Background: When determining eligibility for kidney transplantation, body mass index (BMI) is often a debatable criterion for transplant due to the clinical indication, an elevated BMI has adverse effects on kidney transplant outcomes. There is a current gap in research on the effect BMI has on kidney transplant outcomes.

This dissertation examines the role of CXCR2, a seven transmembrane G- protein coupled receptor, in mediating autocrine as well as paracrine mechanisms during pancreatic cancer progression. Data presented in the initial section demonstrates the aberrant expression of the CXCR2 biological axis in human pancreatic cancer tissue specimens. A study performed within the first section of this dissertation investigates the contribution of CXCR2 signaling in pancreatic cancer initiation. These studies have identified a novel role of CXCR2 in mediating KRAS(G12D) -induced autocrine growth transformation of pancreatic cancer cells. The highly invasive grass species, giant reed (Arundo donax), has been a major contributor to riparian habitat degradation in California for over 50 years. Several modes of vegetative reproduction have allowed this alien species to take advantage of fluvial processes and rapidly spread within California watersheds. A hallmark of cardiac development is the formation of myocardial trabeculations exclusively from the luminal surface of the primitive heart tube. Although a number of genetic defects in the endocardium (Grego-Bessa et al., 2007; Liu et al., 2010) and cardiac jelly (Camenisch et al., 2000) disrupt myocardial trabeculation, the role of cell polarity machinery in driving this process remains unclear. The high degree of heterogeneity observed in breast cancers makes it very difficult to classify cancer patients into distinct clinical subgroups and consequently limits the ability to devise effective therapeutic strategies. In this study, we explore the use of gene mutation profiles to classify, characterize and predict the subgroups of breast cancers. We analyzed the whole exome sequencing data from 358 ethnically similar breast cancer patients in The Cancer Genome Atlas (TCGA) project. Identified somatic and non-synonymous single nucleotide variants were assigned a quantitative score (C-score) that represents the extent of negative impact on the function of the gene. Using …

Over activation of the sympathetic nervous system is prevalent in many forms of cardiovascular disease such as chronic heart failure (CHF) and hypertension. Although increased neuronal renin-angiotensin system activity in presympathetic neurons has been well implicated in mediating this sympatho-excitation, many of the neuronal effects of angiotensin II (Ang II) signaling remain poorly understood. One particular mechanism of Ang II-mediated increases in presympathetic neuronal activity is through reductions in voltage-gated K+ currents. The transmembrane mucin MUC4 is a high molecular weight glycoprotein that is expressed de novo in pancreatic ductal adenocarcinoma (PDAC). MUC4 has been shown to play a tumor-promoting role in malignancies such as PDAC, ovarian cancer and breast cancer. Unlike the normal pancreas, MUC4 is ordinarily expressed by goblet and absorptive cells in the normal colonic epithelium. However, its expression/role in colorectal cancer (CRC) is not well studied.

Endocytic trafficking is an essential process in eukaryotic cells, specifically for the transport of nutrients, membrane components, and receptors. Cargo destined for endocytic traffic is internalized at the cell surface via clathrin-dependent and clathrin-independent pathways, and brought to the early or sorting endosomes. From there, cargo is further trafficked to lysosomes for degradation, trafficked to other compartments in the cell, or recycled back to the cell surface (either directly or via the endocytic recycling compartment).

