Algal Biomass As A Binding Agent For The Densification Of Miscanthus, Sunil Thapa, Daniel B. Johnson, Peter Ping Liu, Thomas Canam

Thomas Canam

Purpose: Lignocellulosic biomass has been regarded as an important future energy source due to its excessive availability; however, the wide application of this material for many applications is restricted by the high costs associated with densification, transportation, thermo-chemical pretreatment and conversion. In order to increase the density of lignocellulosic biomass, it is typically compressed into pellets or briquettes. This frequently requires the addition of additives, which may negatively impact the economics of the process. Environmentally-friendly binding agents that can be obtained inexpensively are therefore desirable. This study examines the change in physicochemical properties of densified Miscanthus straw where algae were …

Modeled Impacts Of Cover Crops And Vegetative Barriers On Corn Stover Availability And Soil Quality, Ian J. Bonner, David J. Muth Jr., Joshua B. Koch, Douglas L. Karlen

David J. Muth

Environmentally benign, economically viable, and socially acceptable agronomic strategies are needed to launch a sustainable lignocellulosic biofuel industry. Our objective was to demonstrate a landscape planning process that can ensure adequate supplies of corn (Zea mays L.) stover feedstock while protecting and improving soil quality. The Landscape Environmental Assessment Framework (LEAF) was used to develop land use strategies that were then scaled up for five U.S. Corn Belt states (Nebraska, Iowa, Illinois, Indiana, and Minnesota) to illustrate the impact that could be achieved. Our results show an annual sustainable stover supply of 194 million Mg without exceeding soil erosion T …

Sensitivity Analysis And Parameter Identification For A Nonlinear Time-Delay System In Microbial Fed-Batch Process, Chongyang Liu

Chongyang Liu

Developing suitable dynamic models of bioprocess is a difficult issue in bioscience. In this paper, considering the microbial metabolism mechanism, i.e., the production of new biomass is delayed by the amount of time it takes to metabolize the nutrients, in glycerol bioconversion to 1,3-propanediol, we propose a nonlinear time-delay system to formulate the fed-batch fermentation process. Some important properties are also discussed. Then, in view of the effect of time-delay and the high number of kinetic parameters in the system, the parametric sensitivity analysis is used to determine the key parameters. Finally, a parameter identification model is presented and a …

Microfabricated Nanotopological Surfaces For Study Of Adhesion-Dependent Cell Mechanosensitivity, Weiqiang Chen, Yubing Sun, Jianping Fu

Weiqiang Chen

Cells exhibit high sensitivity and diverse responses to the intrinsic nanotopography of the extracellular matrix through their nanoscale cellular sensing machinery. A simple microfabrication method for precise control and spatial patterning of the local nanoroughness on glass surfaces by using photolithography and reactive ion etching is reported. It is demonstrated that local nanoroughness as a biophysical cue could regulate a diverse array of NIH/3T3 fi broblast behaviors, including cell morphology, adhesion, proliferation, migration, and cytoskeleton contractility. The capability to control and further predict cellular responses to nanoroughness might suggest novel methods for developing biomaterials mimicking nanotopographic structures in vivo for …

Surface-Micromachined Microfiltration Membranes For Efficient Isolation And Functional Immunophenotyping Of Subpopulations Of Immune Cells, Weiqiang Chen, Nien-Tsu Huang, Boram Oh, Raymond H. W. Lam, Rong Fan, Timothy T. Cornell, Thomas P. Shanley, Katsuo Kurabayashi, Jianping Fu

Weiqiang Chen

An accurate measurement of the immune status in patients with immune system disorders is critical in evaluating the stage of diseases and tailoring drug treatments. The functional cellular immunity test is a promising method to establish the diagnosis of immune dysfunctions. The conventional functional cellular immunity test involves measurements of the capacity of peripheral blood mononuclear cells to produce pro-inflammatory cytokines when stimulated ex vivo. However, this “bulk” assay measures the overall reactivity of a population of lymphocytes and monocytes, making it difficult to pinpoint the phenotype or real identity of the reactive immune cells involved. In this research, we …

Weiqiang Chen

Circulating tumor cells (CTCs) detached from both primary and metastatic lesions represent a potential alternative to invasive biopsies as a source of tumor tissue for the detection, characterization and monitoring of cancers. Here we report a simple yet effective strategy for capturing CTCs without using capture antibodies. Our method uniquely utilized the differential adhesion preference of cancer cells to nanorough surfaces when compared to normal blood cells and thus did not depend on their physical size or surface protein expression, a significant advantage as compared to other existing CTC capture techniques.

Cosmic ray particles image series acquired using a Hamamatsu ORCA-II ERG scientific grade CCD camera, cooled to -60 C. Each image is a consecutive 600 second (10 minute) exposure time with no light to the camera.

