Characterization Of Extracellular Vesicles By Surface-Enhanced Raman Spectroscopy, Nina M. Culum Sep 2021

Characterization Of Extracellular Vesicles By Surface-Enhanced Raman Spectroscopy, Nina M. Culum

Electronic Thesis and Dissertation Repository

Extracellular vesicles (EVs), which are nanoscale vesicles secreted by cells into biofluids, are of research interest due to their roles in intercellular communication. EVs released from mesenchymal stromal cells (MSCs) have tremendous potential in cell-free regenerative medicine, while EVs released from diseased cells are being studied as biomarkers for minimally invasive and early disease detection. Presented in this thesis are gold nanohole arrays for the capture and sensitive detection of EVs by surface-enhanced Raman spectroscopy (SERS), a plasmonic technique capable of single molecule detection. Herein, we have characterized EVs released from MSCs and ovarian cancer cells, with a focus on …

Chemical Reactions At Metal Surfaces: Functionalization Strategies And Spectroscopic Characterization, Sydney Legge Oct 2020

Chemical Reactions At Metal Surfaces: Functionalization Strategies And Spectroscopic Characterization, Sydney Legge

Electronic Thesis and Dissertation Repository

Tremendous interest in increasing the control over surface properties has led to high demand for new efficient methods for chemical surface modifications. In this work, several approaches toward surface functionalization are explored, and modified surfaces are subsequently characterized using uniquely suited spectroscopic techniques. In the first part of this thesis, strain-promoted alkyne-azide cycloaddition (SPAAC) reactions are investigated for their potential to precisely tune the surface properties of gold substrates at the monolayer level. The utility of SPAAC reactions in preparing biorecognition interfaces for cell adhesion is then examined. Polarization modulation infrared reflection-absorption spectroscopy is used to characterize adsorbed monolayers and …

Multiresonant Anisotropic Nanostructures For Plasmon-Mediated Spectroscopies, Gregory Q. Wallace Mar 2018

Multiresonant Anisotropic Nanostructures For Plasmon-Mediated Spectroscopies, Gregory Q. Wallace

Electronic Thesis and Dissertation Repository

To detect and analyze molecular species of interest, analytical sciences and technologies exploit the variation in the chemical properties associated with the analytes. Techniques involving vibrational spectroscopy rely on the unique response observed when a molecule interacts with light. Although these methods can provide the specificity needed for detection, they are traditionally hindered by the need for large quantities of material, and long acquisition times. To minimize these issues, advancements in plasmon-enhanced techniques, such as surface-enhanced Raman spectroscopy (SERS) and surface-enhanced infrared absorption (SEIRA) are being made. Such techniques make use of the strong interaction between an optical field and …

Design And Fabrication Of Integrated Plasmonic Platforms For Ultra-Sensitive Molecular And Biomolecular Detections, Mohammadali Tabatabaei Aug 2015

Design And Fabrication Of Integrated Plasmonic Platforms For Ultra-Sensitive Molecular And Biomolecular Detections, Mohammadali Tabatabaei

Electronic Thesis and Dissertation Repository

One of the major challenges in analytical and biological sciences is to develop a device to obtain unambiguous chemical and structural properties of a material or a probe biomolecule with high reproducibility and ultra-high sensitivity. Moreover, in addition to such a high sensitivity, other cases such as minimum intrusiveness, small amounts of analyte, and short acquisition time and high reproducibility are key parameters that can be valued in any analytical measurements. Among the promising methods to achieve such endeavor, plasmon-mediated surface-enhanced spectroscopic techniques, such as surface-enhanced Raman spectroscopy (SERS), are considered as suitable options. Such techniques take advantage of the …

Rational Design And Advanced Fabrication Of Metallic Nanostructures For Surface-Enhanced Raman Spectroscopy, Betty Cristina Galarreta Aug 2011

Rational Design And Advanced Fabrication Of Metallic Nanostructures For Surface-Enhanced Raman Spectroscopy, Betty Cristina Galarreta

Electronic Thesis and Dissertation Repository

One of the main challenges in analytical science and technology is to develop devices that provide unambiguously the chemical nature of the material of interest with the minimum intrusiveness, the smallest amount of analyte, and the shortest acquisition time. Among the promising methods for such purpose, optical spectroscopy such as surface-enhanced Raman scattering is considered a suitable option. This spectroscopic technique takes advantage of the interaction between an optical field and metallic nanostructures to magnify the electromagnetic field in the vicinity of the nanostructure, resulting in an amplified signal of the vibrational fingerprints of the adsorbed molecules onto the metallic …