This study quantifies the changes in vegetation composition and structure of a fire-excluded Pinus palustris (longleaf pine) wetland savanna restoration site in southeastern in comparison to a proximate contemporary reference site. The restoration site was invaded by hardwood species and off-site pines, and never underwent extensive soil disturbance. The restoration treatments involved logging across portions of the site and the reintroduction of fire across the entire site. All species present in 10m2 quadrats were recorded prior to treatment and throughout the 17 year study at reference and treatment sites. P53 is a tumor suppressor protein, which functions in maintaining the cell cycle. When p53 loses its function, cells may multiply at an uncontrolled rate and form tumors. Prompted by findings that TGFβ stimulates thymine DNA glycosylase (TDG) dependent rapid DNA demethylation and activation of the CDKN2B gene, I investigated the global role of TDG and DNA demethylation in TGFβ signaling in HaCaT cells. Using dot blot analysis, I show that TGFβ treatment increases the global levels of 5-formylcytosine, an intermediate metabolite of active DNA demethylation. Characterization of genomic regions that undergo DNA demethylation and recruitment of TDG indicate that they are both frequent events, but only overlap at 11 genomic locations. Even in the absence of direct killing, predators have a pervasive effect on prey populations through costly anti-predator behavioural responses. In high risk environments, animals can reduce conspicuous behaviours such as courtship displays decreasing exposure at the cost of reproduction. Previous studies typically looked at male behaviour immediately following a predator cue, thus, not considering temporal variations in risk or the impact of female receptivity on male courtship. CK2 is a ubiquitously expressed and constitutively active serine/threonine protein kinase that is implicated in many cellular functions. Previous studies have indicated that the generation of mutants that are less sensitive to inhibition can be advantageous when studying protein kinases. Importantly, studies have demonstrated that mutants of CK2 rendered less sensitive to inhibition are attainable. To extend these observations, mutants of CK2α were designed and evaluated to test their effect on the inhibition of CK2 by CX-4945 using in vitro enzymatic assays followed by the development of inducible cell lines. Bone development and repair involve complex processes that include interaction between cells and their surrounding matrix. In the body, bone sialoprotein (BSP) expression is up-regulated at the onset of mineralization. BSP is a multifunctional acidic phosphoprotein with collagen-binding, hydroxyapatite nucleating, and integrin recognition (RGD sequence, which is important for cell-attachment and signaling) regions. Mice lacking BSP expression (Bsp-/-), exhibit a bone phenotype with reductions in bone mineral density, bone length, osteoclast activation, and impaired bone healing. I have two aims with the publication of this thesis. The first is to contribute to the recent scholarly work that seeks to discredit the popular, yet fallacious, warfare model of science and religion. A nuanced investigation shows that the warfare evident in popular discourse cannot easily be attributed to a conflict between the essence of science and religion. Rather “science” and “religion” are interpreted as something else entirely. The New Atheists equate science with scientism, and religion is often associated with the Biblical inerrantism of many Protestant Christians. The Venus flytrap (Dionaea muscipula), first described by Charles Darwin, is a unique carnivorous plant which acquires prey using leaf blades modified into snap-traps. A recent study showed that prey capture by flytraps was not selective. With this study we examined prey capture rates further by analyzing D. muscipula traps less than 1 em in length to determine if juvenile flytraps differ from adults. Similar to the earlier findings, trap size appeared to play no role in prey capture success. There are, however, plants recalcitrant to established transformation methods, requiring the implementation of novel techniques. This study investigates the viability of cell-penetrating peptide (CPP) transformation using Tat2, a CPP with demonstrated transformation efficiency in plant and animal systems, in two regenerable tissue culture systems, soybean somatic embryos and Arabidopsis protoplasts. In 2000 following a 100-year record drought, approximately 8,000 hectares of S. alterniflora died along the Louisiana coast, which prompted Louisiana Governor Foster to declare a state of emergency. In the United States, colorectal cancer (CRC) is the third most diagnosed cancer and the second leading cause of cancer deaths. In 2015, approximately 143,000 new cases will be diagnosed and 50,000 deaths are predicted. Liver metastasis is the major cause of death. To understand the molecular and cellular mechanisms of liver metastasis, we used an orthotopic mouse model of CRC liver metastasis that recapitulated all stages of tumor growth and metastasis to perform whole genome microarray analysis to identify genetic changes in the liver microenvironment in response to primary tumor growth in the cecum. An integral aspect of innate immune response to viral infections is the ability to detect non-self molecules to initiate antiviral signaling via pattern recognition receptors (PRRs). One subset of these receptors are cytoplasmic receptors that contain double stranded (dsRNA) binding domains, which allow them to identify non-self dsRNA produced during a viral infection and mount a protective cellular response. PKR is a dsRNA-activated eIF2α kinase that is a key regulator of cellular antiviral and stress response pathways. Viruses are the most numerous biological entities in the ocean, playing a key role in microbial ecology and biogeochemical cycling. In order to replicate, viruses must first infect a host and then use the host’s cellular machinery to produce new viral particles. Some of the most abundant viral hosts in the subtropical open ocean are picophytoplankton. However, picophytoplankton growth is typically constrained by nutrient availability. In many regions of the ocean, iron (Fe) is only available in very low concentrations causing growth-limitation of picophytoplankton. Predators kill, but the risk of being killed is also a powerful force affecting survival because scared prey eat less, thereby increasing the likelihood of starvation. Young of most animals are extremely vulnerable to predators and may alter their behaviour to limit detection. I investigated the previously unexplored effects that predation risk has on the behaviour of newly fledged offspring and their parents, and the impact this has on offspring survival. I manipulated predation risk using sound and found that parent song sparrows reduced their feedings, providing 60% less food overall. Histone deacetylases have important roles in development and stress response in plants. To further investigate their function, the HD2D gene, of the plant specific HD2 family, was studied. An hd2d-1 mutant and two HD2D overexpression lines were used in this study. Germination was delayed in hd2d-1 and HD2D overexpression seeds only in the presence of ABA. HD2D was found to positively regulate the expression of members of the ABA-response pathway (ABI1, ABI5, and RD29A) leading to increased resistance to drought and salinity treatments.