While processing the data, I discoverd that the background changed around planes 25 and 227 (see Excel file and jpeg screenshots), so I also processed only planes 025-227 (203 planes total, 2030 minutes, 33.83 hours). the CCD industry "rule of thumb" for a "typical" CCD sensor (i.e. 1/3" CCD) is that one cosmic ray particle strikes a sensor approximately every 30 seconds (assuming not …

Environmental Impacts Of Stover Removal In The Corn Belt, Alicia English, Wallace E. Tyner, Juan Sesmero, Phillip Owens, David J. Muth Jr. Aug 2012

Environmental Impacts Of Stover Removal In The Corn Belt, Alicia English, Wallace E. Tyner, Juan Sesmero, Phillip Owens, David J. Muth Jr.

When considering the market for biomass from corn stover resources erosion and soil quality issues are important to consider. Removal of stover can be beneficial in some areas, especially when coordinated with other conservation practices, such as vegetative barrier strips and cover crops. However, benefits are highly dependent on several factors, namely if farmers see costs and benefits associated with erosion and the tradeoffs with the removal of biomass. This paper uses results from an integrated RUSLE2/WEPS model to incorporate six different regime choices, covering management, harvest and conservation, into a simple profit maximization model to show these tradeoffs.

Human embryonic stem cells (hESCs) have great potentials for future cell-based therapeutics. However, their mechanosensitivity to biophysical signals from the cellular microenvironment is not well characterized. Here we introduced an effective microfabrication strategy for accurate control and patterning of nanoroughness on glass surfaces. Our results demonstrated that nanotopography could provide a potent regulatory signal over different hESC behaviors, including cell morphology, adhesion, proliferation, clonal expansion, and self-renewal. Our results indicated that topological sensing of hESCs might include feedback regulation involving mechanosensory integrin-mediated cell matrix adhesion, myosin II, and E-cadherin. Our results also demonstrated that cellular responses to nanotopography were cell-type …

Integrative Bayesian Analysis Of High-Dimensional Multi-Platform Genomics Data, Wenting Wang, Veerabhadran Baladandayuthapani, Jeffrey S. Morris, Bradley M. Broom, Ganiraju C. Manyam, Kim-Anh Do

Jeffrey S. Morris

Motivation: Analyzing data from multi-platform genomics experiments combined with patients’ clinical outcomes helps us understand the complex biological processes that characterize a disease, as well as how these processes relate to the development of the disease. Current integration approaches that treat the data are limited in that they do not consider the fundamental biological relationships that exist among the data from platforms.

Statistical Model: We propose an integrative Bayesian analysis of genomics data (iBAG) framework for identifying important genes/biomarkers that are associated with clinical outcome. This framework uses a hierarchical modeling technique to combine the data obtained from multiple platforms …

Optimal Switching Control Of A Fed-Batch Fermentation Process, Chongyang Liu, Zhaohua Gong, Enmin Feng, Hongchao Yin

Chongyang Liu

Considering the hybrid nature in fed-batch culture of glycerol biconversion to 1,3-propanediol (1,3-PD) by Klebsiella pneumoniae, we propose a state-based switching dynamical system to describe the fermentation process. To maximize the concentration of 1,3-PD at the terminal time, an optimal switching control model subject to our proposed switching system and constraints of continuous state inequality and control function is presented. Because the number of the switchings is not known a priori, we reformulate the above optimal control problem as a two-level optimization problem. An optimization algorithm is developed to seek the optimal solution on the basis of a heuristic approach …

Nonlinear Dynamical Systems Of Fed-Batch Fermentation And Their Optimal Control, Chongyang Liu, Zhaohua Gong, Enmin Feng, Hongchao Yin

Chongyang Liu

In this article, we propose a controlled nonlinear dynamical system with variable switching instants, in which the feeding rate of glycerol is regarded as the control function and the moments between the batch and feeding processes as switching instants, to formulate the fed-batch fermentation of glycerol bioconversion to 1,3-propanediol (1,3-PD). Some important properties of the proposed system and its solution are then discussed. Taking the concentration of 1,3-PD at the terminal time as the cost functional, we establish an optimal control model involving the controlled nonlinear dynamical system and subject to continuous state inequality constraints. The existence of the optimal …

Modeling And Parameter Identification Involving 3-Hydroxypropionaldehyde Inhibitory Effects In Glycerol Continuous Fermentation, Zhaohua Gong, Chongyang Liu, Yongsheng Yu

Chongyang Liu

Mathematical modeling and parameter estimation are critical steps in the optimization of biotechnological processes. In the 1,3-propanediol (1,3-PD )production by glycerol fermentation process under anaerobic conditions, 3-hydroxypropionaldehyde (3-HPA) accumulation would arouse an irreversible cessation of the fermentation process. Considering 3-HPA inhibitions to cells growth and to activities of enzymes, we propose a novel mathematical model to describe glycerol continuous cultures. Some properties of the above model are discussed. On the basis of the concentrations of extracellular substances, a parameter identification model is established to determine the kinetic parameters in the presented system. Through the penalty function technique combined with an …

