Simulations For Designing And Interpreting Intervention Trials In Infectious Diseases., M Elizabeth Halloran, Kari Auranen, Sarah Baird, Nicole E Basta, Steven E Bellan, +Several Additional Authors

Global Health Faculty Publications

BACKGROUND: Interventions in infectious diseases can have both direct effects on individuals who receive the intervention as well as indirect effects in the population. In addition, intervention combinations can have complex interactions at the population level, which are often difficult to adequately assess with standard study designs and analytical methods.

DISCUSSION: Herein, we urge the adoption of a new paradigm for the design and interpretation of intervention trials in infectious diseases, particularly with regard to emerging infectious diseases, one that more accurately reflects the dynamics of the transmission process. In an increasingly complex world, simulations can explicitly represent transmission dynamics, …

Recent studies have found an association between functional variants in TREM2 and PLD3 and Alzheimer's disease (AD), but their effect on cognitive function is unknown. We examined the effect of these variants on cognitive function in 1449 participants from the Wisconsin Registry for Alzheimer's Prevention, a longitudinal study of initially asymptomatic adults, aged 36–73 years at baseline, enriched for a parental history of AD. A comprehensive cognitive test battery was performed at up to 5 visits. A factor analysis resulted in 6 cognitive factors that were standardized into z scores (∼N [0, 1]); the mean of these z scores was …

Purpose

Early cardiac toxicity is a risk associated with adjuvant chemotherapy plus trastuzumab. However, objective measures of cardiac function and health-related quality of life are lacking in long-term follow-up of patients who remain cancer free after completion of adjuvant treatment.

Patients and Methods

Patients in NSABP Protocol B-31 received anthracycline and taxane chemotherapy with or without trastuzumab for adjuvant treatment of node-positive, human epidermal growth factor receptor 2–positive early-stage breast cancer. A long-term follow-up assessment was undertaken for patients who were alive and disease free, which included measurement of left ventricular ejection fraction by multigated acquisition scan along with patient-reported …

Some Dimension Reduction Strategies For The Analysis Of Survey Data, Jiaying Weng, Derek S. Young Dec 2017

Some Dimension Reduction Strategies For The Analysis Of Survey Data, Jiaying Weng, Derek S. Young

Statistics Faculty Publications

In the era of big data, researchers interested in developing statistical models are challenged with how to achieve parsimony. Usually, some sort of dimension reduction strategy is employed. Classic strategies are often in the form of traditional inference procedures, such as hypothesis testing; however, the increase in computing capabilities has led to the development of more sophisticated methods. In particular, sufficient dimension reduction has emerged as an area of broad and current interest. While these types of dimension reduction strategies have been employed for numerous data problems, they are scantly discussed in the context of analyzing survey data. This …

Bayesian Prediction Intervals For Assessing P-Value Variability In Prospective Replication Studies, Olga A. Vsevolozhskaya, Gabriel Ruiz, Dmitri Zaykin Dec 2017

Bayesian Prediction Intervals For Assessing P-Value Variability In Prospective Replication Studies, Olga A. Vsevolozhskaya, Gabriel Ruiz, Dmitri Zaykin

Biostatistics Faculty Publications

Increased availability of data and accessibility of computational tools in recent years have created an unprecedented upsurge of scientific studies driven by statistical analysis. Limitations inherent to statistics impose constraints on the reliability of conclusions drawn from data, so misuse of statistical methods is a growing concern. Hypothesis and significance testing, and the accompanying P-values are being scrutinized as representing the most widely applied and abused practices. One line of critique is that P-values are inherently unfit to fulfill their ostensible role as measures of credibility for scientific hypotheses. It has also been suggested that while P-values …

Using Mixed Effects Modeling To Quantify Difference Between Patient Groups With Diabetic Foot Ulcers, Rachel French Dec 2017

Using Mixed Effects Modeling To Quantify Difference Between Patient Groups With Diabetic Foot Ulcers, Rachel French

Mahurin Honors College Capstone Experience/Thesis Projects

When diabetes progresses, many patients suffer from chronic foot ulcers. In a study described in Matrix Metalloproteinases and Diabetic Foot Ulcers (Muller et al., 2008), sixteen patients with diabetic foot ulcers were examined throughout a twelve week healing period. During this period, levels of matrix metalloproteinases (MMP-1), their inhibitors (TIMP-1), and the extracellular matrix in a wound area were measured at distinct time intervals for each patient. The ratios of these healing components are vital in determining whether a wound will heal or become chronic and never properly heal. Connecting Local and Global Sensitivities in a Mathematical Model for Wound …

Flow Anisotropy Due To Thread-Like Nanoparticle Agglomerations In Dilute Ferrofluids, Alexander Cali, Wah-Keat Lee, A. David Trubatch, Philip Yecko

