Twin-Rainbow Metrology. I. Measurement Of The Thickness Of A Thin Liquid Film Draining Under Gravity, Charles L. Adler, James A. Lock, Ian P. Rafferty, Wayne Hickok

We describe twin-rainbow metrology, a new optical technique used to measure the thickness of thin films in a cylindrical geometry. We also present an application of the technique: measurement of the thickness of a Newtonian fluid draining under gravity. We compare these measurements with fluid mechanics models. A systematic analysis of the mode structure of diffusive relaxations in 1 MDa hydroxypropylcellulose(HPC):water is presented. New methods and data include (1) use of integral spectral moments to characterize nonexponential decays, (2) spectra of small probes in concentrated HPC solutions, (3) temperature dependence of the mode structure, and (4) comparison of optical probe spectra and spectra of probe-free polymer solutions. We find that (1) probe and polymer relaxations are in general not the same; (2) the apparent viscometric crossover near ct≈6 g/l is echoed by probe behavior; (3) our HPC solutions have a characteristic dynamic length, namely the 50 nm …

In this work, we present the development and validation of a tandem mass spectrometry method for the quantitative determination of methoxyamine (CH3ONH2), a potential new chemotherapeutic agent, in human and mouse plasma. Methoxyamine together with the internal standard (I.S.) methoxyl-D3-amine was directly derivatized in plasma sample with a novel chemical agent 4-(N,N-diethylamino)benzaldehyde. The product solution was injected into an on-line Oasis® HLB extraction column ( mm) for analyte extraction. After the elution of extractives, the derivatized analytes were monitored by the positive-electrospray-ionization mass spectrometry (ESI-MS-MS). Thrombin activated factor Va (factor VIIa, residues 1-709 and 1546-2196) has an apparent dissociation constant (Kd,app) for factor Xa within prothrombinase of approximately 0.5 nM. A protease (NN) purified from the venom of the snake Naja nigricollis nigricollis, cleaves human factor V at Asp697, Asp1509, and Asp1514 to produce a molecule (factor VNN) that is composed of a Mr 100,000 heavy chain (amino acid residues 1-696) and a Mr 80,000 light chain (amino acid residues 1509/1514-2196). Factor VNN, has a Kd,app for factor Xa of 4 nm and reduced clotting activity. Cleavage of factor VIIa by NN at Asp697 results …

We have recently demonstrated that amino acid region 323-331 of factor Va heavy chain (9 amino acids, AP4') contains a binding site for factor Xa (Kalafatis, M., and Beck, D. O. (2002) Biochemistry 41, 12715-12728). To ascertain which amino acids within this region are important for the effector and receptor properties of the cofactor with respect to factor Xa, we have synthesized three overlapping peptides (5 amino acids each) spanning the amino acid region 323-331 and tested them for their effect on prothrombinase complex assembly and function. Mixing is an integral component of most polymer processing operations as material properties are highly influenced by the quality of mixing. The degree of distributive mixing (system homogeneity) is assessed by calculating the evolution of Renyi relative entropies for the minor component along a continuous processing equipment. The Renyi entropy involves a β parameter, which represents weighting given to the concentration of the minor component in small, localized regions. Different aspects of mixing can thus be analyzed, from the amount of void spaces to the concentration of the region where mixing is the worst in terms of the minor component. …

A space-borne optical tweezer apparatus for use with colloidal crystallization experiments has been characterized. The trapping force has been measured as a function of index mismatch between colloidal microspheres and the surrounding fluid and as a function of particle size. This work also presents a method to determine the refractive index of a colloidal microsphere, which is then used to calculate the applied trapping force for the case of an arbitrary background fluid. This is useful for work with dense colloidal suspensions when the usual (e.g., Stokes flow) trap force measurement methods do not apply, as well as microrheological studies …

The transient control of diverse biological responses that occurs in response to varied forms of stress is often a highly regulated process. During the interferon (IFN) response, translational repression due to phosphorylation of eukaryotic initiation factor 2α, eIF2α, by the double-stranded RNA-dependent protein kinase, PKR, constitutes a means of inhibiting viral replication. Here we show that the transient nature of the IFN response against acute viral infections is regulated, at least in part, by RNase L. During the IFN antiviral response in RNase L-null cells, PKR mRNA stability was enhanced, PKR induction was increased, and the phosphorylated form of eIF2α …

