Siloles and 9-heterofluorenes (Si, Ge) are conjugated π-electron systems containing Group 14 atoms which demonstrate unique properties such as high electron affinity, mobility, and bathochromically shifted optical spectra relative to their all-carbon analogs. This is due to their low-lying lowest-unoccupied molecular orbital (LUMO) which is resultant of the σ*-π* conjugation between the exocyclic groups at the Group 14 center and the butadiene unit of the ring. High-resolution biomacromolecular structure elucidation is fundamentally important to structure-based drug design and basic research into complex biochemical processes. Cryo-EM is an emerging alternative to XRD and NMR that is complementary in many ways relative to XRD and NMR. Materials approaches to cryo-EM are anticipated to greatly facilitate the cryo-EM process, allowing progress toward a more high-throughput application of cryo-EM to address challenges in structural biology. ^ Various affinity-based approaches inspired by approaches previously introduced for 2D crystallization were developed for facilitation of cryo-EM. Part 1. Lead compound SK-03-92 represents a new scaffold for antibiotic drug discovery. Development of a new process for the synthesis of analogs has led to the development of a number of new ligands with even more potent activity against gram-positive bacteria, including drug-resistant strains of S. aureus. Compounds 36 and 38 represent some of the most potent analogs developed thus far, and preliminary results indicate that they are also not cytotoxic. Lipids control a variety of complex biological processes. Bulk lipids such as phosphatidylcholine (PC), phosphatidylserine (PS), and phosphatidylethanolamine (PE) represent the major components of cellular membranes. In addition, unilamellar vesicles composed of lipids (liposomes) are valuable for delivery applications since they can encapsulate and transport drugs and other agents. In order to maximize delivery efficiency and target specific membranes, the ability to trigger and control vesicle-vesicle fusion is desirable. Quinonoids are quinone derivatives that have carbonyl or carbonyl equivalent and even number of double bonds embedded in six member rings. As a result of the intrinsic α,β-unsaturated ketone or imine structures, quinonoids, such as quinone monoketals, quinols, quinol ethers and quinone imine ketals, can accommodate a wide range of reactions including 1,2-additon, 1,4-addtion, SN2’ reaction (allylic substitution) to the α-carbon of the carbonyl or imine and cycloaddition reactions (e.g. Diels-Alder reaction). The Mukaiyama oxidative cyclization has been a major area of study within the Pagenkopf group and within the field of THF containing natural products. This document contains a brief discussion of the major divisions of THF synthesis; SN2, cycloadditions, and transition mediated strategies. The development of the Mukaiyama cyclization is described from the earliest applications of cobalt acac type ligand in Mukaiyama’s work to the initial discovery of the cyclization reactivity. Glycosyl transfer within the active site of an enzyme involves the arrangement of the gly-cosyl donor and glycosyl acceptor in close proximity to each other. To mimic this pro-cess, intramolecular glycosylations are carried out where the donor and acceptor are linked together via a spacer. Correct orientation, rigidity and position of the spacer play a vital role in the selectivity and yield of reactions in this “molecular clamp method”. Metastable layered perovskites containing interlayer transition metals can readily be obtained by simple ion exchange reactions on receptive hosts, such as those of the Dion-Jacobson and Ruddlesden-Popper structure types. In this work, we focused on adding to the library of layered perovskites by not only creating new compounds, but by also showing their ability to be further manipulated, and by studying the stability of the series through thermal behavior studies.

Various propargyl alcohols were synthesized via Grignard reactions. These alkynic alcohols are tethered with dimethyl and biphenyl silicon tethers. These terminal alkyne substrates are reacted with RuHCl(CO)(H2IMes)(PPh3) as a catalyst under high pressure (80 psi) ethylene atmosphere to afford the 5-exo-dig trans-silylvinylation product in poor yield. Internal alkyne substrates are found to be much more efficiently transformed into the silylvinylation product, favoring one cycloisomer product over the other.

Metal-organic nanotubes (MONTs) are an emerging class of discrete materials that are the 1D variant of metal-organic frameworks (MOFs). MONTs have potential to become an alternative 1D material to carbon nanotubes, metal oxide nanotubes, and boron nitride nanotubes because they possess an organic ligand that can be functionalized and tuned for specific applications. Despite this potential, only a handful of structures have been reported and only two examples of discrete MONTs exist in the literature. It is thus imperative to develop general methods to prepare and characterize discrete MONTs to bring them to the forefront of the scientific literature.

This dissertation focuses on the synthesis of dauer pheromone analogs for biological evaluation, the development of visible light promoted O-glycosylation methods and the development of an Ir(ppy)3 catalyzed C-H hydroxylation method. Chapter 1 describes the synthesis of dauer pheromone analogs. C.elegans is a small nematode that enters a dauer stage when encountering unfavorable environmental conditions. The dauer stage is initiated by the nematodes chemosensation of the dauer pheromone caused by the down regulation biochemical pathways TGF-â and IGF-1. This work is focused on the design, synthesis and characterization of block copolymers (BCPs) containing both conjugated and polyelectrolyte blocks. This type of rod-coil BCP is considered to have microphase separation which is different compared to coil-coil BCPs. The conjugated block used for studies is regioregular poly(3-alkylthiophene) (P3AT), which can be synthesized by Grignard metathesis (GRIM) polymerization. The polyelectrolyte block used is polystyrene sulfonate with well-defined structure, which can be synthesized by atom transfer radical polymerization (ATRP). “Supramolecular chemistry” powered by non-covalent interactive forces forms the crux in the area of host-guest chemistry. Supramolecular assemblies often have different chemical and physical properties than that of its individual molecular entities and are used to develop novel functional materials. Our expertise involves making functional materials from macrocycles, which contain two urea groups and two rigid C shaped spacer groups. These individual macrocyclic components can self-assemble through hydrogen bonding and other non-covalent interactions to form porous supramolecular assemblies that can be used as confined reaction environments and as ligands to synthesize novel metal organic materials.

Exploratory crystal growth is a versatile technique and one that can advance materials discovery. Through exploratory crystal growth, it is possible to synthesize many new materials that can have applications in many fields, including solid-state lighting. There exist many adaptations for exploratory crystal growth, including the use of alkali halide flux growth and hydroflux growth, both reported herein. By changing variants in these techniques like reactant ratios, temperature, reaction profile, and, most importantly here, the flux compositions, it is possible to change the products formed or fine tune reactions for product purity.

The reaction of coordination compounds of palladium, rhodium, ruthenium and nickel with thiolated ortho-carborane derivatives- 1,2-dimercapto-o-carborane (1,2-(HS)2-1,2- C2B10H10 ) and 9,12- dimercapto-o-carborane (9,12-(HS)2-1,2- C2B10H10 ) and with carbornanyl dicarboxylate has been studied. To further investigate the chemistry of o-carboranyl dithiolate, both of the reported literature dithiols have been employed to study the reactivity with some of the above mentioned transition metal coordination compounds. The 1,2-dimercapto-o-carborane reacts with bis(triphenylphosphine)palladium(II) chloride [PdCl2(PPh3)2] to form a square planar bis(triphenylphosphine)palladium(II) carboranyl dithiolate (PdC2B10S2(PPh3)2) complex.
