Beta-N-methylamino-L-alanine (BMAA) is a non-protein amino acid produced by many cyanobacteria, and thought to induce neurotoxic effects through excitotoxicity, contributing to neurodegenerative diseases such as Amyotrophic Lateral Sclerosis/Parkinsonism-dementia complex (ALS-PDC) and Alzheimer’s. The ubiquitous nature of cyanobacteria, and evidence of biomagnification through our food web, creates a dire need for the development of an analytical platform that will provide accurate identification and quantification of BMAA amounts in our ecosystem and potential food supply. Coastal forest retreat in the Florida Keys during the 20th century has been attributed to a combination of sea level rise and hurricane storm surge impacts, but the interactions between these two disturbances leading to forest decline are not well understood. The goal of my research was to assess their effects over a period spanning more than two decades, and to examine the relationships between these press and pulse disturbances and freshwater availability in pine rockland, hardwood hammock, and supratidal scrub communities. Amines in the atmosphere are of interest because of their likely role in new particle formation, and because of anthropogenic emissions of amines at post-combustion carbon capture (PCCC) facilities. A conceptual framework for considering the partitioning of a monobasic amine (Am = unprotonated "free-base form) from the gas phase to atmospheric particulate matter (PM) is presented for cases when the PM may be composed of multiple liquid phases. Natural and anthropogenic emissions of aromatic hydrocarbons from biomass burning, agro-industrial settings, and fossil fuel combustion contribute precursors to secondary aerosol formation (SOA). How these compounds are processed under humid tropospheric conditions is the focus of current attention to understand their environmental fate. This work shows how catechol thin films, a model for oxygenated aromatic hydrocarbons present in biomass burning and combustion aerosols, undergo heterogeneous oxidation at the air–solid interface under variable relative humidity (RH = 0–90%). Managed honey bee, Apis mellifera L., colonies are contracted to pollinate fruit and nut orchards improving crop quality and yield. Colonies placed in orchards are potentially exposed to pyrethroid insecticides used for broad-spectrum pest control. Pyrethroids have been reported to pose minimal risk to bees due to their low application rates in the field and putative repellent properties. This repellency is believed to alter foraging behavior with the benefit of preventing bees from encountering a lethal dose in the field. However, sub-lethal exposure to pyrethroids may adversely impact bee behavior potentially resulting in social dysfunction or disruption of foraging.

Several potential new phosphorus-containing flame retardant molecules were evaluated for heat release reduction potential by incorporation of the molecules into a polyurethane, generated from methylene diphenyl diisocyanate and 1,3-propane diol. The heat release reduction potential of these substances was evaluated using the pyrolysis combustion flow calorimeter (PCFC). The polyurethanes were prepared in the presence of the potential flame retardants via solvent mixing and copolymerization methods to qualitatively evaluate their potential reactivity into the polyurethane prior to heat release testing. The functionality of the flame retardants was epoxide based that would potentially react with the diol during polyurethane synthesis. Flammability testing …

Mangos (Mangifera indica L.) are tropical/subtropical fruits belonging to the plant family Anacardiaceae. Anthracnose is the most deleterious disease of mango both in the field and during postharvest handling. It is most commonly caused by the Colletotrichum gloeosporioides complex. Mangiferin, a xanthanoid compound found in at least twelve plant families worldwide (Luo et al., 2012), is present in large amounts of the leaves and edible mangos. Even though this compound plays a pivotal role in the plant’s defense against biotic and abiotic stressors, no correlations been made between the compound and mango anthracnose resistance.

Development and application of target and non-target techniques for routine analysis, identification of transformation products, and characterization of unknown compounds in water matrices using liquid-chromatography high-resolution mass spectrometry (HRMS) were explored in this dissertation. A novel analytical method based on online-SPE-LC-HRMS was developed for the detection of 18 drugs of abuse (DOAs) in raw sewage water from a college campus. Results showed the presence of 14 DOAs for which amphetamine and 11-nor-9-carcoxy-THC (THC metabolite) were the most prevalent and had the highest potential consumption rates.

