Mechanomyography; Lasers For Non-Invasive Quantification Of Muscle Recovery From Exercise Induced Fatigue, Nicholas Rosser, Darryl Mcandrew, Donald Iverson

Darryl J McAndrew

Muscle fatigue is a common clinical sign and possible precursor to musculoskeletal injury. Existing methods of detecting muscle fatigue qualify changes within muscle or across joints, but are expensive, invasive or difficult to use. A technique of mechanomyography utilising maximal percutaneous stimulation (known as the Muscle Displacement Laser {MDL}) is an easy non-invasive assessment tool, which may have considerable application in measuring muscle fatigue and determining recovery. Specifically, this study attempted to validate the MDL as a tool to measure muscle fatigue and determine the time-course of recovery from Delayed Onset of Muscle Soreness in human biceps brachii muscle. Methods: …

Teasaponin Improves Central Leptin Sensitivity In High-Fat Diet-Induced Obese Mice, Y Wu, Xu-Feng Huang, Y Yu Sep 2013

Teasaponin Improves Central Leptin Sensitivity In High-Fat Diet-Induced Obese Mice, Y Wu, Xu-Feng Huang, Y Yu

Yizhen Wu

Purpose: Leptin promotes negative energy balance by suppressing energy intake (EI) and elevating energy expenditure through its central action. Central leptin resistance is a hallmark of diet-induced obesity (DIO). Oral administration of teasaponin (TS) can reduce body weight and fat mass. However, little is known if TS has benefit effect in improving central leptin sensitivity. This study investigated the effects of TS on central leptin sensitivity and leptin signalling in the hypothalamic arcuate nucleus (Arc) of DIO mice. Methods: After 15 weeks of high-fat diet, DIO mice (n=40) were divided into two groups received either intraperitoneal (ip) injection of TS …

Time-Dependent Ghrelinergic Signaling And Hyperphagia Induced By Olanzapine, Q Zhang, M He, J Lian, C Deng, Xu-Feng Huang Aug 2013

Time-Dependent Ghrelinergic Signaling And Hyperphagia Induced By Olanzapine, Q Zhang, M He, J Lian, C Deng, Xu-Feng Huang

Qingsheng Zhang

Olanzapine is widely used to treat schizophrenia and other mental diseases. Interestingly, although increased body weight is evident throughout the course of olanzapine treatment, elevated food intake only appears in relatively shorter term. Olanzapine can modulate ghrelin levels, which may upregulate neuropeptide Y (NPY) secretion at hypothalamus. Brain-specific homeobox transcription factor (BSX) may also be involved in ghrelin-induced hyperphagia. PURPOSE: To investigate the effect of olanzapine on central NPY, BSX and plasma ghrelin levels, and their relationship to food intake. METHODS: Rats were treated with olanzapine (1mg/kg, orally, 3x/day, n=12/group) or vehicle for 1 week, 2 weeks, or 5 weeks. …

Ozone-Induced Dissociation On A Modified Tandem Linear Ion-Trap: Observations Of Different Reactivity For Isomeric Lipids, Berwyck Poad, Huong Pham, Michael Thomas, Jessica Hughes, J Campbell, Todd Mitchell, Stephen Blanksby May 2013

Ozone-Induced Dissociation On A Modified Tandem Linear Ion-Trap: Observations Of Different Reactivity For Isomeric Lipids, Berwyck Poad, Huong Pham, Michael Thomas, Jessica Hughes, J Campbell, Todd Mitchell, Stephen Blanksby

Berwyck L. J. Poad

Ozone-induced dissociation (OzID) exploits the gas-phase reaction between mass-selected lipid ions and ozone vapor to determine the position(s) of unsaturation. In this contribution, we describe the modification of a tandem linear ion-trap mass spectrometer specifically for OzID analyses wherein ozone vapor is supplied to the collision cell. This instrumental configuration provides spatial separation between mass-selection, the ozonolysis reaction, and mass-analysis steps in the OzID process and thus delivers significant enhancements in speed and sensitivity (ca. 30-fold). These improvements allow spectra revealing the double-bond position(s) within unsaturated lipids to be acquired within 1 s: significantly enhancing the utility of OzID in …

Developmental Vitamin D Deficiency Alters Mk-801-Induced Behaviours In Adult Offspring, James Kesby, Jonathan O'Loan, Suzanne Alexander, Chao Deng, Xu-Feng Huang, John Mcgrath, Darryl Eyles, Thomas Burne Feb 2013