Department of Applied Mathematics and Statistics Faculty Scholarship and Creative Works

Improved knowledge of the magnetic field dependent flow properties of nanoparticle-based magnetic fluids is critical to the design of biomedical applications, including drug delivery and cell sorting. To probe the rheology of ferrofluid on a sub-millimeter scale, we examine the paths of 550 μm diameter glass spheres falling due to gravity in dilute ferrofluid, imposing a uniform magnetic field at an angle with respect to the vertical. Visualization of the spheres’ trajectories is achieved using high resolution X-ray phase-contrast imaging, allowing measurement of a terminal velocity while simultaneously revealing the formation of an array of long thread-like accumulations of magnetic …

A Customized Quantitative Pcr Microrna Panel Provides A Technically Robust Context For Studying Neurodegenerative Disease Biomarkers And Indicates A High Correlation Between Cerebrospinal Fluid And Choroid Plexus Microrna Expression, Wang-Xia Wang, David W. Fardo, Gregory A. Jicha, Peter T. Nelson

Sanders-Brown Center on Aging Faculty Publications

MicroRNA (miRNA) expression varies in association with different tissue types and in diseases. Having been found in body fluids including blood and cerebrospinal fluid (CSF), miRNAs constitute potential biomarkers. CSF miRNAs have been proposed as biomarkers for neurodegenerative diseases; however, there is a lack of consensus about the best candidate miRNA biomarkers and there has been variability in results from different research centers, perhaps due to technical factors. Here, we sought to optimize technical parameters for CSF miRNA studies. We examined different RNA isolation methods and performed miRNA expression profiling with TaqMan® miRNA Arrays. More specifically, we developed a customized …

Dynamic Child Growth Prediction: A Comparative Methods Approach, Andrada Ivanescu, Ciprian M. Crainiceanu, William Checkley Dec 2017

Dynamic Child Growth Prediction: A Comparative Methods Approach, Andrada Ivanescu, Ciprian M. Crainiceanu, William Checkley

Department of Applied Mathematics and Statistics Faculty Scholarship and Creative Works

We introduce a class of dynamic regression models designed to predict the future of growth curves based on their historical dynamics. This class of models incorporates both baseline and time-dependent covariates, start with simple regression models and build up to dynamic function-on-function regressions. We compare the performance of the dynamic prediction models in a variety of signal-to-noise scenarios and provide practical solutions for model selection. We conclude that (a) prediction performance increases substantially when using the entire growth history relative to using only the last and first observation; (b) smoothing incorporated using functional regression approaches increases prediction performance; and (c) …

Load Model Verification, Validation And Calibration Framework By Statistical Analysis On Field Data, Xiangqing Jiao, Yuan Liao, Thai Nguyen

Electrical and Computer Engineering Faculty Publications

Accurate load models are critical for power system analysis and operation. A large amount of research work has been done on load modeling. Most of the existing research focuses on developing load models, while little has been done on developing formal load model verification and validation (V&V) methodologies or procedures. Most of the existing load model validation is based on qualitative rather than quantitative analysis. In addition, not all aspects of model V&V problem have been addressed by the existing approaches. To complement the existing methods, this paper proposes a novel load model verification and validation framework that can systematically …

Family caregivers have the potential to help patients manage those symptoms, as well as their own stress, if they are equipped with the proper resources. Electronic health (eHealth) systems may be able to provide those resources. Very sick patients may not be able to use such systems themselves to report their symptoms but family caregivers could.

Objective: The aim of this paper was to assess the effects on cancer patient symptom distress of an eHealth system that alerts clinicians to significant changes in the patient’s symptoms, as …

Trend And Acceleration: A Multi-Model Approach To Key West Sea Level Rise, John Tenenholtz Nov 2017

Trend And Acceleration: A Multi-Model Approach To Key West Sea Level Rise, John Tenenholtz

FIU Electronic Theses and Dissertations

Sea level rise (SLR) varies depending on location. It is therefore important to local residents, businesses and government to analyze SLR locally. Further, because of increasing ice melt and other effects of climate change, rates of SLR may change. It is therefore also important to evaluate rates of change of SLR, which we call sea level acceleration (SLA) or deceleration.