The fidelity of DNA replication is achieved in a multiplicative process encompassing nucleobase selection and insertion, removal of misinserted nucleotides by exonuclease activity, and enzyme dissociation from primer/templates that are misaligned due to mispairing. In this study, we have evaluated the effect of altering these kinetic processes on the dynamics of translesion DNA replication using the bacteriophage T4 replication apparatus as a model system. The effect of enhancing the processivity of the T4 DNA polymerase, gp43, on translesion DNA replication was evaluated using a defined in vitro assay system. Small angle neutron scattering (SANS) experiments were used to characterize binary nanodroplets composed of D2O and H2O. The droplets were formed by expanding dilute mixtures of condensible vapor in a N2 carrier gas through a supersonic nozzle, while maintaining the onset of condensation at a fixed position in the nozzle. It is remarkable, given the small coherent scattering length density of light water, that even the pure H2O aerosol gave a scattering signal above background. The scattering spectra were analyzed assuming a log-normal distribution of droplets. On average, the geometric radius of the nanodroplets rg was rg=13 (±1) nm, the …

The paper presents the theoretical framework for the development of a new solvent boundary method for computer simulations and its possible application to the simulation of nitric oxide synthase. Different approaches for the construction of a solvent boundary potential are reviewed and their strengths and weaknesses are discussed. A new solvent boundary potential is proposed which combines the mean field force approximation with the Green's function approach for treating long-range electrostatic interactions, and introduces a novel strategy to treat electrostriction effects due to ions crossing the solvent boundary. The scattering far zone for light transmitted through a sphere following p - 1 internal reflections by a family of near-grazing incident rays is subdivided into a lit region and a shadow region. The sharpness of the ray theory transition between the lit and the shadow regions is smoothed in wave theory by radiation shed by electromagnetic surface waves. It is shown that when higher-order terms in the physical optics approximation to the phase of the partial-wave scattering amplitudes are included, the transition between the lit and the shadow regions becomes a two-ray-to-zero-ray transition, called a superweak caustic in analogy …

A reversed-phase HPLC assay has been developed to determine the concentration of the anti-metabolite 2′,2′-difluorodeoxycytidine (gemcitabine, dFdC) in human plasma over the concentration range of 0.5–150 μM (0.13–39.44 μg/ml), and 2′,2′-difluorodeoxyuridine (dFdU), the deaminated, inactive metabolite, over the range of 1.0–227 μM (0.26–60 μg/ml). After the addition of 20 nmol 2′-fluorodeoxycytidine (FdC) as an internal standard, 0.5-ml samples of plasma were subjected to acetonitrile precipitation, followed by analysis using a gradient reversed-phase HPLC assay with UV detection. Transient kinetic analyses further support the role of the clamp-loader in bacteriophage T4 as a catalyst which loads the clamp onto DNA through the sequential hydrolysis of two molecules of ATP before and after addition of DNA. Additional rapid-quench and pulse-chase experiments have documented this stoichiometry. The events of ATP hydrolysis have been related to the opening/closing of the clamp protein through fluorescence resonance energy transfer (FRET). In the absence of a hydrolysable form of ATP, the distance across the subunit interface of the clamp does not increase as measured by intramolecular FRET, suggesting gp45 cannot be loaded onto DNA. …

Detection Sensitivity Optimization Of Optical Signals Generated By Fiber Optic Bragg Gratings Under Dynamic Excitation, John Lekki, James A. Lock

The dynamic response of a fiber optic Bragg grating to mechanical vibrations is examined both theoretically and experimentally. The theoretical expressions describing the consequences of changes in the grating's reflection spectrum are derived for partially coherent beams in an interferometer. The analysis is given in terms of the dominant wavelength, optical bandwidth, and optical path difference of the interfering signals. Changes in the reflection spectrum caused by a periodic stretching and compression of the grating are experimentally measured using an unbalanced Michelson interferometer, a Michelson interferometer with a nonzero optical path difference. Anomers of monovalent and divalent β-d-galactopyranosyl-(1→4)-2-acetamido-2-deoxy-d-gluco-pyranosides were synthesized under different glycosylation conditions, and evaluated for in vitro antimetastatic activity. Three compounds showed promising inhibitory effects on cancer cell attachment, spreading, migration, and invasion. Six divalent O-β-d-galactopyranosyl-(1→4)-2-acetamido-2-deoxy-d-glucopyranosides (29–34) were synthesized and their antimetastatic activities were studied.

We analyze the optical caustic produced by light refracted at the curved meniscus surrounding a cylindrical rod standing partially out of a liquid-filled container. When the rod is tilted from the vertical or when light is diagonally incident, the caustic is a four-cusped astroid with two of its cusps obscured by the rod's shadow. If a portion of the flat end of the rod is raised above the water level, the caustic evolves into a pattern of five interlocking cusps. The five cusps result from symmetry breaking of a three-cusped surface perturbation caustic. (C) 2003 Optical Society of America.

