Recent pharmaceutical developments have investigated using supramolecular nanoparticles in order to increase the bioavailability and solubility of drugs delivered in various methods. Modification of the carbohydrate cyclodextrin increases the ability to encapsulate hydrophobic pharmaceutical molecules by forming a carrier with a hydrophobic core and hydrophilic exterior. Guest molecules are commonly added to these inclusion complexes in order to add stability and further increase targeting abilities of the carriers. One such guest molecule is adamantine combined with a poly(ethylene glycol) chain. Carbohydrates are important to cellular communication, recognition, and pathogenesis, making them a useful synthetic target for application in therapeutics and diagnosis. Synthetic carbohydrate presenting scaffolds can replicate or interrupt the binding interactions that occur in nature. Due to the weak nature of the monosaccharide-protein bond, most carbohydrate presenting scaffolds display saccharides in a multivalent manner to improve binding. Recently there has been an effort to combine carbohydrate containing scaffolds with polymersomes (vesicles composed of polymers), due to the structural resemblance of polymersome membranes to biological membranes. Single molecule studies can provide information of biological molecules which otherwise is lost in ensemble studies. A wide variety of fluorescence-based techniques are utilized for single molecule studies. While these tools have been widely applied for imaging soluble proteins, single molecule studies of transmembrane proteins are much more complicated. A primary reason for this is that, unlike membrane proteins, soluble proteins can be easily isolated from the cellular environment. One approach to isolate membrane proteins into single molecule level involves a very low label expression of the protein in cells.