Black carbon (BC) is a complex mixture of polycondensed aromatic compounds produced by the incomplete combustion of biomass during events such as wildfires and the burning of fossil fuels. Black carbon was initially considered to be a refractory form of organic matter. However, recent studies have shown that BC can be quite mobile and reactive in the terrestrial environment. Black carbon can be translocated from soils and sediments in the form of dissolved BC (DBC). The intertidal zone is an ideal habitat to investigate effects of global warming because species living in it are very close to their physiological limits. Initial studies of invertebrate physiological responses to heat stress have employed relatively abrupt increases in temperature. My research investigates effects of ecologically more realistic temperature profiles on feeding in the intertidal hermit crab, Pagurus sameulis. Recent work in the Wright lab showed that feeding in this species is inhibited by an abrupt increase in temperature. Because temperature change in the natural environment of Pagurus is much more gradual, I hypothesize that such a gradual temperature …

At the Irvine Ranch Water District, two types of secondary treatment are utilized: a sludge and Dual Media Filters (DMF) process and Membrane Bioreactor (MBR) process. This study was proposed to analyze the electrical conductivity (EC) of each secondary effluent and possible reasons for differences. After testing the two effluents using an electrical conductivity probe and ion chromatography, it was discovered that there is no significant difference between the two, though secondary treatment raises the nitrate levels in general.

The Toxic Substances Control Act (TSCA) is the central form of chemical regulation existent in the United States today, yet scientists are often unaware or uncertain of its provisions. Violations of TSCA by unknowing chemists set industry and government unnecessarily at odds. A lecture on TSCA was developed for undergraduate students that uses the concept of green chemistry to promote interest and incentivize learning. The occurrences of cyanotoxin and organic contaminants threaten drinking water sources and are a serious human health and environmental concern. The control of these problematic contaminants and the remediation of the associated contaminants are critical for ensuring safe drinking water to significant populations. Advanced oxidation processes (AOPs) have received considerable attention as a potential water treatment for various pollutants. In this dissertation, advanced oxidative degradation of four problematic water toxic contaminants (CYN, iopamidol, 4-methylcyclohexane methanol and propylene glycol phenyl ether) were studied to develop the fundamental understanding required to assess AOPs as a potential water treatment process.

The purpose of this research is to investigate the molecular constitution of silk and the specific causes of post-production degradation in silk material. This capstone project focused specifically on the conditions and degradation of ancient Chinese silk products provided by the Chinese National Silk Museum. Modern silk products were artificially degraded and compared to the ancient silk using Py-GC-MS and comparative analytic techniques. This investigation revealed that the ancient silk products most likely underwent degradation in elevated temperatures while in the presence of moisture and the absence of sunlight or UV radiation during long term burial.

