Interaction Of Human Follicle Stimulating Hormone Receptor With Caveolin Within The Cell Membrane, Jordan Pereira

The human FSH receptor is a g protein-coupled receptor (GPCR) expressed on the surface of granulosa cells in the ovary and Sertoli cells in the testes. FSHR has a sequence of amino acids consistent with a caveolin interaction motif (φXφXXXXφXXφ) found between amino acids 479-489 in the primary receptor sequence in the putative 4th transmembrane domain. Caveolin is a protein found in cell membrane micro domains such as lipid rafts. These densely packed regions of the membrane are enriched for sphingolipids and cholesterol and are thought to be involved in signal transduction. The blacklegged tick (Ixodes scapularis) is the primary vector of Borrelia burgdorferi, the causative agent of Lyme disease. When a tick is not questing or feeding, the majority of its life is spent within the soil. Abiotic factors within soil have been shown to affect tick molting and survival across all life stages. Soil pH, however, has not been heavily investigated. In this field study, I investigated the effects of soil pH and texture on engorged nymphal ticks. Lipid raft domains have been shown to be important in receptor signaling, especially for G Protein Coupled Receptors (GPCR). Follicle stimulating hormone is a vital reproductive signal that relies on effective signaling of its GPCR. The goal of this study was to test whether or not the follicle stimulating hormone receptor (FSHR) utilizes lipid raft domains when signaling. The main methods used were fluorescent antibodies and various forms of microscopy, including confocal microscopy. Lizards of the genus Anolis communicate through visual displays involving the opening and closing of an expandable flap of skin, called a dewlap, located on their throat. In some habitats up to ten species of Anolis are found in close proximity to one another. For a dewlap color to be effective an individual of the same species should be able to easily distinguish it from the dewlap color of other species located in the same habitat. Ixodes scapularis, or the black-legged tick, is the major vector of Lyme disease in the U.S. I. scapularis has expanded its range in recent decades, making the study of factors affecting its distribution a high priority. Studying the effects of various conditions in the soil could help in predicting range expansions, because ticks spend the majority of their lives in contact with the soil. Karenia brevis is species of dinoflagellate responsible for most of the harmful algal blooms that occur in the Gulf of Mexico. These blooms can be detrimental to the environment and the economy of a coastal region due to the brevetoxins produced by Karenia brevis. Currently, the cause of these blooms, as well as the mechanisms of associated toxin production, are unknown. Efforts to characterize Karenia brevis at a molecular level are ongoing. West Nile virus (WNV), a positive sense single stranded RNA flavivirus, is prevalent in many countries worldwide and is a growing threat in the United States. Some patients infected with WNV develop severe neuroinvasive disease including meningitis or encephalitis which may lead to death. The recently characterized cytokine interleukin-17 (IL-17) has been implicated in several autoimmune diseases, neuroinflammatory conditions, and immune responses to various microbial infections. Biodegradability is an important consideration in the design of novel chemicals. At the end of their function, chemicals should break down into harmless degradation products to avoid toxic effects as a result of environmental persistence. To test the biodegradability potential of novel chemicals, a protocol designed by the Organisation for Economic and Cooperative Development (OECD) is used. This protocol states that in a mixture of the chemical of interest, a microbial inoculate, and mineral media, 70% of the chemical must degrade within a 10-day window of a 28-day experiment in order to be considered readily biodegradable. Neurotrophic factors are proteins that play an important role in the development and maintenance of neurons. Recent studies have shown that neurotrophic factors may hold promise for treating damage to the nervous system caused by trauma or diseases, such as amyotrophic lateral sclerosis or Parkinson’s disease. Glial cell line-derived neurotrophic factor (GDNF) is expressed in skeletal muscles and affects peripheral motor neurons. The theory of sexual selection has proven to be an important factor in understanding the mechanisms behind the evolution of secondary sexual traits. The Hamilton and Zuk (1982) hypothesis of parasite-mediated sexual selection postulates that birds evolved ornamentation to indicate parasite resistance to potential mates. My research explores the relationship between parasite load and plumage coloration in hunter-harvested mourning doves. Plumage from seven body areas was collected, ectoparasites were quantified and blood smears made so that intracellular hematozoan parasites could be quantified. Due to the prevalence of diabetes and lack of simple endocrine experiments, a laboratory protocol was developed in order to increase student knowledge of the disease and the diabetic testing process. In this paper, we present the protocol for manipulating the glucose concentration of commercially available bovine blood. The bovine blood was manipulated using Ringer's solution to which glucose was added, and six samples were prepared (pre-prandial and postprandial, normal; pre-prandial and postprandial, pre-diabetic; pre-prandial and postprandial, diabetic). Concerns over food production and processing, along with increasing outbreaks of foodborne illnesses and antibiotic resistant infections have led many consumers to seek alternative food sources in the organic and local food markets. This study compared conventional, USDA certified organic, and local farmer's market milk types to determine whether there is any noticeable benefit to purchasing organic or locally harvested milk in terms of their bacterial populations and level of antibiotic resistance. Octocorals are a diverse group of organisms, and within the Caribbean they contribute substantially to coral-reef cover. In this thesis, two different studies performed on these marine organisms are detailed. In Section I, the percent calcium carbonate composition of several species was analyzed. Octocorals produce sclerites: calcium carbonate crystals that serve as the skeletal elements of the coral. Determining the percent calcium carbonate composition of eight Caribbean octocoral species can give comparative insight into the form and function in these animals.
