Cyclodextrins (CD) are a family of oligosaccharides that can non-covalently and precisely bind small hydrophobic drug molecules, such as antibiotics and anticancer drugs, through well-defined host-guest interactions inside the inner cavity of the macrocycle. Ring-opening metathesis polymerization (ROMP) is a versatile, functional group tolerant method of polymerization that is underexplored in biomedical applications relative to the more common controlled chain-growth polymerizations. In this dissertation, I describe my efforts to combine the supramolecular chemistry of CDs with that of ROMP using specialized functional monomers to synthesize several CD-based polymeric drug delivery nanomaterials. Electrochemistry provides an alternative pathway to handle redox organic reactions, especially for certain chemical process that can be only achieved through electrochemical process. Herein, there research projects are introduced to enrich the whole synthetic community, including an anodic oxidative cyclization studies to develop methodology on heterocyclic ring skeleton constructions, an electroreduction pathway exploration to more effectively obtain hydrazine from nitroso compounds, as well as a study in paired electrolysis process on which both anode and cathode are simultaneously employed to maximize overall atom and energy efficiency. Future directions on these projects are also demonstrated at the end of each chapters..

Enantioselective Synthesis Of Β-Amino Acid Derivatives Using Amidine-Based And Bifunctional Organocatalysts, Matthew Robert Straub

Two new enantioselective methodologies have been developed that have important implications for the asymmetric synthesis of β-amino acids and their derivatives. First, chiral amidine-based catalyst (ABC) HBTM-2 catalyzed an asymmetric cyclocondensation between in situ activated fluoroacetic acid and N-sulfonyl aldimines to give α-fluoro-β-lactams in highly enantioenriched form, achieving modest to excellent diastereoselectivities. These reactive lactams can then be quenched with various alcohols and amines to deliver the α-fluoro-β-amino acid derivatives in moderate isolated yields. Secondly, bifunctional double hydrogen bond donor-amine organocatalysts enable the catalytic alcoholysis of various racemic N-carbalkoxy-3-substituted isoxazolidin-5-ones, resulting in their kinetic resolution. Interest in the development of soft-material artificial molecular muscles has inspired scientists to pursue novel stimuli-responsive systems capable of undergoing change in the physical and mechanical properties of a material in response to external stimuli. This interest has been driven primarily by advances, or the desire for advances, in fields such as soft robotics, microfluidics, and bio-compatible drug-delivery systems. In this work, redox-responsive viologens and their ability to form stable radical-cation complexes were evaluated as a method of actuation in stimuli-responsive materials, specifically hydrogels.

Two new cascade transformations of α,β-unsaturated thioesters catalyzed by amidine-based catalysts have been developed. First, a reagent-free transformation of o-formylaryl cinnamoyl thioesters catalyzed by HBTM-2 to produce 2-substituted thiochromenes achieved high enantioselectivities and yields while forming carbon dioxide as the only byproduct. Second, a highly diastereo- and enantioselective tandem rearrangement of less reactive enone thioesters into tricyclic thiochromanes in the presence of electron-rich amidine-based catalysts was developed. These catalysts were designed with the help of DFT calculations. H-PIP, the first chiral amidine-based catalyst synthesized in our group, performed the best overall in the thiochromane synthesis.

Multidrug resistant (MDR) Gram-negative bacteria represent a global health crisis. Traditional antibiotics create selective pressure and breed resistance, making our current drugs less and less effective. Targeting of bacterial virulence factors has gained interest as an alternative strategy to potentially circumvent this problem. Virulent Acinetobacter baumannii synthesize and secrete pre-acinetobactin, a small molecule metal chelator that scavenges life-sustaining iron and is critical for establishing infection. The work described herein sheds light on the unusual properties of a non-enzymatic isomerization from pre-acinetobactin to acinetobactin and proposes a siderophore-swapping mechanism by which A. baumannii expands its pH window for virulence. The development of palladium catalysis has been influential in a wide range of organic transformations, in particular C-C coupling, C-Heteroatom coupling and C-H functionalization reactions. These catalytic transformations normally proceed through the Pd0/II catalytic cycle. These reactions are remarkably useful, however, they suffer from two major problems: -hydride elimination and palladium black deposition. To circumvent these problems, recent reports have been focused on developing novel organic transformations proceeding through high-valent palladium and nickel catalytic cycles.
