Buprenorphine, a novel long-acting analgesic, was developed with the intention of two purposes: analgesia and opioid use disorder. Regarding its pharmacodynamics, it is a partial agonist at mu receptors, an inverse agonist at kappa receptors, and an antagonist at delta receptors. For the purpose of analgesia, three formulations of buprenorphine were developed: IV/IM injectable formulation (Buprenex®), transdermal patch formulation (Butrans®), and buccal film formulation (Belbuca®). Related to opioid dependence, the formulations developed were subcutaneous extended release (Sublocade®), subdermal implant (Probuphine®), and sublingual tablets (Subutex®). Lastly, in order to avoid misuse of buprenorphine for opioid dependence, two combination formulations paired with …

When a global pandemic hits during a longitudinal study of biology student success, researchers can unearth rich information about student resilience. By sharing case studies from two demographically different midsized 4-year institutions, this article illustrates the aspects of student self-efficacy beliefs that were undercut by the shift to emergency remote instruction (ERI) in introductory biology courses in Spring 2020: agency and belonging. By assessing student predictions of exam performance and analyzing themes from 276 student narrative surveys, we highlight the power of a careful balance between cognitive and social interventions to help students recover. Students in this study showed a …

Stroke is a devastating neurological event with limited treatment opportunities. Recent advances in understanding the underlying pathogenesis of cerebral ischemia support the involvement of multiple biochemical pathways in the development of the ischemic damage. Fenamates are classical non-steroidal anti-inflammatory drugs but they are also highly subunit-selective modulators of GABAA receptors, activators of IKS potassium channels and antagonists of non-selective cation channels and the NLRP3 inflammosome. In the present study we investigated the effect of mefenamic acid (MFA) in a rodent model of ischemic stroke and then addressed the underlying pharmacological mechanisms in vitro for its actions in vivo. The efficacy …

Wounds in the mouth, occurring after oral surgery, take time to heal. No ointment can be added to help with the healing process because mouth saliva will constantly wash it away. In order to combat this problem, we propose engineering a normal flora microbe to grow at the site of injury and secrete a recombinant growth factor to promote healing of the damaged tissue. Our goal is to have the yeast Pichia pastoris produce human basic fibroblast growth factor (bFGF), which aids in cellular proliferation. P. pastoris is a good choice for this application because not only is it considered …

OBJECTIVES: In earlier work (Lambert et al., 2017) we demonstrated the efficacy of new landmarks for inferior alveolar nerve (IAN) block injections. While the latter study focused on adult morphology, the landmarks were shown to be effective in cases where the M3/3 were present but unerupted. Based on these result a question arose as to their applicability of the landmarks to children and adolescents, given the known changes in mandibular foramen position during growth. Here we quantify the position of the mandibular foramen relative to standard landmarks during growth and assess the new landmarks applicability for IAN …
Root Analogue Implant Creation: Establishing Procedures For The General Dentist, Alicia Hazdovac, Sarah Osborne, Gary D. Richards
Root Analogue Implant Creation: Establishing Procedures For The General Dentist, Alicia Hazdovac, Sarah Osborne, Gary D. Richards

The possibility of replacing damaged teeth with custom-made root analogue implants was initially suggested around 1969 and reintroduced by Lungren et al. in 1992. The latter authors created custom implants in titanium and reported good osteointegration and aesthetic results. Following this effort there has been an increasing focus on custom root analogue implants as a replacement for ‘screw-type’ implants. Given the availability of software to manipulate 3D image data and the rapid rise of 3D printing technology, the question arises as to whether a general dentist can produce their own implants. Here we test the feasibility of practitioners producing custom …

Recent concerns about the prevalence of obstructive sleep apnea (OSA) and its negative sequelae have led to investigations of how manipulation of the dentofacial complex may affect airway problems. CBCT can be used to measure airway dimensions of patients. Extraction of premolars is indicated as part of orthodontic treatment for patients who have severe crowding or protrusion of incisors. Some have suggested that the reduction in arch circumference after premolar extraction would lead to decreased airway volume, which may predispose a patient for sleep disordered breathing (SDB). The purpose of this study was to determine whether there is a significant …

Nucleoside-diphosphate-kinases (NDKs) are leaderless, multifunctional enzymes. The mode(s) of NDK secretion is currently undefined, while extracellular translocation of bacterial NDKs is critical for avoidance of host pathogen clearance by opportunistic pathogens such as Porphyromonas gingivalis. P. gingivalis-NDK during infection inhibits extracellular-ATP (eATP)/P2X7-receptor mediated cell death in gingival epithelial cells (GECs) via eATP hydrolysis. Furthermore, depletion of pannexin-1-hemichannel (PNX1) coupled with P2X7-receptor blocks the infection-induced eATP release in GECs, and P. gingivalis-NDK impacts this pathway. Ultrastructural and confocal microscopy of P. gingivalis-co-cultured GECs or green-fluorescent-protein (GFP)-P. gingivalis-NDK transfected GECs revealed a perinuclear/cytoplasmic localization of NDK. eATP stimulation induced NDK recruitment to …

The cytolethal distending toxin (Cdt) is produced from a number of bacteria capable of causing infection and inflammatory disease. Our previous studies with Actinobacillus actinomycetemcomitans Cdt demonstrate not only that the active toxin subunit functions as a phosphatidylinositol-3,4,5-triphosphate (PIP3) phosphatase but also that macrophages exposed to the toxin were stimulated to produce proinflammatory cytokines. We now demonstrate that the Cdt-induced proinflammatory response involves the activation of the NLRP3 inflammasome. Specific inhibitors and short hairpin RNA (shRNA) were employed to demonstrate requirements for NLRP3 and ASC as well as caspase-1. Furthermore, Cdt-mediated inflammasome activation is dependent upon upstream signals, including reactive …
