Towards The Total Synthesis Of Thioviridamide: Thiyl Radical Approach To The Beta-Thioenamide Linkage Formation, Jung-Hoon Kang

We developed an approach to the β-thioenamide linkage contained in the S-(2-aminovinyl)cysteine (avCys) residue of thioviridamide.1,2 Kinetic and thermodynamic control of radical additions of thiols to ynamides were studied for the formation of β-thioenamide linkage. Thiyl radicals are electrophilic and ynamides are electron-rich alkynes. This complementary polarity of the radical and acceptor increases the likelihood of a successful radical addition reaction. Because little is known about these types of compounds (β-thioenamides), we were unsure what kinds of yields and stereoselectivities (cis vs. trans) to expect. In 1988 data on coral reef community composition were collected from two areas, Akumal and Chemuyil, Quintana Roo, Mexico, ranging from 5-35m depth. These areas were revisited in 2005 and data were collected by the same methods and at the same depths as in 1988. Data from 1988 and 2005 were compared to determine if the coral reefs had undergone significant changes, and what specific changes had occurred. Chi-square analysis determined that community composition data collected in 1988 are significantly different from data collected in 2005 at all sites and depths within the categories of corals, gorgonians, sponges, and macroalgae. …

The identification of alternative protein sources for partial or whole replacement of fishmeal (FM) is a priority in the development of suitable, low cost feed formulations for the Florida pompano (Trachinotus carolinus). To evaluate alternative feed ingredients for the potential replacement of FM in diets fed to juvenile pompano, a series of four experiments were conducted. For the past ten years the Fleming research group has been involved in the research of allene compounds. We have explored the synthesis of allenes as well as several reactions involving allenes, such as oxidation and nitrene addition. Recently, we have explored the synthesis and photochemistry of allenic alcohols tethered to silicon. There are literature examples using allenes in synthesis, but very few examples exist using allenes in photochemical cycloaddition reactions. We have found that a diisopropylallenyloxy(cinnamyloxy)silane derivative undergoes [2+2] photochemical cycloaddition to produce a cyclobutylsilane product when irradiated for 60-75 minutes.

Coastal lagoons and bays vary in shape and size in response to antecedent topography, geologic processes and sea level rise. Variations in shape and environmental conditions of coastal basins are believed to influence the distribution of benthic sub-environments and the exchange of water with the ocean and other adjacent coastal systems. Gargathy Inlet and its coastal lagoon vary spatially from the inlet, where the greatest depths are observed, to the mainland, dominated by shallow intertidal areas, colonized by marsh. Purpose: This in vitro study compared the radiopacity between human primary and permanent enamel and dentin. It also compared the radiopacity between human anterior and posterior teeth.

Methods: Extracted primary and permanent teeth were used in this study. One mm enamel and one mm dentin slices were cut from each extracted tooth. Radiographs were taken of each enamel and dentin specimen 1 mm in thickness along with an aluminum step-wedge using procedures specified in International Standards Organization (ISO) 4049:2000. Adiabatic calorimetry provides accurate and precise specific heat (Cp) data. From this data, thermodynamic functions may be calculated. Cu2OCl2, melanothallite, became of interest as part of a study of a particular thermochemical cycle. The experimental specific heat data and the calculated thermodynamic functions are reported here. Free energies of formation, calculated from the thermodynamic functions, suggest the particular cycle of interest with this compound as an intermediate is not feasible; uncertainty as to the accuracy of CuO and CuCl2 data used in the calculations indicate further study may be necessary. The synthesis of five chiral DBFOX (dibenzofuran-oxazoline) ligands with either aryl or benzyl substituents will be presented. The requisite amino alcohols were obtained with high enantioselectivity either commercially (DBFOX/Bn), via Sharpless asymmetric aminohydroxylation (DBFOX/Nap, DBFOX/t-BuPh, DBFOX/Pip), or via phase-transfer catalyzed asymmetric alkylation (DBFOX/MeNap). These ligands, complexed with Mg(NTf2)2, were used as Lewis acid promoters of enantioselective radical conjugate additions to α/β-unsaturated nitro-amides/esters. Chemokines are important chemical signals that guide lymphocyte movement within the immune system and promote the organization and functions of germinal centers (GCs) in the secondary lymphoid tissues. Previous studies have shown that GC T cells exhibit high expression of chemokine receptor 4, CXCR4, but that these cells are unable to migrate to the ligand for this receptor, the chemokine CXCL12. This “migratory paralysis” to CXCL12 was found to be correlated with the expression of two Regulators of G-protein Signaling, RGS13 and RGS16 in the GC T cells. Widespread fish stocking has led to a worldwide decline in naturally fishless lakes and their associated communities. Little is known about the historical distribution or native communities of these freshwater ecosystems. The objectives of this study were to: 1) develop a quantitative method to remotely detect naturally fishless lakes in Maine, 2) conduct a landscape-scale assessment of unique attributes of fishless lake macroinvertebrate communities, 3) identify macroinvertebrate bioindicators of fish absence, and 4) assess effects of introduced fish on native macroinvertebrates. Raman spectroscopy, laser-induced fluorescence, and sum-frequency generation (SFG) spectroscopy are used to investigate the behavior of compressible mobile phases and stationary phases under a variety of chromatographic conditions. Efforts to understand and optimize separations employing compressible mobile phases have been limited by a lack of understanding of the mobile phase density gradient. Mobile phase compressibility leads to gradients in linear velocity and solute retention and affects separation speed and efficiency, especially in packed columns. The Dania Beach Erojacks Artificial Reef was deployed off the coast of Broward County, Florida on December 31, 1967 as a way to help combat beach erosion. Over the last forty years, the linear pile of concrete hexapods has become an important habitat, for scleractinian corals, octocorals, algae, crustaceans, mollusks, and fish. Ribosomes translate the genetic information encoded by mRNA into proteins. Defective mRNAs can cause stalling of translating ribosomes. The molecule tmRNA (transfer-messenger RNA) rescues stalled ribosomes in eubacteria. Together with its protein partner SmpB, tmRNA mimics a tRNA by entering the ribosomal A site and linking an alanine residue to the growing polypeptide chain. The ribosome then abandons the defective mRNA template and resumes translation on tmRNA, adding ten more amino acids to the nascent polypeptide. Molecular dynamics simulation was used to study ion permeation through different membrane proteins embedded in a lipid bilayer (DMPC) with different saline solutions. The potential of mean force (PMF) for ion transport was obtained by umbrella sampling simulations. A revised MacKerell force field for tryptophan residues was studied using gramicidin A (gA) channel as a test model. The revised force field contribution to the Na+ PMF was consonant with the prediction from the experimental results, but in stark contrast to the prediction of the CHARMM force field, version 22, for the tryptophan side-chain. Pygmy sperm whales (Kogia breviceps) and dwarf sperm whales (Kogia sima) strand frequently in the southeastern United States (SEUS). To detect seasonal trends in Kogia sp. strandings across the SEUS, all 979 stranding events from 1977 through 2005 were segregated by month. A peak in strandings occurred in the late summer and early fall (July – October). PURPOSE: To evaluate sealant microleakage using four different application techniques.

