There are approximately 48,000 known cave systems in the United States of America, with caves formed in carbonate karst terrains being the most common. Epigenic systems develop from the downward flow of meteoric water through carbonate bedrock and the solutional enlargement of interconnected subsurface conduits. Despite carbonate karst aquifers being globally extensive and important drinking water sources, microbial diversity and function are poorly understood compared to other Earth environments. Beach closures and public health protection are confounded by analytical procedures that result in delays in notification of adverse water quality conditions and the lack of affordable analytical methods to identify pollutant sources. Attempts have been made to develop predictive frameworks using ancillary hydrometeorological data to statistically anticipate deteriorated water quality. Many urban coastal beaches are impacted by river runoff. In Kenosha Wisconsin, beach sanitary survey data from two beaches adjacent to the mouth of the Pike River were examined to ascertain whether simple river-lake mixing models identified river influence on coastal water quality and improved predictions of beach advisories. …

Sewage contamination from failing infrastructure and sewer overflows is a major environmental and human health concern in waterways, especially in urban communities bordering the Great Lakes such as Milwaukee, Wisconsin. Culture-based fecal indicator bacteria, such as Escherichia coli, enterococci, and fecal coliforms are traditionally used to indicate the presence of a human health risk due to fecal contamination. These indicators, however, fail to distinguish between sources of fecal contamination (human vs. non-human). Microalgae can be grown on municipal wastewater media to both treat the wastewater and produce feedstock for algae biofuel production. However the reliability of treatment must be demonstrated, as well as high areal algae productivity on recycled wastewater media and efficient sedimentation harvesting. This processes was studied at pilot scale in the present research.

Rare Occurrences Of Free-Living Bacteria Belonging To Sedimenticola From Subtidal Seagrass Beds Associated With The Lucinid Clam, Stewartia Floridana, Aaron M. Goemann

Lucinid clams and their sulfur-oxidizing endosymbionts comprise two compartments of a three-stage, biogeochemical relationship among the clams, seagrasses, and microbial communities in marine sediments. A population of the lucinid clam, Stewartia floridana, was sampled from a subtidal seagrass bed at Bokeelia Island Seaport in Florida to test the hypotheses: (1) S. floridana, like other lucinids, are more abundant in seagrass beds than bare sediments; (2) S. floridana gill microbiomes are dominated by one bacterial operational taxonomic unit (OTU) at a sequence similarity threshold level of 97% (a common cutoff for species level taxonomy) from 16S rRNA genes; …

Sanitation, renewable energy, and food security are among the most pressing global development needs of the century, especially for small cities with rapid population growth. Currently, 53% of the world’s population either lacks access to improved sanitation or discharges fecal waste to the environment without treatment. Furthermore, 80% of food consumed in developing regions is produced by 500 million small farms, and while many of them are still rain-fed, irrigated agriculture is increasing. Limestone dissolution in karst environments is likely due to geochemistry of the water, the actions of microbial communities, and the effect of water flow. We explored the rate of limestone dissolution and will examine here the microbial communities associated with the limestone. A conduit within the brackish cave, Double Keyhole Spring, on the coast of central west Florida was the site of the experiment. PVC pipes (5cm x 16cm) were filled with crushed limestone that was screened to a 1.9cm – 2.54cm size range. This research models and analyzes the inactivation of Bacillus spores following a radiation exposure and the process enacted by the Bacillus spore to repair the resulting damage. Irradiation of a spore and the medium surrounding the spore induces chemical reactions that produce reactive oxygen species (ROS). This research will consider the reaction- diffusion of these ROS throughout the spore. These ROS can react with the spore's DNA and enzymes to degrade them to such an extent that the DNA cannot be repaired or replicated, thus causing spore death. This thesis began by verifying that nanoscale zero valent iron (nZVI) synthesis methods could be scaled up and implemented at the field scale in a safe manner. This led to successful demonstration of nZVI injection and mobility under constant head gravity injection into a contaminated utility corridor in Sarnia, Ontario. Where field studies have fallen short in the past was linking the somewhat qualitative field geochemical parameters to other evidence of nZVI transport. The several million years preceding the Cretaceous/Paleogene (K/Pg) boundary has been the focus of many studies. Changes in ocean circulation and sea level, extinctions, and major volcanic events have all been documented for this interval. Important research questions these changes raise include the climate dynamics during the warm, but not hot, time after the decay of the Late Cretaceous greenhouse interval and the stability of ecosystems prior to the mass extinctions at the end-Cretaceous.

Investigations into the distribution and ecosystem functions of fruiting amoebae revealed that local-scale environmental conditions can largely explain broad biogeographical patterns in species assemblage, the way in which amoeboid predators shape bacterial communities and how this top-down influence may affect global biogeochemical processes in a changing climate. The distribution and assemblage of protosteloid amoebae on the islands of New Zealand and Hawaii did not yield any expected patterns of island biogeography, and conformed to other global regions studied. Due to the production of methane (CH4) under flooded-soil conditions, rice (Oryza sativa L.) cultivation is a major contributor to agricultural CH4 emissions. Studies examining CH4 emissions from rice have only recently been initiated in Arkansas and no data have been collected from rice produced on clay soils in Arkansas. The scale of predator-prey interactions varies from the iconic (e.g., lions and gazelles) to the microscopic (e.g., protists and bacteria). Bdellovibrio-and-like-organisms (BALOs) are bacteria that prey upon other Gram-negative bacteria, including pathogens. In particular, the halophilic genus Vibrio, many species of which are disease agents, has been shown to be susceptible to attack by BALOs. One motivation of the present research is to assess the potential for biocontrol of Vibrio by Bacteriovorax. The household biosand filter (BSF) is one of the world’s most utilized point-of-use (POU) water treatment tools where drinking water is not reliably potable. The feasibility of using ferrocement for construction, the filter volume, and the effect of temperature on removal are unclear, however. The following field and laboratory research was carried out in Panama and at Michigan Technological University. Field testing of ferrocement biosand filters (FBSFs) in rural Panama yielded average coliform and E.coli removals that compared favorably to plastic and cement BSFs. The majority of dengue cases in the United States were acquired in endemic areas by travelers or immigrants. "Soap Lake, located in Washington State, is a meromictic, soda lake. Many bacterial isolates retrieved from Soap Lake have been noted to possess resistance to multiple antibiotics. A likely explanation for the wide range of antibacterial resistance exhibited by these strains is due to the impact of high alkalinity on the antibiotics themselves and not due to the presence of antibiotic resistance genes. Despite our dependency on treatment facilities to condition wastewater for eventual release to the environment, our knowledge regarding the effects of treated water on the local watershed is extremely limited. Responses of these lotic systems to the treated wastewater effluent have been traditionally investigated by examining the benthic macroinvertebrate assemblages and community structure; however, these studies do not address the microbial diversity of the water systems.
