This Article describes the molecular recognition of peptides containing an N-terminal methionine (Met) by the synthetic receptor cucurbit[8]-uril (Q8) in aqueous solution and with submicromolar affinity. Prior work established that Q8 binds with high affinity to peptides containing aromatic amino acids, either by simultaneous binding of two aromatic residues, one from each of two different peptides, or by simultaneous binding of an aromatic residue and its immediate neighbor on the same peptide. The additional binding interface of two neighboring residues suggested the possibility of targeting nonaromatic peptides, which have thus far bound only weakly to synthetic receptors. The development of novel approaches to signal amplification in aqueous media could enable new diagnostic platforms for the detection of water-soluble analytes, including biomolecules. This paper describes a fluorogenic polymerization approach to amplify initiator signal by the detection of visible fluorescence upon polymerization in real-time. Fluorogenic monomers were synthesized and co-polymerized by atom transfer radical polymerization (ATRP) in water to reveal increasing polymer fluorescence as a function of both reaction time and initiator concentration. The molecular recognition of peptides and proteins in aqueous solution by designed molecules remains an elusive goal with broad implications for basic biochemical research and for sensors and separations technologies. This paper describes the recognition of N-terminal tryptophan in aqueous solution by the synthetic host cucurbit[8]uril (Q8). Q8 is known to form 1:1:1 heteroternary complexes with methyl viologen (MV) and a second aromatic guest. This paper describes the molecular recognition of the tripeptide Tyr-Leu-Ala by the synthetic receptor cucurbit[8]uril (Q8) in aqueous buffer with nanomolar affinity and exceptional specificity. This combination of characteristics, which also applies to antibodies, is desirable for applications in biochemistry and biotechnology but has eluded supramolecular chemists for decades. Building on prior knowledge that Q8 binds to peptides with N-terminal aromatic residues, a library screen of 105 peptides was designed to test the effects of residues adjacent to N-terminal Trp, Phe, or Tyr. Mitochondria, vital organelles existing in almost all eukaryotic cells, play a crucial role in energy metabolism and apoptosis of aerobic organisms. In this work, we report two new flavone-based fluorescent probes, MC-Mito1 and MC-Mito2, for monitoring mitochondria in living cells. These two probes exhibit remarkably low toxicity, good cell permeability, and high specificity; these probes complement the existing library of mitochondrial imaging agents. The new dyes give nearly no background fluorescence, and their application does not require tedious postwashing after cell staining. The appreciable tolerance of MC-Mito2 encourages a broader range of biological applications for understanding the cell degeneration and …

We report the photophysical properties, binding-induced turn-on emission, and fluorescence imaging of the cellular uptake and distribution of lapatinib, an EGFR/ERBB inhibitor. Lapatinib, a type II, i.e. inactive state, inhibitor that targets the ATP binding pocket of the EGFR family of receptor tyrosine kinases. DFT calculations predict that the 6-furanylquinazoline core of lapatinib should exhibit an excited state with charge transfer character and an S0 to S1 transition energy of 3.4 eV. Absorption confirms an optical tran- sition in the near UV to violet, while fluorescence spectroscopy shows that photoemission is highly sensi- tive to solvent polarity. The hydrophobicity of …

Samples taken at different points in the wastewater treatment process of a triazine-manufacturing plant were scanned by fluorescence spectroscopy, in the wavelength range of 200-900 nm. Reproducibly, the fluorescence spectra revealed one single major peak at excitation and emission wavelengths of 258 and 370 nm respectively. Aqueous solutions of purified active compounds, including Atrazine, Propazine, Simazine, Terbuthylazine, Metolachlor, and Benoxacor, were also scanned. No significant fluorescence was observed in these standard solutions at concentrations up to 100 mg/L. Selected plant samples as well as standard solutions of Atrazine, Metolachlor, and toluene were further analyzed using high-performance liquid chromatography with absorbance …

