N-Succinyl-L,L-2,6-Diaminopimelic Acid Desuccinylase (DapE) is a bacterial enzyme located in the lysine biosynthetic pathway of all Gram-negative and most Gram-positive species of bacteria including the notorious ESKAPE pathogens, or pathogenic strains of bacteria that have developed significant levels of resistance to currently available antibiotics. As a means to combat this challenge of growing antimicrobial resistance, we have identified the bacterial enzyme DapE as a conserved, novel enzymate target within these resistant bacterial strains,as DapE is responsible for the production of m-DAP and lysine which are ultimately employed in bacterial cell wall synthesis. Few studies examine Black school psychologists' experiences of racial microaggressions in the work environment. However, based on the available literature on the impact of racial microaggression on Black school psychology graduate students, it can be argued that Black school psychology practitioners' experiences with racial microaggressions are similar to graduate students in school psychology training programs. Vascular smooth muscle cells (SMCs) surround many blood vessels throughout the body,where they dynamically alter vessel diameter to regulate blood pressure, provide structural integrity, and absorb shock on a beat-to-beat timescale. As smooth muscle function fails, profound vascular disease ensues, often with tragic results- even death. Smooth muscle myosin 2 (SM2) is the dominant motor protein that actuates contractility and allows SMCs to perform these vital functions. The goal of the research was to expand the data available in the social service sector associated with understanding the role of board member diversity within nonprofit organizations. To this end, this study applies Bolman and Deal’s 4-frame model of organizational learning lenses to school improvement indicators on the Illinois 5Essentials Survey. Through taking Bolman and Deal’s Online 4-frame Self-assessment, I am able to identify the organizational learning lens least integrated into my own leadership worldview. An under-investigated target of lysine acetylation is the bacterial ribosome. Although lysine acetylations on the ribosome are common and conversed in diverse bacterial species, little work has been done to understand how lysine acetylation affects the bacterial ribosome. The goal of this work is to determine if lysine acetylation has functional impact on the bacterial ribosome.I have identified in vitro and in vivo effects of nonenzymatic, AcP-dependent lysine acetylation on translation and the ribosome. In vitro acetylation of transcription-translation reactions causes a translation-specific defect that is unaffected by the addition of the CobB deacetylase. Philosophy of music, especially in the 20th and 21st centuries, presides over a relatively narrow range of field-specific ontological and metaphysical questions. I claim that a focus on classical music and a reliance on analogies to the plastic arts constitutes an unhelpful (but pervasive) methodology in philosophy of music, one that stands in tension with its purported aim of accurately accounting for “the ways we talk, think, and act” in relation to music and musical works (Rohrbaugh 2003, 179). A three-fold approach was taken to develop a method for the detection of heavy metals in different matrices and had to be performed sequentially. The first was to develop a method capable of quantifying lead in complex matrices by graphite furnace atomic absorption (GFAA). The developed method needed to meet specific analytical requirements, namely, to be robust, sensitive, and accurate. Initially, four different sample preparation methods were explored for the quantification of lead using Escherichia coli (E. coli) as a sample matrix. This project reimagines a first-century reception of the Gospel of Mark within a historically reconstructed (yet hypothetical) performance event. In particular, it considers the disciples' character and characterization through the lens of performance criticism. Questions concerning the characterization of the disciples have been relatively one-sided in New Testament scholarship, in favor of their negative characterization. This project demonstrates why such assumptions need not be necessary when we (re-)consider the oral/aural milieu in which the Gospel of Mark was first composed and received by its earliest audiences. This dissertation is a critical critique of Queer Theory as an academic field. It argues that queer theory’s establishment as an academic discipline and as a periodizing and historicizing force through the bibliographies that make up its canon has upheld and supported the very normative models of temporality and progression that the field claims to resist. While Pope Francis has recently called for “overcoming colonizing mentalities” through the lens of what he calls “integral ecology,” crucial aspects of the colonial paradigm remain neglected or underexamined in this approach: sexuality, gender, and the negotiation of religious difference. In this study, we sought to profile changes in intestinal epithelial cell miRNA expression after alcohol and burn injury and elucidate their impact on inflammation and barrier integrity. Hematopoiesis is dynamically controlled by an epigenetic landscape that regulates gene transcription. Ten-eleven translocation 2 (TET2) is a Fe2+ and α-ketoglutarate (α-KG)-dependent dioxygenase that regulates the transcriptional status of genes associated with hematopoietic stem cell self-renewal and multiple stages of lineage commitment and differentiation. TET2 mutations are commonly detected in almost all types of hematopoietic malignancies, including myeloproliferative disorder, acute myeloid leukemia, and peripheral T-cell lymphomas. In most cases, TET2 is a founder mutation that occurred in hematopoietic stem/progenitor cells (HSPCs). Gram-positive bacteria produce small autoinducing peptides (AIPs), which act to regulate expression of genes that promote adaptive traits including virulence. The Gram-positive pathogen Staphylococcus aureus generates a cyclic AIP that controls expression of virulence factors via the accessory gene regulatory (Agr) system. S. aureus strains belong to one of four Agr groups (I, II, III, and IV), and each group harbors allelic variants of AgrD, the precursor of AIP. In a prior screen for S. aureus virulence factors, the Alonzo lab identified MroQ, a putative peptidase. A ΔmroQ mutant closely resembled a Δagr mutant and had significant defects in AIP …

The TANK-binding kinase 1 (TBK1) is a cytoplasmic, serine/threonine kinase and a critical activator of the innate immune system. Reports have implicated TBK1 as a promoter of multiple cancer types. Acute myeloid leukemia (AML) is a cancer arising from the hematopoietic stem/progenitor cells (HSPCs). Although ~70% of AML patients achieve complete remission with chemotherapy, at least half will relapse due to the failure to eradicate leukemia stem cells (LSCs). As our previous data indicate the TLR-TAK1/TICAM-1 axes regulate LSCs, we suspect TBK1 also regulates LSCs.