The present thesis will review the annual average sea level data compiled at the Key West tidal gauge in Key West, Florida. We use a multi-model approach that compares the results of various models on that data set. The goal …

Convergence Analysis And Error Estimates For A Second Order Accurate Finite Element Method For The Cahn–Hilliard–Navier–Stokes System, Amanda E. Diegel, Cheng Wang, Xiaoming Wang, Steven M. Wise

Mathematics and Statistics Faculty Research & Creative Works

In this paper, we present a novel second order in time mixed finite element scheme for the Cahn–Hilliard–Navier–Stokes equations with matched densities. the scheme combines a standard second order Crank–Nicolson method for the Navier–Stokes equations and a modification to the Crank–Nicolson method for the Cahn–Hilliard equation. in particular, a second order Adams-Bashforth extrapolation and a trapezoidal rule are included to help preserve the energy stability natural to the Cahn–Hilliard equation. We show that our scheme is unconditionally energy stable with respect to a modification of the continuous free energy of the PDE system. Specifically, the discrete phase variable is shown …

Entropy Production In A Fluid-Solid System Far From Thermodynamic Equilibrium, Bong Jae Chung, Blas Ortega, Ashuwin Vaidya

Department of Applied Mathematics and Statistics Faculty Scholarship and Creative Works

Abstract.: The terminal orientation of a rigid body in a moving fluid is an example of a dissipative system, out of thermodynamic equilibrium and therefore a perfect testing ground for the validity of the maximum entropy production principle (MaxEP). Thus far, dynamical equations alone have been employed in studying the equilibrium states in fluid-solid interactions, but these are far too complex and become analytically intractable when inertial effects come into play. At that stage, our only recourse is to rely on numerical techniques which can be computationally expensive. In our past work, we have shown that the MaxEP is a …

Kallmes, Carlos M. Jimenez, Christopher Putman, Juan Cebral

Department of Applied Mathematics and Statistics Faculty Scholarship and Creative Works

BACKGROUND AND PURPOSE: Intracranial aneurysms originating at the posterior communicating artery are known to have high rupture risk compared with other locations. We tested the hypothesis that different angioarchitectures (ie, branch point configuration) of posterior communicating artery aneurysms are associated with aneurysm hemodynamics, which in turn predisposes aneurysms to rupture.

MATERIALS AND METHODS: A total of 313 posterior communicating artery aneurysms (145 ruptured, 168 unruptured) were studied with image-based computational fluid dynamics. Aneurysms were classified into different angioarchitecture types depending on the location of the aneurysm with respect to parent artery bifurcation. Hemodynamic characteristics were compared between ruptured and unruptured …

Using Multilevel Outcomes To Construct And Select Biomarker Combinations For Single-Level Prediction, Allison Meisner, Chirag R. Parikh, Kathleen F. Kerr

UW Biostatistics Working Paper Series

Biomarker studies may involve a multilevel outcome, such as no, mild, or severe disease. There is often interest in predicting one particular level of the outcome due to its clinical significance. The standard approach to constructing biomarker combinations in this context involves dichotomizing the outcome and using a binary logistic regression model. We assessed whether information can be usefully gained from instead using more sophisticated regression methods. Furthermore, it is often necessary to select among several candidate biomarker combinations. One strategy involves selecting a combination on the basis of its ability to predict the outcome level of interest. We propose …

Constructing A Confidence Interval For The Fraction Who Benefit From Treatment, Using Randomized Trial Data, Emily J. Huang, Ethan X. Fang, Daniel F. Hanley, Michael Rosenblum Oct 2017

Constructing A Confidence Interval For The Fraction Who Benefit From Treatment, Using Randomized Trial Data, Emily J. Huang, Ethan X. Fang, Daniel F. Hanley, Michael Rosenblum

Johns Hopkins University, Dept. of Biostatistics Working Papers

The fraction who benefit from treatment is the proportion of patients whose potential outcome under treatment is better than that under control. Inference on this parameter is challenging since it is only partially identifiable, even in our context of a randomized trial. We propose a new method for constructing a confidence interval for the fraction, when the outcome is ordinal or binary. Our confidence interval procedure is pointwise consistent. It does not require any assumptions about the joint distribution of the potential outcomes, although it has the flexibility to incorporate various user-defined assumptions. Unlike existing confidence interval methods for partially …

Dodrant-Homomorphic Encryption For Cloud Databases Using Table Lookup, Thomas Schwarz Oct 2017

Dodrant-Homomorphic Encryption For Cloud Databases Using Table Lookup, Thomas Schwarz

Mathematics, Statistics and Computer Science Faculty Research and Publications

Users of large commercial databases increasingly want to outsource their database operations to a cloud service providers, but guaranteeing the privacy of data in an outsourced database has become the major obstacle to this move. Encrypting all data solves the privacy issue, but makes many operations on the data impossible in the cloud, unless the service provider has the capacity to decrypt data temporarily. Homomorphic encryption would solve this issue, but despite great and on-going progress, it is still far from being operationally feasible. In 2015, we presented what we now call dodrant-homomorphic encryption, a method that encrypts numeric values …
