Spinal cord injury (SCI) is a significant public health concern that leaves patients with a multitude of life-long disabilities. Major complications of SCI apart from paralysis, include deficits in bladder and bowel function. Skeletal muscle constitutes a highly plastic and malleable tissue that rapidly and profoundly adapts to various environmental and physiological stimuli. This response takes place by means of regulated signaling transduction which elicits changes in gene expression, biochemical, and metabolic properties. Adult muscle is composed by alignment of multinucleated, post mitotic muscle fibers formed during embryonic and neonatal development. Residing in the niche of mature muscle fibers is a small population of undifferentiated muscle progenitors termed satellite cells. The Louisville and Jefferson County Metropolitan Sewer District (MSD) has developed a Long-Term Control Plan (LTCP) to address the combined sewer overflow (CSO) issue through a combination of gray and green infrastructure (GI) stormwater management practices in the city of Louisville. In support of this effort, the MSD installed a series of GI stormwater control measures (SCMs) to abate the CSO volumes in an urban sewershed basin. Background: In the U.S., prostate cancer (PCA) metastasis is associated with a 5-yr survival rate of 29% and disproportionately affects men of African descent. Single nucleotide polymorphisms (SNPs) may serve as genetic markers to identify individuals at high risk of developing PCA. Methods: We examined main and joint effects of 119 inflammatory and immune response-associated SNPs among men of African descent (535 controls, 279 cases) via logistic regression (LR) and entropy-based-multi-factor dimensionality reduction (MDR) modeling. Quality program evaluation investigates both the process and the outcomes of an intervention in order to accurately determine its effectiveness and impact. In the present study, the effectiveness of CHOICES, a brief, applied-theatre intervention that addresses cyberbullying and suicide was evaluated. The fidelity of implementation of the intervention was assessed to determine whether the delivery of CHOICES was consistent with the original goals of the program, as well as consistent across multiple deliveries. Investigation of the fidelity of implementation revealed that CHOICES was not implemented with high levels of fidelity. The study also determined whether exposure to CHOICES led to …

The type IIa sodium-phosphate cotransporter (Npt2a) is expressed in the apical membrane of the renal proximal tubule and is responsible for the reabsorption of the majority of the filtered load of phosphate. Parathyroid hormone (PTH) is secreted by the parathyroid gland in response to a decrease in serum calcium or an increase in serum phosphorus, and acutely induces phosphaturia through the rapid stimulation of endocytosis of Npt2a and its subsequent lysosomal degradation. Chronic PTH stimulation leads to sustained phosphaturia, but the mechanisms for the chronic regulation of Npt2a by PTH remain unclear. We hypothesize that PTH decreases Npt2a mRNA levels …

What is not clear is why some students, despite the pervasiveness of stressors are able to do well in school and maintain/improve their health, while other students struggle to maintain their health and do poorly in school. Our current knowledge on cement hydration during setting is based on the discrete observation of hydrated paste. An advanced micro/nano-level technique which can perform the in-situ observation on the continuous hydration of cement paste is demanded. In this study, Raman spectroscopy (RS) was chosen as such a method to continuously investigate wet pastes. The objective of this research is to explore the hydration process and microstructural development of fresh pastes with this technique. This research was conducted in three phases. Host cells respond to bacterial lipid A through Toll-like receptor 4 (TLR4). Activation of TLR4 by lipid A triggers a response that involves two main adaptor proteins, MyD88 and TRIF. MyD88-dependent gene expression is associated with proinflammatory protein production, while TRIF-dependent gene expression is essential for optimal activation of adaptive immunity by antigen-presenting cells. Detoxified, monophosphoryl lipid A agonists (MPLA or synthetic MLA) were previously suggested to elicit TRIF-biased TLR4 signaling; that is, induction of weaker MyD88-associated gene expression but relatively intact TRIF-dependent gene expression when compared to fully active diphosphoryl lipid A (lipid A). My dissertation provides theoretical insights on individual level entrepreneurial identity in three inter-related essays, drawing on multiple theories and disciplines. In the first essay, I advance a dual-process model of identity development, contributing to the debates in the broad identity literature. I articulate that the intrinsic prospection of identity is another fundamental process of identity formation, besides the social construction of identity, the prevalent notion in the literature. I propose a comprehensive conceptual framework of entrepreneurial identity. Noise reduction testing is completed for two single steel plate constructions with different thicknesses and for two double wall constructions with different air gap thicknesses that include the same two plates used in the single plate constructions. Results compare favorably with known theory and experimental results from previous literature. These favorable comparisons include the direct relationship between mass per unit area and transmission loss for single plate constructions, the presence of a noise reduction valley near the mass-air-mass frequency, the dependency of the mass-air-mass frequency on the air gap thickness, and the existence of a bridge frequency. The reaction of an aminooxy moiety (RONH2) with an aldehyde or ketone carbonyl, an oximation reaction, results in the formation of robust oxime ether linkages. Oximation is a form of click chemistry and is chemoselective, occurs in a variety of solvents including water, and produces high yields with little to no purification. We were inspired to exploit the advantages of oximation reactions by attaching aminooxy-functionalized molecules to solid supports, thus allowing us to employ aminooxy chemistry in ways that cannot be achieved using solution phase. A relationship between periodontal disease and oral squamous cell carcinoma (OSCC) has been demonstrated in recent studies. Porphyromonas gingivalis’ antigen have been found in oral cancer cells. The embryonic program “epithelial-mesenchymal transition” (EMT) is thought to trigger cancer invasion by allowing tumor cell propagation. The objective of this study was to examine the role of P. gingivalis in the expression of ZEB1, vimentin, and repression of miRNA-200 family as EMT prognostic markers in cancer cell metastasis. Detrusor-sphincter dyssynergia after chronic spinal cord injury (SCI) leads to over-distention, high bladder pressures and vesico-ureteric reflux into the kidney, resulting in repeated infections and pyelonephritis. Given the importance/limited knowledge of SCI effects on the upper urinary tract, this study assessed expression of two proteins in the kidneys of contused (T9) male rats relative to controls that are indicative of tissue stress/damage (TGFβ, CD11b). Lunasin is a 44 amino acid peptide derived from the soybean seed that has been shown to have cancer chemopreventive and chemotherapeutic properties. In this study, we investigated the potential utility of lunasin as a chemotherapeutic in a melanoma model. Initial studies showed that lunasin has little activity against established melanoma cell lines in vitro using adherent culture methods; however, lunasin’s in vitro activity was significantly higher in non-adherent colony-forming assays in soft agar and oncosphere assays. In tumor cells, glutamine metabolism provides anaplerotic carbon for the TCA cycle, fatty acid synthesis, and precursors for the production of nucleotides and glutathione. This metabolic alteration is primarily driven by disruptions in oncogenic or tumor suppressor function and results in glutamine dependency for tumor cell survival. Troglitazone, a PPARγ agonist, has been reported to alter glutamine metabolism. This thesis project addresses whether troglitazone treatment could disrupt glutamine metabolism in glutamine-dependent tumor cells. Results obtained with troglitazone treatment include: dose-dependent inhibition of cell proliferation, glutamine uptake, glutaminolytic protein expression, steady-state ATP levels, and glutamine carbon flux into the TCA cycle. …