METHODS: The four application techniques evaluated were acid etch only, Adper L-Pop bonding, Optibond bonding, and Enamel Loc One-Step Sealant. Sixty-four extracted third molar teeth were assigned on one of four application techniques (n=16) and Clinpro sealants applied according to the manufacturer's instructions. The teeth were thermocycled for 500 cycles, dyed, and embedded in resin. The samples were then sectioned and evaluated for microleakage on the buccal and lingual surfaces. Statistical methods employed to analyze the data were the Kruskal-Willis rank test and the Mann-Whitney U test.

Categorization of animals and vehicles in different contexts was investigated in three experiments using event related potentials (ERPs). The presence of a background and congruency of the background in relation to the object were both manipulated in order to determine the effects of context on visual processing. The [beta]-oxidation system in sunflower (Helianthus annuus L.) cotyledons is distinguished by the existence of two different thiolase isoforms, Thiolase I (acetoacetyl-CoA thiolase, EC 2.3.1.9 AACT) and Thiolase II (3-oxoacyl-CoA thiolase, EC 2.3.1.16 OACT). Glyoxysomal AACT is the last enzyme in the p-oxidation of fatty acids in plant glyoxysomes. Glyoxysomal AACT has been successfully cloned, expressed and purified from sunflower cotyledons (Dyer et al., 2006). In this paper we investigate the optimal conditions for both expression and purification. These include the choice of vector, the growth conditions and the purification parameters for the maximum possible yield of AACT protein.

The vast overuse of antibiotics along with extrachromosomal gene insertion between different bacterial species has led to the Darwinian consequence of antibiotic resistance. Amongst the contributing factors to the rise of bacterial resistances are the production of ß-lactamases, which pose a major impediment to treatment with ß-lactam drugs such as penicillins and carbapenems. SPM-1 is a metallo-ß-lactamase (MBL) that confers antibiotic resistance to bacteria through zinc-mediated hydrolysis of ß-lactams. Bacteria that express the SPM-1 metalloenzyme show reduced sensitivity to ß-lactam drugs. The thesis takes as input information that has been generated from videotaping walking individuals and converting their gaits into numerical data that measures the locations of various points on the body through time. Soils disturbed by strip mining practices may have increased colloid loads moving to groundwater resources, also enhancing the transport of contaminants into our water resources. We hypothesize that contaminant transport within soils following mining is enhanced by colloid mobility. Two sites were chosen for this study, a 30-year old reclaimed strip mine in southwest Virginia and a recently mined area from eastern Kentucky. Intact reclaimed soil monoliths were retrieved from sandstone derived soils in southwestern Virginia. Reclaimed monoliths from eastern Kentucky were recreated in the lab. Amphibian populations are declining worldwide due to factors such as habitat degradation, fragmentation and destruction. I conducted a study to explore the use of created ponds in a forested habitat by breeding amphibians, specifically Rana sylvatica and Ambystoma maculatum. The objectives were to examine the movement of these animals after leaving the ponds, the survival and movement of juveniles, how both respond to fragmentation, and how similar the created ponds were to natural ones. The behaviors of oyster larvae are difficult to monitor or experimentally manipulate, especially in field conditions. As a result, little is known of the fate of oysters in the larval portion of their life cycle, prior to recruitment. At the transition from pelagic larvae to benthic adults, larvae are likely to come into contact with many invertebrates resident on oyster reefs. Of these, fouling epifauna are generally believed to reduce the settlement of interspecific larvae through competitive exclusion and predation.
