This project focuses on the optimization of benzothiazole-based chromophores to utilize them as fluorescent tags functionalized onto stimuli-responsive, or “smart,” polymers as non-viral gene delivery vectors for gene therapy applications. Blue-fluorescent emissive chromophores will allow tracking capabilities for the transfection pathway of the vector to be monitored as it delivers the DNA payload within intercellular space. The most appropriate chromophore must be covalently bound to a free amine within the vector, which is accomplished through NAS chemistry from fluorophenyl-benzothiazoles (F-BTZ-CBz) derivatives to amidated hyperbranched poly(ethyleneimine)(HPEI-IBAm0.61). A series of 3,5-difluorinated systems, which are activated towards nucleophilic aromatic substitution (NAS) by the strongly electron withdrawing benzoxazole or benzothiazole groups, located meta to the fluorines were prepared and fully characterized. The monomers also carry various N-heterocyclic species on the non-fluorinated ring. The corresponding poly(arylene ether)s, some of which were copolymers with triphenylphosphine oxide-based monomers, were prepared via standard NAS polycondensation reactions. Incorporation of the monomers containing N-heterocycle units was determined by NMR spectroscopy. Characterization of the thermal properties was done using thermogravimetric analysis (TGA) and differential scanning (DSC). Most of the polymers displayed good film forming properties when …

A series of hydroquinone-based poly(arylene ether) copolymers, with truly pendent sulfonated phenyl sulfonyl groups and either benzothiazole or benzoxazole groups was synthesized for use as proton exchange membranes (PEM). Two homopolymers series, benzothiazole and benzoxazole based, were prepared via meta activated nucleophilic aromatic substitution polycondensation reactions. Two copolymer series of N-heterocyclic and sulfonated phenyl sulfonyl groups were synthesized via a similar method. The ratio of 3,5-difluoro-3'-sulfonated diphenylsulfone to 2-[3,5-bis-(4-hydroxyphenoxy)]benzothiazole or 2-[3,5-bis-(4-hydroxyphenoxy)]benzoxaole was varied to afford ion-exchange capacities ranging from 0.60 to 1.39 meq and 0.62 to 1.42 meq, respectively.
