Chemical Tools And Treatments For Neurological Disorders And Infectious Diseases, Andrea Lemus
Chemical Tools And Treatments For Neurological Disorders And Infectious Diseases, Andrea Lemus
USF Tampa Graduate Theses and Dissertations
Neurological disorders have multiple etiologies such as genetics and age. Stress related disorders are becoming more common and diagnosed more often. Mental health disorders are no longer being treated as purely psychological. Mechanisms into the underlying factors of these disorders continue to bring this area to new frontiers. Chapter 2 of this dissertation focuses on the efforts made to prove the association of the protein FKBP51 to the stress response. We synthesized and used a molecular tool to discover and prove these interactions.
The study of neurodegenerative diseases is of continued importance as the world reaches record numbers of an …
Chemical Investigations Of Fungal Natural Products For Drug Discovery, Danielle H. Demers
Chemical Investigations Of Fungal Natural Products For Drug Discovery, Danielle H. Demers
USF Tampa Graduate Theses and Dissertations
Natural products have, historically, played an important role in drug discovery. Nevertheless, drug resistance, pathogen evolution, and global climate change threaten human health and nearly all current anti-infective treatments on the market today. It is undeniable that new drug discovery efforts are needed with increasing urgency.
Bolstered by a rich history of discovering treatments in the world around us, natural products chemists continue to look to the environment with increasing understanding and emerging technologies that allow efficient, effective isolation of new chemical entities. This thesis will describe one such endeavor.
Focusing on fungal natural products, herein is described the isolation …
2,4-Disubstituted Quinazolines With Antileishmanial Or Antibacterial Activity, Megan Marie Barber
2,4-Disubstituted Quinazolines With Antileishmanial Or Antibacterial Activity, Megan Marie Barber
USF Tampa Graduate Theses and Dissertations
Herein 47 2,4-disubstituted quinazolines were synthesized and tested against Leishmania donovani intracellular amastigotes. A structure-activity relationship was conducted and lead to the identification of quinazolines with EC50s in the single digit and high nanomolar range with favorable antileishmanial selectivity indexes. Quinazoline 2.6 and 2.31 underwent in vivo efficacy studies in murine models of visceral leishmaniasis, reducing liver parasitemia by 12 % and 24 %, respectively, when given by the intraperitoneal route at 15 mg/kg/day x 5 days. The antileishmanial efficacy and easy of synthesis make the 2,4-disubstituted quinazoline compound series a suitable platform for the future development of antileishmanial agents. …
Anti-Parasitic And Anti-Bacterial Agents: Studies On 1,4-Dihydropyridines And 2,4-Diaminoquinazolines, Kurt Steven Van Horn
Anti-Parasitic And Anti-Bacterial Agents: Studies On 1,4-Dihydropyridines And 2,4-Diaminoquinazolines, Kurt Steven Van Horn
USF Tampa Graduate Theses and Dissertations
Thirty-three 1,4-dihydropyridine diastereomeric pairs were synthesized and the structure-activity relationship studied in a Plasmodium falciparum in vitro model. Twenty-nine of these derivatives contained a 6-position oxygen, with 2.31, 2.32, 2.52 and 2.53 having single and double digit nanomolar activities. This SAR study revealed some insightful information about the 1,4-dihydropyridine substitution pattern. Substitution at the 7-position other than 3,4-dimethoxy severely reduced the activity. 4-phenyl substitution with 2- or 4- halo or methyl formed active compounds while substitution at the 3-position or with methoxy or conjugated aryl systems resulted in inactive compounds. The 2-position was found to majorly affect the activity, with …
