PolC is the polymerase responsible for genome duplication in many Gram-positive bacteria and represents an attractive target for antibacterial development. We have determined the 2.4-Å resolution crystal structure of Geobacillus kaustophilus PolC in a ternary complex with DNA and dGTP. The structure reveals nascent base pair interactions that lead to highly accurate nucleotide incorporation. A unique β-strand motif in the PolC thumb domain contacts the minor groove, allowing replication errors to be sensed up to 8 nt upstream of the active site. PolC exhibits the potential for large-scale conformational flexibility, which could encompass the catalytic residues. The structure suggests a …

A promising avenue in the development of high-energy pulsed chemical HF/DF lasers and amplifiers is the utilization of a photon-branched chain reaction initiated in a two-phase active medium, that is, a medium containing a laser working gas and ultradispersed passivated metal particles. These particles are evaporated under the action of IR laser radiation which results in the appearance of free atoms, their diffusion into the gas, and the development of a photon-branching chain process, which involves photons as both reactants and products. X-ray absorption spectroscopy (XAS) has emerged as one of the premier tools for investigating the structure and dynamic properties of metals in cells and in metal containing biomolecules. Utilizing the high flux and broad energy range of X-rays supplied by synchrotron light sources, one can selectively excite core electronic transitions in each metal. Spectroscopic signals from these electronic transitions can be used to dissect the chemical architecture of metals in cells, in cellular components and in biomolecules at varying degrees of structural resolution. We developed an approach to the β-thioenamide linkage contained in the S-(2-aminovinyl)cysteine (avCys) residue of thioviridamide.1,2 Kinetic and thermodynamic control of radical additions of thiols to ynamides were studied for the formation of β-thioenamide linkage. Thiyl radicals are electrophilic and ynamides are electron-rich alkynes. This complementary polarity of the radical and acceptor increases the likelihood of a successful radical addition reaction. Because little is known about these types of compounds (β-thioenamides), we were unsure what kinds of yields and stereoselectivities (cis vs. trans) to expect. Nonlinear optical (NLO) and electro-optical (EO) properties of π-conjugated systems have been the subject of intense interest during the past several decades. In this mini-review we focus on semiconducting materials based on alkyne π-conjugation, with particular emphasis on those examples from our laboratory of chromophores containing a silole core. Resonance Raman (RR) studies of intermediates generated by cryoreduction of the oxyferrous complex of the D251N mutant of cytochrome P450cam (CYP101) are reported. Owing to the fact that proton delivery to the active site is hindered in this mutant, the unprotonated peroxo-ferric intermediate is observed as the primary species after radiolytic reduction of the oxy-complex in frozen solutions at 77 K. In as much as previous EPR and ENDOR studies have shown that annealing of this species to ∼180 K results in protonation of the distal oxygen atom to form the hydroperoxo intermediate, this system has been exploited to …

The photochemical evolution of an anthropogenic plume from the New-York/Boston region during its transport at low altitudes over the North Atlantic to the European west coast has been studied using a Lagrangian framework. This plume, originally strongly polluted, was sampled by research aircraft just off the North American east coast on 3 successive days, and then 3 days downwind off the west coast of Ireland where another aircraft re-sampled a weakly polluted plume. Changes in trace gas concentrations during transport are reproduced using a photochemical trajectory model including deposition and mixing effects. One of the signature activities during this time continues to be the intensive extraction, processing, and combustion of fossil fuels. While fossil fuels themselves are naturally-occurring, they are most often millions of years old and associated with deeply buried strata. A simple and economical fluorometer using blue LEDs excitation sources and simple PMT detection had been built, assembled, optimized, and employed for measurement of fluorescence from riboflavin (vitamin B2). Surfactants have been known to enhance the intensity of fluorescence of fluorescent compounds. Fluorescence analysis of riboflavin in the presence of various anionic, cationic, and nonionic surfactants was also conducted to determine if they could improve analysis. However, the surfactants employed did not seem to have any meaningful enhancement; in fact, some actually diminished the fluorescence intensity of riboflavin. Progress toward the syntheses of new tetranuclear bimetallic complexes of copper(II) and ruthenium(II) was realized. The designed triads comprise a central binuclear copper(II) complex with a tetraiminodiphenolate macrocyclic Robson-type compartmental ligand. In the envisioned complexes, the macrocyclic core is further functionalized by attachment of two polypyridyl ruthenium(II) complexes. A novel dibrominated dicopper(II) Robson complex was formed by the 2:2:2 condensation reaction of 4-bromo-2,6-diformylphenol and 1,3- diaminopropane with cupric chloride. Similarly, a new dibrominated dizinc(II) was synthesized from zinc tetrafluoroborate and the same diamine and dialdehyde. The new dicopper(II) complex did not heterocouple with borylated substrates under explored Suzuki reaction conditions. …

