Chemical Analysis Of Metabolites From Mangrove Endophytic Fungus, Sefat E Munjerin

Natural products hold a significant place in drug discovery for their abundant and unique secondary metabolites. These secondary metabolites which encompass a range of organic compounds such as alkaloids, phenols, peptides, flavonoids, polyketides, and terpenoids, can easily be exploited for drug development. Endophytic fungi, residing harmoniously with its host, have emerged as generous producers of bioactive secondary metabolites, displaying efficacy against a wide array of human pathogens, including the challenging ESKAPE pathogens.

Natural products have played major roles in drug discovery and provided sources for many new drugs. Secondary metabolites, also called natural products, are organic compounds that are produced from fungi, bacteria, and other living organisms. Secondary metabolites produced by endophytic fungi include alkaloids, phenols, peptides, enzymes, polyketides, and terpenes. Endophytic fungi are rich sources of secondary metabolites and demonstrate bioactivity against many human pathogens including the ESKAPE pathogens. Chemical ecology is the study of chemical interactions between organisms and their environment mediated by small molecules involved in nonessential physiological functions, known as secondary metabolites. These compounds can be crucial to survival of the organism, and have also provided the field of medicine with some of history’s most influential drugs, referred to in that context as natural products. Antarctica is a dynamic and understudied environment, which affords the opportunity to examine the chemical ecology of unique organisms while simultaneously evaluating their novel chemistry for potential therapeutic properties as natural products.

Natural products have been a fundamental source of medicinal scaffolds for decades; with sixty percent of marketed drugs. Many synthetic chemists are focused on synthesizing potent and nontoxic compounds for pharmaceutical targets, however, nature is still proving to be a source of new bioactive compounds. Produced by the host organism for defense, reproduction and communication, secondary metabolites also demonstrate promising bioactivity against human pathogens. Hence, natural product chemists continue their quest for new leads.

New methodology has been utilized to provoke or increase targeted metabolic pathways in microbes. The low hanging fruit of natural products has been discovered over the last 50 years. To continue finding new metabolites to be used as possible drug candidates, methodology development such as those proposed herein are necessary. This methodology uses extracts from known pathogenic bacteria to elicit production of latent biosynthetic pathways from environmental bacterial isolates that may be active against the original pathogenic strains. Despite the critical role algae serve as primary producers, increases or accumulation of certain algae may result in Harmful Algal Blooms (HABs). Algal toxins from these blooms contribute significantly to incidences of food borne illness, and evidence suggests HABs are expanding in frequency and distribution. Mitigation of these HABs without knowledge of the ecological purpose and biochemical regulation of their toxins is highly unlikely. The production, function, and potential of secondary metabolites produced by the dinoflagellates Karenia brevis and Pyrodinium bahamense, were investigated.

The marine realm and, in particular, the Antarctic benthos is largely unexplored and understudied. The chemical investigation reported herein reveals not only the biodiversity, but how that biodiversity manifests remarkable chemical diversity. In our continuing study of the nudibranch Austrodoris kerguelenensis, we have isolated a diverse suite of diterpenoid glyceride esters, palmadorins D - S (2.32 - 2.47), one of which is the first reported halogenated diterpene from a dorid nudibranch. Utilizing genomic data from collaborators, we have investigated the chemical diversity from phylogenetically unique specimens collected in close proximity to one another.