Invasive plant species are major threats to the biodiversity and ecosystem stability. The purpose of this study is to understand the impacts of invasive plants on soil nutrient cycling and ecological functions. Soil samples were collected from rhizosphere and non-rhizosphere of both native and exotic plants from three genera, Lantana, Ficus and Schinus, at Tree Tops Park in South Florida, USA. Experimental results showed that the cultivable bacterial population in the soil under Brazilian pepper (invasive Schinus) was approximately ten times greater than all other plants. The introduction of particulate and oxides of nitrogen (NOx) after-treatment controls on heavy-duty vehicles has spurred the need for fleet emissions data to monitor their reliability and effectiveness. The University of Denver has developed a new method for rapidly measuring heavy-duty vehicles for gaseous and particulate fuel specific emissions. The method was recently used to collect 3,088 measurements at a Port of Los Angeles location and a weigh station on I-5 in northern California. The Viking missions to Mars, which took place in the 1970’s, performed several experiments on martian soil in an attempt to discern if there was microbial life present. In one set of experiments, a nutrient solution containing amino acids and organic substrates tagged with carbon-14 was injected into sample cells containing martian soil, and the evolution of labeled CO2 was monitored. The evolution of labeled CO2 indicated that there was a process occurring that broke down the nutrients in solution and released CO2 as a byproduct. Secondary organic aerosol particles (SOA) formed from the oxidation of monoterpenes can impact the Earth’s radiation balance, act as cloud condensation nuclei and negatively affect human health. In the initial Visiting Faculty Program application, we proposed the use of an ultraviolet-visible spectrometer equipped with a liquid waveguide capillary flow cell to determine the extent to which SOA absorb visible light. The inclusion of Concordia University in the Secondary Organic Aerosol From Forest Emissions Experiment (SOAFFEE) laboratory campaign at PNNL necessitated a change in the proposed experiments. While the magnitude of radiation absorbed by greenhouse gases is known to a high certainty, the absorption of radiation by atmospheric aerosol particles is not. The micro-orifice uniform deposit impactor– droplet freezing technique (MOUDI-DFT) combines particle collection by inertial impaction (via the MOUDI) and a microscope-based immersion freezing apparatus (the DFT) to measure atmospheric concentrations of ice nucleating particles (INPs) as a function of size and temperature. In the first part of this study we improved upon this recently introduced technique. Using optical microscopy, we investigated the non-uniformity of MOUDI aerosol deposits at spatial resolutions of 1, 0.25 mm, and for some stages when necessary 0.10 mm. The inner shelf of the East China Sea is a river-dominated margin characterized by fine-grained mud deposits and a rapid sedimentation rate. Three short sediment cores (similar to 2.7m in length) were examined to characterize spatial variations in magnetic mineral diagenesis. The sediment cores were analyzed for sedimentation rates, magnetic properties, particle size distribution, organic carbon, and total sulfur content. The two more proximal cores with higher sedimentation rates (similar to 2.2cm/yr and similar to 0.96cm/yr) do not exhibit obvious effects of reductive dissolution of magnetite with increasing depth, which is consistent with their lower total sulfur content. Over the past half century, analytical pyrolysis has proven itself to be an effective means for the semiquantitative characterization of complex macromolecular organic substances. It has been demonstrated that instruments such as Py-FID, Py-MS, and in particular, Py-GC/MS can provide valuable geochemical insights when applied to a wide variety of problems in environmental science. The more widespread use of analytical pyrolysis methods in the evaluation of environmental pollution is recommended, because of their relatively low cost and information-rich results.

An abandoned chemical plant in Asturias (Spain) was studied using a multi-faceted molecular fingerprinting approach, demonstrating that it is possible to: (1) carefully unravel tangled evidence resulting from multiple pollution sources, and (2) recognize major contaminants largely ignored by conventional analyses. This methodology employed a battery of GC-MS analyses of liquid chromatographic fractions of soil extracts, plus the pyrolysis products of the soil extract's asphaltene fraction and the whole soil itself. In this example, coal tar distillation and the subsequent production of naphthalene, phenols and polymer resins are responsible for most of the soil contamination. We describe the publicly available data from the Global OZone Chemistry And Related trace gas Data records for the Stratosphere (GOZCARDS) project and provide some results, with a focus on hydrogen chloride (HCl), water vapor (H2O), and ozone (O3). This data set is a global long-term stratospheric Earth system data record, consisting of monthly zonal mean time series starting as early as 1979. The data records are based on high-quality measurements from several NASA satellite instruments and the Atmospheric Chemistry Experiment Fourier Transform Spectrometer (ACE-FTS) on SCISAT. We examine consistency aspects between the various data sets. …

Fungal spores as a prominent type of primary biological aerosol particles (PBAP) have been incorporated into the COSMO-ART (Consortium for Small-scale Modelling- Aerosols and Reactive Trace gases) regional atmospheric model. Two literature-based emission rates for fungal spores derived from fungal spore colony counts and chemical tracer measurements were used as a parameterization baseline for this study. A third, new emission parameterization for fluorescent biological aerosol particles (FBAP) was adapted to field measurements from four locations across Europe. FBAP concentrations can be regarded as a lower estimate of total PBAP concentrations. Size distributions of FBAP often show a distinct mode at …