Hantaviruses, family Bunyaviridae, are present throughout the globe in a variety of mouse, rat, mole, vole, shrew, or bat species. Hantaviruses persist for the lifetime of the animal reservoir, while causing no signs or symptoms of disease. Only the rodent-borne hantaviruses cause disease in humans. In contrast, a “spillover” infection of a hantavirus into a nonreservoir rodent species results in an asymptomatic acute infection. We and others in the field are interested in understanding the biology of these virus-host interactions and mechanisms that underlie these three very different outcomes. It is not complete in its detail or ambition but instead focuses on themes and topics of self-transformation, reinvention and the unintentional consequences of several nervous breakdowns and experiments with psychedelic substances. This thesis is just a beginning for a longer more drawn out memoir that will explore the themes above but also discuss family, one’s sense of self worth in the world and how patterns between fathers and sons repeat themselves. In the field of magnetocaloric heat pumps, many system models have been made. What most of these models lack is heat transfer occurring while there is no fluid motion. It is the objective of this paper to analytically determine and experimentally verify heat transfer with no bulk fluid motion. The addition of a pure dwell period to the typical magnetocaloric cycle was shown to have a positive performance impact in specific conditions. The objective of this study was to evaluate the presence of mental and physical disability in children's animation, and to understand image of mental or physical disability the animation portrayed. Three animated movies and one 61 episode animated series were coded for verbal references to mental illness and physical disability. Visual images and thematic elements portraying disability were also examined. All of the animations contained verbal as well as visual references to disability. Verbal references appeared with an average of 13.2 references per hour of animation. The connotations of these references often went against negative stereotypes of disability. These findings …

This dissertation study explored the relationship between school-based parent involvement and early reading outcomes by positing that different types of parent involvement activities reflect access to different forms of cultural capital and therefore should be analyzed as separate constructs. Exploratory factor analysis (EFA) and confirmatory factor analysis (CFA) techniques were used to establish the factor structure underlying measures of school-based parent involvement available in the Early Childhood Longitudinal Study-Kindergarten Cohort of 2011 (ECLS-K: 2011). Altered energy metabolism is an established hallmark of cancer cells. Fructose-2,6-bisphosphate is an allosteric activator of glycolysis and its concentration in a cell is dictated by the 6-phosphofructo-2-kinase/fructose-2,6-bisphosphatases (PFKFB1-4) family of bifunctional enzymes. The PFKFB family of enzymes are characterized by their different kinase: bisphosphatase activities and were originally determined to be tissue-specific. More recent data however suggest that multiple isoforms are co-expressed in both normal and neoplastic tissues. PFKFB4 is highly expressed in human cancer, strongly induced by hypoxia, and required for the survival of cancer cells.
