Aptamer-Based Voltammetric Biosensing For The Detection Of Codeine And Fentanyl In Sweat And Saliva, Rosa Lashantez Cromartie Nov 2021

Aptamer-Based Voltammetric Biosensing For The Detection Of Codeine And Fentanyl In Sweat And Saliva, Rosa Lashantez Cromartie

FIU Electronic Theses and Dissertations

Despite the many governmental and medicinal restrictions created to combat the opioid epidemic in the United States, opioid abuse and overdose rates continue to rise. The development of an aptamer-based voltammetric sensor and biosensor is described in this dissertation. The aim was to develop a low-cost, sensitive, and specific aptamer-based sensor for on-site, label-free determination of codeine and fentanyl in biological fluids. To do this, the surfaces of screen-printed carbon electrodes (SPCE) were modified with gold nanoparticles (AuNPs), followed by the addition of single-stranded DNA aptamers. These were covalently bound to the electrode surface. Operations of the sensors were collected …

An Electrochemical Immunoassay System For Measuring Circulating Protein Biomarkers Of Pediatric Soft Tissue Sarcoma, Ivy Antwi Aug 2021

An Electrochemical Immunoassay System For Measuring Circulating Protein Biomarkers Of Pediatric Soft Tissue Sarcoma, Ivy Antwi

Electronic Theses and Dissertations

Measurement of circulating protein biomarkers associated with disease can facilitate early detection, help guide treatment strategies and improve patient outcomes beyond current standards of care. The combination of inexpensive 3D-printed flow cells and electrochemical biosensors has recently emerged as a viable platform for low-cost, reliable biomarker measurements. Here, we report an electrochemical immunoassay system based on simple graphite electrode arrays, 3D-printed flow cells, and signal-generating magnetic bead bioconjugates for simultaneous detection of three biomarker proteins (cancer antigen 125 (CA-125), midkine (MK) and osteopontin (OPN)) associated with pediatric soft tissue sarcomas (PSTS). Magnetic bead bioconjugates are functionalized with large amounts of …

Two Dimensional Nanoparticles/Nanozymes For Biosensing Augmented By Machine Learning, Nidhi Chintan Nandu Aug 2021

Two Dimensional Nanoparticles/Nanozymes For Biosensing Augmented By Machine Learning, Nidhi Chintan Nandu

Legacy Theses & Dissertations (2009 - 2024)

Biosensing is an ever-evolving field with many resources devoted towards new approaches in sensing and refining the existing methods. For a long time, such approaches involved use of state-of-the-art instruments and were often proved to be time consuming and expensive. Nanoparticles with their unique properties, inexpensive synthesis and ease of manipulation have found purpose in myriad fields including biosensing. Different nanoparticles based on their composition, morphology, dimensionality, and surface modifications have already gained foot hold in the biosensor ranks. The unique opto-electric properties due to the quantum effect in the nanoparticles have also made them one of the leading alternatives …

Novel Sensing Concept For Organophosphate Monitoring, Cheyenne Bowman Jan 2021

Novel Sensing Concept For Organophosphate Monitoring, Cheyenne Bowman

Legacy Theses & Dissertations (2009 - 2024)

Organophosphates are a group of insecticides that are commonly used as chemical warfare agents (CWAs), since they can act as nerve agents by inhibiting acetylcholinesterase. Acetylcholinesterase is a vital enzyme of the central nervous system (CNS). This enzyme facilitates the breakdown of acetylthiocholine into thiocholine and acetate. The major goal of the research will be to provide a preliminary rapid, sensitive, and reliable method for sensing organophosphates. Current methods are enzyme inhibition-based – that is, a decrease in absorbance or signal yields a positive indicator for the presence of analyte. Herein, two different novel approaches to organophosphate sensing are described. The …
