Pmr6, A Pectate Lyase–Like Gene Required For Powdery Mildew Susceptibility In Arabidopsis, John P. Vogel, Ted K. Raab, Celine Schiff, Shauna C. Somerville

Ted K. Raab

The plant genes required for the growth and reproduction of plant pathogens are largely unknown. In an effort to identify these genes, we isolated Arabidopsis mutants that do not support the normal growth of the powdery mildew pathogen Erysiphe cichoracearum. Here, we report on the cloning and characterization of one of these genes, PMR6. PMR6 encodes a pectate lyase-like protein with a novel C-terminal domain. Consistent with its predicted gene function, mutations in PMR6 alter the composition of the plant cell wall, as shown by Fourier transform infrared spectroscopy. pmr6-mediated resistance requires neither salicylic acid nor the ability to perceive …

The model VFSMOD was used to investigate the effect of varying watershed characteristics and buffer dimensions on the sediment trapping efficiency of vegetative filters. This investigation allows for a better understanding of how watershed characteristics, buffer dimensions, and storm characteristics impact the performance of vegetative filters. Using VFSMOD, relationships that estimate sediment trapping efficiency from the ratio of filter area and field area were developed for different sites. These relationships have been applied for field assessment of concentrated flow.

Reactivity Profiles Of Ligands Of Mammalian Retinoic Acid Receptors: A Preliminary Corepa Analysis, Steven P. Bradbury, G. T. Ankley, O. G. Mekenyan, V. . Kamenska, P. K. Schmieder Jan 2002

Retinoic acid and associated derivatives comprise a class of endogenous hormones that bind to and activate different families of retinoic acid receptors (RARs, RXRs), and control many aspects of vertebrate development. Identification of potential RAR and RXR ligands is of interest both from a pharmaceutical and toxicological perspective. The recently developed COREPA (COmmon REactivity PAttern) algorithm was used to establish reactivity profiles for a limited data set of retinoid receptor ligands in terms of activation of three RARs (a, b, g) and an RXR (a). Conformational analysis of a training set of retinoids and related analogues in terms of thermodynamic …

Narrowing Historical Uncertainty: Probabilistic Classification Of Ambiguously Identified Tree Species In Historical Forest Survey Data, David J. Mladenoff, Sally E. Dahir, Eric V. Nordheim, Lisa A. Schulte, Glenn G. Guntenspergen Jan 2002

Narrowing Historical Uncertainty: Probabilistic Classification Of Ambiguously Identified Tree Species In Historical Forest Survey Data, David J. Mladenoff, Sally E. Dahir, Eric V. Nordheim, Lisa A. Schulte, Glenn G. Guntenspergen

Historical data have increasingly become appreciated for insight into the past conditions of ecosystems. Uses of such data include assessing the extent of ecosystem change; deriving ecological baselines for management, restoration, and modeling; and assessing the importance of past conditions on the composition and function of current systems. One historical data set of this type is the Public Land Survey (PLS) of the United States General Land Office, which contains data on multiple tree species, sizes, and distances recorded at each survey point, located at half-mile (0.8-km) intervals on a 1-mi (1.6 km) grid. This survey method was begun in …

Familial Als-Associated Mutations Decrease The Thermal Stability Of Distinctly Metallated Species Of Human Copper/Zinc Superoxide Dismutase, Daryl K. Eggers, J. A. Rodriguez, J. S. Valentine, J. A. Roe, A. Tiwari, R. H. Brown, L. J. Hayward Jan 2002

Familial Als-Associated Mutations Decrease The Thermal Stability Of Distinctly Metallated Species Of Human Copper/Zinc Superoxide Dismutase, Daryl K. Eggers, J. A. Rodriguez, J. S. Valentine, J. A. Roe, A. Tiwari, R. H. Brown, L. J. Hayward

Daryl K. Eggers

We report the thermal stability of wild type (WT) and 14 different variants of human copper/zinc superoxide dismutase (SOD1) associated with familial amyotrophic lateral sclerosis (FALS). Multiple endothermic unfolding transitions were observed by differential scanning calorimetry for partially metallated SOD1 enzymes isolated from a baculovirus system. We correlated the metal ion contents of SOD1 variants with the occurrence of distinct melting transitions. Altered thermal stability upon reduction of copper with dithionite identified transitions resulting from the unfolding of copper-containing SOD1 species. We demonstrated that copper or zinc binding to a subset of “WT-like” FALS mutants (A4V, L38V, G41S, G72S, D76Y, …
