1/D2 synergism using transgenic R6/1 mice bearing the human huntingtin allele (Htt). Performance is key to survival. From day-to-day foraging events, to reproductive activities, to life-or-death crises, how well an organism performs these tasks can determine success or failure. Selection, therefore, both natural and sexual, act upon performance, and performance demands on individuals shape a population’s morphological and physiological trait distributions. While studies of morphological adaptations to ecological pressures implicitly center on the idea that responses to selection improve performance via changes in morphology, the relationships between morphology, performance, and fitness are not always well understood. Wing Shape Variation In The Mimetic Butterfly Papilio Dardanus (Papilionidae) And Its Unpalatable Nymphalid Models, Miles Hegedus

Papilio dardanus displays female-limited polymorphic mimicry of multiple model species. Butterfly wing shape is species-specific and can influence mimetic signaling, but has not been characterized in this species. We used elliptical fourier analysis to investigate whether mimetic P. dardanus female forms have converged on the wing shape of their respective models. Although both models and mimics varied in forewing and hind wing shape, we found no evidence of forewing shape convergence between them. Overall, forewings did not differ in shape between sexes in P. dardanus, nor in four non-mimetic Papilio used for comparison. Similarly, there were no hind wing …

In mammals, calcium influx is required for oocyte maturation and egg activation, as it supports the persistent calcium oscillations induced by fertilization. These oscillations are required for the initiation of embryo development. The molecular identities of the plasma membrane calcium-permeant channels that underlie calcium influx are not established. Among these channels, Transient Receptor Potential Vanilloid, member 3 (TRPV3) allows divalent cations, namely strontium (Sr2+) and calcium (Ca2+) with high permeability, into cells, and its expression pattern seems to predict an essential role in the initiation of development. In eukaryotes, DNA repair mechanisms detect and repair damaged DNA. DNA damage is primarily caused by a variety of exogenous and endogenous sources. Several types of damage to DNA are repaired by different kinds of DNA repair pathways. This dissertation focused on repair of N-methylpurines (NMPs) and double-strand breaks (DSBs) in human fibroblasts.

NMPs, including N7-methylguanine (7MeG) and N3-methyladenine (3MeA), can be induced by environmental methylating agents (e.g. the soil fumigant methyl bromide), chemotherapeutics (e.g. nitrogen mustards), and natural cellular methyl donors like S-adenosylmethionine. In the past couple of decades, there has been an increase in the occurrence of wildfire events in the United States. The U.S. Forest Service estimates that over 73,000 wildfires on average occur annually in the U.S., burning about 7.3 million acres of land. Globally there is a biodiversity crisis, with many groups of species threatened with extinction due to changes in the environment and human impacts. Amphibians are one such group and according to the IUCN, over 30% of amphibians are threatened by extinction. There are many factors have that can explain the decline of amphibians including pollution, habitat loss, climate change and disease. One factor is chytridiomycosis, an emerging infectious disease caused by the chytrid Batrachochytrium dendrobatidis. The chytrid enters the keratinized skin of the amphibian and asexually reproduces, where it disrupts host functions, often leading to host death.

This capstone is a data-driven investigation into New York City's rat problem. By using publicly available government data to map rat activity in NYC, I identified several socio-economic variables that correlate with rat populations at the community district, borough, and city-scale. I used these findings (mainly that rat problems are linked to lower incomes) as the basis of an investigation, which includes interviews with residents, experts, and city officials. Prof. Bobby Corrigan, urban rodentologist and formerly with the NYC Department of Health criticizes the city's efforts for the first time on the record.

The use of prescribed fire is integral to the restoration of open woodland habitats in the southeast, including shortleaf pine (Pinus echinata) woodlands in the Ouachita Mountains. Mature pine habitats maintained with recurrent disturbances have an open understory with a rich floristic diversity that provides quality habitat for many wildlife species, including the endemic and endangered red-cockaded woodpecker (Picoides borealis). Plants store starch during the day for use at night. This process of transitory starch degradation is mostly attributable to the β-Amylase (BAM) family, which are starch exohydrolases that cleave the penultimate α-1,4 glycosidic bonds of starch to release maltose. BAM2 was recently characterized as a catalytically active, K+-requiring tetramer with sigmoidal kinetics and cooperativity. All other catalytically active BAMs display Michaelis-Menten kinetics, no cooperativity, and do not require salt, making BAM2’s characteristics intriguing. Due to a lack of a crystal structure, a monomeric homology model of BAM2 was generated using I-TASSER based on a BAM5 from soybean. …

Sudden Infant Death Syndrome (SIDS) is a leading cause of infant mortality. Alterations in brainstem development of Serotonin (5HT) and γ-aminobutyric acid (GABA) are linked to its cause. The sympathetic premotor neurons located in the Nucleus of the Raphe Pallidus (NRP) in the brainstem have both 5-HT1A and GABA-A receptor subtypes that mediate protective cardiovascular responses to environmental stress. It is hypothesized that alteration in these receptors at the NRP will also modify protective thermoregulatory responses to hypoxic stress such as hypothermia. During autumn and winter, mallards (Anas platyrhynchos) maximize fitness through their spatiotemporal distribution to avoid mortality risks while balancing trade-offs to access foods to undergo migration and maintain homeostasis. The mouse brain can be studied over vast spatial scales ranging from microscopic imaging of single neurons to macroscopic measurements of hemodynamics acquired over the majority of the mouse cortex. However, most neuroimaging modalities are limited by a fundamental trade-off between the spatial resolution and the field-of-view (FOV) over which the brain can be imaged, making it difficult to fully understand the functional and structural architecture of the healthy mouse brain and its disruption in disease. Huntington’s disease (HD) is a debilitating neurodegenerative disorder with a complex pathophysiology. Despite extensive studies to study the disease, the sequence of events through which mutant Huntingtin (mHtt) protein executes its action still remains elusive. Movement and habitat selection by Greater Sage-grouse (Centrocercus uropasianus) is of great interest to wildlife managers tasked with applying conservation measures for this iconic western species. Current technology has created small and lightweight GPS (Global Positioning Systems) transmitters that can be attached to sage-grouse. Using GIS software and statistical programs such as Program R, land managers can analyze GPS location data to assess how sage-grouse are geospatially interacting with their habitats. Although each species is widely distributed, information about their diet and activity patterns is lacking, especially within Kansas. Regulation of gene expression is essential to life. Post-transcriptional regulation of gene expression is a complex process with many inputs that lead to changes in localization, translation and stability of mRNAs. The translation and stability of many mRNAs is regulated by cis-elements, such as mRNA-structure or codon optimality; and by trans-acting factors such as RBPs and miRNAs. Here I report on the complex interactions between RBPs, miRNAs and characteristics of their target mRNAs in respect to effects on translation and RNA stability.

IAV successfully infects a diverse host range and adaptation of new viral strains to humans may cause pandemic events with high morbidity and mortality.