Terahertz (THz) time domain spectroscopy (TDS) was assessed as a nondestructive evaluation technique for aircraft composites. Material properties of glass fiber composite were measured using both transmission and reflection configuration. The interaction of THz with a glass fiber composite was then analyzed, including the effects of scattering, absorption, and the index of refraction, as well as effective medium approximations. THz TDS, in both transmission and reflection configuration, was used to study composite damage, including voids, delaminations, mechanical damage, and heat damage. Lipid A is a glucosamine-based phospholipid that anchors carbohydrate molecules to the outer membrane of most Gram-negative bacteria. During severe infection, this amphiphilic macromolecule invoke a strong and acute pro-inflammatory response that leads to sepsis and ultimately to septic shock, organ failure and death. Resonance Raman spectroscopy is employed to characterize heme site structural changes arising from conformational heterogeneity in deoxyMb and ligated derivatives, i.e., the ferrous CO (MbCO) and ferric cyanide (MbCN) complexes. The spectra for the reversed forms of these derivatives have been extracted from the spectra of reconstituted samples. Dramatic changes in the low-frequency spectra are observed, where newly observed RR modes of the reversed forms are assigned using protohemes that are selectively deuterated at the four methyl groups or at the four methine carbons. We report the design of an amphiphilic polyamine (polymer 1) based on poly(2-alkenyl azlactone) that strongly couples the formation of polyelectrolyte complexes at aqueous/liquid crystal (LC) interfaces to ordering transitions in the LC. We demonstrate that the addition of a strong anionic polyelectrolyte to aqueous solutions in contact with polymer 1-laden LC interfaces (prepared by Langmuir-Schaefer transfer of monolayers of polymer 1 onto micrometerthick films of nematic LC) triggers ordering transitions in the LCs. We further demonstrate that changes in the ordering of the LCs (i) are driven by electrostatic interactions between the polyelectrolytes, (ii) involve multivalent interactions between the …

Compared to conventional acrylic monomer systems, thiol-ene photopolymerization, an efficient click process leading to the formation of dense and uniform molecular networks, has several distinct advantages including relative insensitivity to oxygen inhibition, high monomer conversion and low shrinkage. This work is divided into two parts: the adsorption of propane on the magnesium oxide (100) surface and the synthesis of anodized aluminum oxide. The adsorption properties of propane on the MgO (100) surface were investigated using high-resolution volumetric isotherm techniques and a computational study was accomplished using Materials Studio. From the adsorption work, the two-dimensional isothermal compressibility, the isosteric heat of adsorption, the differential enthalpy, and the differential entropy of adsorption can be calculated. The thermodynamics of molecular hydrogen adsorbed on MgO (100) surfaces has been examined using high resolution volumetric adsorption isotherm techniques. These studies were undertaken using highly perfect, narrow size distributed MgO nanocubes with essentially single (100) facet exposure and extremely high chemical purity. A narrowly spaced (in temperature) series of isotherms were performed between the temperatures of ~7K and the triple point (13.8 K) using H2 and D2 gas.

A minimum of five, and in some cases seven, discrete adsorption steps are clearly observed in each isotherm trace. Analysis of these data indicate that the monolayer film forms …

This dissertation summarizes the recent use of boron and transition metal halides to form new carbon-carbon bonds. Several novel reactions have been discovered. These include the haloallylation of aryl aldehydes with boron trihalide; alkyne-aldehyde couplings mediated by boron trihalide to give either (Z,Z)-1,5-dihaloalkenes or (Z,E)-1,5- dihaloalkenes; dehydroxylation of benzylic alcohols and in situ generated benzylic alcohol salts with iron(III) chloride and triethylsilane to afford diarylmethanes; and, the alkenylation of allylic, benzylic, and propargylic alcohols with phenylacetylenes and titanium(IV) halide. This article describes a method for fabricating 3D microfluidic devices by stacking layers of patterned paper and double-sided adhesive tape. Paper-based 3D microfluidic devices have capabilities in microfluidics that are difficult to achieve using conventional open-channel microsystems made from glass or polymers. In particular, 3D paper-based devices wick fluids and distribute microliter volumes of samples from single inlet points into arrays of detection zones (with numbers up to thousands). Photopolymerization of reverse microemulsion demonstrates strong influences from dynamical and chemical factors on morphology of formed polymer systems. Upon polymerization the clear microemulsion with nanoscale structures transforms into opaque films with larger aggregates that scatter light. Studied here were microemulsions based on acrylate and on thiol-ene chemistries. The difference in the two reaction mechanisms results in the phase separation occurring at different stages of the material formation. Consequently, the morphology of the material demonstrates distinct dissimilarities in the two systems. In addition, the chemical structure of the monomers promotes these morphological differences.