A new class of rainbows is created when a droplet is illuminated from the inside by a point light source. The position of the rainbow depends on both the index of refraction of the droplet and the position of the light source, and the rainbow vanishes when the point source is too close to the center of the droplet. Here we experimentally measure the position of the transmission and one-internal-reflection total-internal-reflection rainbows, and the standard (primary) rainbow, as a function of light-source position. Coronas are simulated in color by use of the Mie scattering theory of light by small droplets through clouds of finite optical thickness embedded in a Rayleigh scattering atmosphere. The primary factors that affect color, visibility, and number of rings of coronas are droplet size, width of the size distribution, and cloud optical thickness. The color sequence of coronas and iridescence varies when the droplet radius is smaller than similar to6-mum. As radius increases to approximately 3.5 mum, new color bands appear at the center of the corona and fade as they move outward. As the radius continues to increase …

A reversed-phase high-performance liquid chromatography method with electrospray ionization and mass spectral detection is described for the determination of capecitabine, 5′-deoxy-5-fluorocytidine and 5′-deoxy-5-fluorouridine in human plasma with 5-chloro-2′-deoxyuridine as the internal standard. An on-line sample clean-up procedure allows dilution of the plasma sample with the initial mobile phase. The linear dynamic range is 0.0500–10.0 μg/ml for capecitabine, and 0.0500–25.0 μg/ml for the metabolites, 5′-deoxy-5-fluorocytidine and 5′-deoxy-5-fluorouridine, respectively. This method has been used to analyze plasma samples from patients receiving capecitabine in combination with oxaliplatin.

A series of seven focus groups were conducted in various locations throughout northern Ohio to identify and assess coastal resources management training needs across the Ohio Great Lakes basin. The focus groups were comprised of a cross-section of professionals who make decisions affecting watershed areas or Lake Erie coastal areas. Six of the focus groups included decision-makers considered previous and potential users of coastal resources management training (non-providers). A seventh focus group included decision-makers who provide training in coastal resources management (providers).

A computational model has been developed to investigate how the magnetostatic interactions affect the hysteresis and magnetization curves for hexagonal arrays of magnetic nanowires. The magnetization coupling between nanowires arises from the stray fields produced by the other nanowires composing the array such that the field at each nanowire is the sum of the external field and the interaction field with the other nanowires. Co0.45Fe0.55 alloy nanowires with 12 to 35 nm diameter and 12 μm length were fabricated by electrodeposition in porous anodic alumina templates. The initial magnetization curves reveal that the zero magnetization state is not unique and is determined by the field history (acdemagnetization process) leading to the zero average moment state. For acdemagnetization processes with the field applied parallel to the nanowire axis, the subsequent magnetization curves suggest that an individual nanowire behaves as a single domain with neighboring nanowires being antiparallel to each other in the zero magnetization state. Purpose: Since preclinical studies have shown more than additive cytotoxicity and DNA damage with the combination of gemcitabine and 5-fluoro-2′-deoxyuridine (FUDR), we studied this combination in a phase I trial. Methods: Gemcitabine alone was given in cycle 1 as a 24-h, 2-h or 1-h i.v. infusion weekly for 3 of 4 weeks; if tolerated, a 24-h i.v. infusion of FUDR was added with oral leucovorin. The Old Woman Creek National Estuarine Research Reserve (OWC), Ohio Department of Natural Resources-Coastal Management Program (ODNRCMP), and Ohio Sea Grant College Program engaged the Great Lakes Environmental Finance Center (GLEFC) to aid them in developing a coastal resources management training initiative for Ohio coastal decision-makers. Coastal decision-maker professionals are expected to formulate effective strategies and apply interdisciplinary approaches to solving problems and policy issues that affect the environments of coastal regions. Coastal decision-makers and training providers in the Ohio Lake Erie basin participated in seven focus groups to generate information on coastal resources knowledge and training needs. Six of the focus groups were comprised of a cross-section of professionals who make decisions affecting watershed areas or Lake Erie coastal areas (non-providers); the seventh was comprised of training providers (providers). Discussions centered on two main areas: core knowledge needs and training needs.

We study a family of diffusion models for risk reserves which account for the investment income earned and for the inflation experienced on claim amounts. After we defined the process of the conditional probability of ruin over finite time and imposed the appropriate boundary conditions, classical results from the theory of diffusion processes turn the stochastic differential equation to a special class of initial and boundary value problems defined by a linear diffusion equation.