Developmental Vitamin D Deficiency Alters Mk-801-Induced Behaviours In Adult Offspring, James Kesby, Jonathan O'Loan, Suzanne Alexander, Chao Deng, Xu-Feng Huang, John Mcgrath, Darryl Eyles, Thomas Burne

Xu-Feng Huang

Rationale Developmental vitamin D (DVD) deficiency is a candidate risk factor for developing schizophrenia in humans. In rodents DVD deficiency induces subtle changes in the way the brain develops. This early developmental insult leads to select behavioural changes in the adult, such as an enhanced response to amphetamine-induced locomotion in female DVD-deficient rats but not in male DVDdeficient rats and an enhanced locomotor response to the Nmethyl- D-aspartate (NMDA) receptor antagonist, MK-801, in male DVD-deficient rats. However, the response to MK- 801-induced locomotion in female DVD-deficient rats is unknown. Therefore, the aim of the current study was to further examine …

Ventromedial Hypothalamic Npy Y2 Receptor In The Maintenance Of Body Weight In Diet-Induced Obesity In Mice, Yulin Li, Xu-Feng Huang, Chao Deng, Qing Wang, Timothy South, Yinghua Yu

Xu-Feng Huang

This study examined changes in neuropeptide Y (NPY) Y2 receptor binding in the brains of C57BL/6 mice in response to different levels of high-fat diets via three dietary intervention methods: high-fat diet, switching from high- to low-fat diet and finally, energy restricted high-fat diet. Forty-five C57Bl/6 male mice were fed a high-fat diet for 8 weeks and then classified as diet-induced obese (DIO) or diet-resistant (DR) mice according to the highest and lowest body weight gainers, respectively. The DIO and DR mice were then randomly divided into three groups each and either continued on their high-fat diet ad libitum (DIO-H …

Temporal And Site-Specific Brain Alterations In Cb1 Receptor Binding In High Fat Diet-Induced Obesity In C57bl/6 Mice, Xu-Feng Huang, Timothy South

Xu-Feng Huang

The cannabinoid CB1 receptor has been implicated in the regulation of appetite and the consumption of palatable foods. This experiment aimed to explore the involvement of the CB1 receptor in the early and late stages of high fat diet-induced obesity in C57BL/6 mice. The C57Bl/6 mice were placed on a high fat (HF) or low fat/high carbohydrate (LF) diet for 3 or 20 weeks. Quantitative autoradiography revealed that binding of [3H] CP-55,940 (CB1 receptor ligand) was elevated following 3 weeks of HF feeding in areas including the medial/ventral anterior olfactory nucleus (22.1%), agranular insular cortex (24.0%) and the hypothalamus (31.5%) …

Brain-derived neurotrophic factor (BDNF) and its receptor tyrosine kinase B (TrkB) are closely associated with the regulation of energy homeostasis, but their roles in diet-induced obesity have not been explored. Using dietary interventions, this study examined regional changes of BDNF and TrkB mRNA expression in different brain regions of diet-induced obese (DIO) and resistant (DR) mice in response to high-fat (HF), energy-restricted pair-feeding and low fat (LF) diets. Using in situ hybridization, DIO mice had significantly decreased levels of BDNF mRNA expression (¿32% to ¿37%) and TrkB (¿21% to ¿23%) in the hippocampus compared to DR mice on an HF …

Decreased Plasma Peptide Yy Accompanied By Elevated Peptide Yy And Y2 Receptor Binding Densities In The Medulla Oblongata Of Diet-Induced Obese Mice, Xu-Feng Huang, Chao Deng, Yean Tan, Gita Rahardjo

Xu-Feng Huang

It is well known that the peripheral peptide YY (PYY)-central neuropeptide Y (NPY) Y2 receptor axis plays an important role in promoting negative energy balance regulation. Both the hypothalamus and medulla oblongata express a high level of Y2 receptors; however, the functional role of this receptor in chronic high-fat diet-induced obesity has not been fully examined. Using quantitative autoradiography, this study measured binding densities of total [125I]PYY and Y2 receptors in the hypothalamus and medulla of chronic high-fat diet-induced obese (DIO), obese-resistant, and low-fat-fed mice. Plasma PYY was also measured using RIA after 22 wk of dietary intervention. The results …