A simple salicylic acid selective fluorescence receptor 1 was designed by combining 1-pyrenecarboxaldehyde and 1-(3-aminopropyl)imidazole. The selective sensing of salicylic acid resulted in a significant increase in monomer emissions due to the p–p interactions between the benzene and pyrene rings. The nature of the interactions between receptor 1 and salicylic acid was investigated further by 1H NMR spectroscopy, and the energy minimised structure of the complex between receptor 1 and salicylic acid was optimised. Boronic acids are versatile reagents for the chemical synthesis of organic molecules. They and other boron-containing compounds can be detected readily by the interruption of the excited-state intramolecular proton transfer (ESIPT) of 10-hydroxybenzo[h]quinolone. This method is highly sensitive and selective, and useful for monitoring synthetic reactions and detecting boron-containing compounds on a solid support.

The properties of two forms of polyaniline (PAni) synthesised under acidic and basic conditions have been investigated both individually and as combined complexes. The PAni polymerised within alkaline media was redox inactive and non-conducting while the PAni emeraldine salt (ES) was electroactive and conducting. Raman, electron spin resonance, UV-Vis and fluorescence spectroscopies were used to monitor the changes in electronic properties of these conducting polymer composites. Solution cast films of alkaline synthesised (A-PAni) with the PAni ES resulted in an increase in the high spin polaron population suggesting that it acts as a pseudodopant. We present experimental and theoretical studies of two dimensional periodic arrays of elliptical plasmonic patch nanoantennas. Experimental and simulation results demonstrate that the azimuthal symmetry breaking of the metal patches leads to the occurrence of even and odd resonant cavity modes and the excitation geometries dependent on their modal symmetries. We show that the cavity modes can be described by the product of radial and angular Mathieu functions with excellent agreements with both simulations and experiments. The effects of the patch periodicity on the excitation of the surface plasmon and its coupling with the cavity modes are also discussed.

Thioflavin T (ThT) dye fluorescence is used regularly to quantify the formation and inhibition of amyloid fibrils in the presence of anti-amyloidogenic compounds such as polyphenols. However, in this study, it was shown, using three polyphenolics (curcumin, quercetin and resveratrol), that ThT fluorescence should be used with caution in the presence of such exogenous compounds. The strong absorptive and fluorescent properties of quercetin and curcumin were found to significantly bias the ThT fluorescence readings in both in situ real-time ThT assays and single time-point dilution ThT-type assays. Sulforhodamine B (SRB), a common fluorescent dye, is often considered to be a purely hydrophilic molecule, having no impact on bulk or interfacial properties of aqueous solutions. This assumption is due to the high water solubility of SRB relative to most fluorescent probes. However, in the present study, we demonstrate that SRB is in fact an amphiphile, with the ability to adsorb at an air/water interface and to incorporate into sodium dodecyl sulfate (SDS) micelles. A new fluorescent imidazolium-based cholestane receptor 4 bearing a pyrene moiety was synthesized. The binding ability of 4 toward various dicarboxylic acids was examined by UV–vis and fluorescence spectroscopy. Receptor 4 showed the highest binding constant for oxalic acid among all the tested dicarboxylic acids. Oxalic acid formed a complex with 4 with a 1:2 ratio in ethanol.

In this research, laser-excited fluorescence was examined for sensitive detection of aqueous dityrosine. Samples were excited with a 6.3-mW, 325-nm helium-cadmium laser focused into a small volume-fluorescence cell with a 10-cm lens. The resulting fluorescence emission was collected perpendicular to the excitation and detected with two different schemes. An optical bandpass filter was used with a photomultiplier tube for sensitive quantitative measurement, while a photodiode array detector was used in conjunction with a spectrograph for qualitative characterization of fluorescence emission spectra. The CFCl radical has been produced in the gas phase by both IR multiphotonphotolysis of C2F3Cl and He metastable reaction with C2F3Cl. Single vibronic level fluorescence and excitation spectra taken of this species have yielded sufficient information to determine certain vibronic constants. The excited state was found to have an origin of T0=25 283±5 cm−1 with vibrational frequencies: ω2′=394±3 cm−1 and ω3′=739±5 cm−1.
