Alfalfa (Medicago sativa) is an autotetraploid, allogamous and heterozygous species. Cultivated varieties are synthetic cultivars, usually obtained through 3 or 4 generations of panmictic reproduction of a set of various numbers of parents. The parents can be clones, half-sib or full-sib families. The breeders apply selection pressure for some agronomic traits, to induce changes in the genetic background. The objective of this study was to investigate the differentiation level among seven cultivars originating from one breeding program, and between these cultivars and the breeding pool, with eight SSR markers.

  1. Molecular markers are effective tools to support traditional approaches in plant genetic resources management.

Lolium perenne, as one of the most important forage grasses of temperate regions, combines a number of very useful characteristics, e.g., good seedling establishment, with a low resistance to drought and limited winter hardiness. Trait selection and introgression can be greatly enhanced by the use of molecular markers in a genetic linkage map. The aim of this project was the generation of a genomic microsatellite library which when combined with microsatellites developed from a Genethresher database would give good genome coverage coupled to high levels of marker polymorphism.

Two small Indian mongooses (Herpestes auropunctatus) were live-captured in 2012 at separate locations on the Hawaiian Island of Kaua'i, which was previously considered to be free of this invasive species. Genotypes from these two individuals were compared to genotypes of H. auropunctatus from the islands of Hawai'i (n =39), O'ahu (n =91), Maui (n = 39), and Moloka'i (n = 19) to determine the island of origin of the Kaua'i individuals. The oceanic whitetip shark, Carcharhinus longimanus, is a circumtropical pelagic shark of high conservation concern (IUCN Red List: “Critically Endangered” in the Western North and Western Central Atlantic and “Vulnerable” globally). I present the first, population genetic assessment of the oceanic whitetip shark on a global scale, based on analysis of two mitochondrial genome regions (entire 1066-1067 bp control region and 784 bp partial ND4 gene), and nine nuclear microsatellite loci. No population structure was detected within the Western Atlantic. Isolated in scattered remnants, less than 0.1% of Iowa’s original tallgrass prairie remains. The small populations remaining are at risk for reduced genetic diversity, inbreeding depression, and outbreeding depression. In light of these concerns, we used microsatellite analysis to assess the genetic structure of butterfly milkweed (Asclepias tuberosa) populations on prairie remnants in northwest Iowa. We compared remnant populations with a restoration population at Dordt College in Sioux Center, Iowa, and with an Oklahoma seed source. The genus Arctomecon (Papaveraceae) is comprised of three narrowly endemic rare species that are largely restricted to gypsum soils of the eastern Mojave Desert. The small, remaining populations of these species have become increasingly isolated by urban development and habitat fragmentation. Arctomecon humilis is federally listed as endangered due to its limited distribution within a ~15 km radius of an actively expanding city. Organizations involved with land management and conservation have called for greater insight into the genetic variation and population structure of the remaining subpopulations as they make important decisions regarding where to focus their efforts and resources.

Fragmentation over evolutionary time scales following vicariant and dispersal events has long been recognised as a dominant process in biological diversification and speciation; while anthropogenic habitat fragmentation in recent times is considered a threat to the long-term persistence of species and ecosystems. The eastern freshwater cod Maccullochella ikei is Australia’s largest endangered freshwater fish species. Abundant in the Clarence and Richmond River systems at the time of European settlement, populations crashed in the early 1900s causing local extinctions and range reduction. DNA from non-invasive sources is increasingly being used as molecular tags for markrecapture population estimation. These sources, however, provide small quantities of often contaminated DNA, which can lead to genotyping errors that will bias the population estimate. We describe a novel approach, called Genotyping Uncertainty Added Variance Adjustment (GUAVA), to address this problem. GUAVA incorporates an explicit model of genotyping error to generate a distribution of complete-information capture histories that is used to estimate the population size. This approach both reduces the genotyping-error bias and incorporates the additional uncertainty due to genotyping error into the variance of the estimate. We …