Optimal Control Of A Fed-Batch Fermentation Involving Multiple Feeds, Chongyang Liu, Zhaohua Gong, Zhaoyi Huo, Bangyu Shen

Chongyang Liu

A nonlinear dynamical system, in which the feed rates of glycerol and alkali are taken as the control functions, is first proposed to formulate the fed-batch culture of 1,3-propanediol (1,3-PD) production. To maximize the 1,3-PD concentration at the terminal time, a constrained optimal control model is then presented. A solution approach is developed to seek the optimal feed rates based on control vector parametrization method and improved differential evolution algorithm. The proposed methodology yielded an increase by 32.17% of 1,3-PD concentration at the terminal time.

Improving Biomass Logistics Cost Within Agronomic Sustainability Constratints And Biomass Quality Targets, Kevin L. Kenney, J. Richard Hess, William A. Smith, David J. Muth Jr. Jan 2012

Improving Biomass Logistics Cost Within Agronomic Sustainability Constratints And Biomass Quality Targets, Kevin L. Kenney, J. Richard Hess, William A. Smith, David J. Muth Jr.

David J. Muth

Equipment manufacturers have made rapid improvements in biomass harvesting and handling equipment. These improvements have increased transportation and handling efficiencies due to higher biomass densities and reduced losses. Improvements in grinder efficiencies and capacity have reduced biomass grinding costs. Biomass collection efficiencies (the ratio of biomass collected to the amount available in the field) as high as 75% for crop residues and greater than 90% for perennial energy crops have also been demonstrated. However, as collection rates increase, the fraction of entrained soil in the biomass increases, and high biomass residue removal rates can violate agronomic sustainability limits. Advancements in …

Advanced biofuels will be developed using cellulosic feedstock rather than grain or oilseed crops that can also be used for food and feed. To be sustainable, these new agronomic production systems must be economically viable without degrading the soil and other natural resources. This review examines six agronomic factors that collectively define many of the limits and opportunities for harvesting crop residue for biofuel feedstock in the midwestern United States. The limiting factors include soil organic carbon, wind and water erosion, plant nutrient balance, soil water and temperature dynamics, soil compaction, and off-site environmental impacts. These are discussed in relationship …

Modeling And Optimal Control Of A Nonlinear Dynamical System In Microbial Fed-Batch Fermentation Process, Chongyang Liu, Zhaohua Gong, Enmin Feng

Chongyang Liu

The mathematical model and optimal control of microbial fed-batch fermentation is considered in this paper. Since it is decisive for increasing the productivity of 1,3-propanediol (1,3-PD) to optimize the feeding rate of glycerol and the switching instants between the batch and feeding processes in the fermentation process, we propose a new nonlinear dynamical system to formulate the process. In the system, the switching instants are variable and the feeding rate of glycerol is regarded as the control function. Some important properties of the proposed system and its solution are then discussed. To maximize the concentration of 1,3-PD at the terminal …

Computational Method For Inferring Objective Function Of Glycerol Metabolism In Klebsiella Pneumoniae, Zhaohua Gong, Chongyang Liu, Enmin Feng, Qingrui Zhang

Chongyang Liu

Flux balance analysis (FBA) is an effective tool in the analysis of metabolic network. It can predict the flux distribution of engineered cells, whereas the accurate prediction depends on the reasonable objective function. In this work, we propose two nonlinear bilevel programming models on anaerobic glycerol metabolism in Klebsiella pneumoniae (K. pneumoniae) for 1,3-propanediol (1,3-PD) production. One intends to infer the metabolic objective function, and the other is to analyze the robustness of the objective function. In view of the models’ characteristic an improved genetic algorithm is constructed to solve them, where some techniques are adopted to guarantee all chromosomes …

Modelling And Optimal Control For Nonlinear Multistage Dynamical System Of Microbial Fed-Batch Culture, Chongyang Liu, Zhaohua Gong, Enmin Feng, Hongchao Yin

Chongyang Liu

In this paper, we propose a new controlled multistage system to formulate the fed-batch culture process of glycerol bio-dissimilation to 1,3-propanediol (1,3-PD) by regarding the feeding rate of glycerol as a control function. Compared with the previous systems, this system doesn’t take the feeding process as an impulsive form, but a time-continuous process, which is much closer to the actual culture process. Some properties of the above dynamical system are then proved. To maximize the concentration of 1,3-PD at the terminal time, we develop an optimal control model subject to our proposed controlled multistage system and continuous state inequality constraints. …

The goal of planning a horizontal well path is to obtain a trajectory that arrives at a given target subject to various constraints. In this paper, the optimal control problem subject to a nonlinear multistage dynamical system (NMDS) for horizontal well paths is investigated. Some properties of the multistage system are proved. In order to derive the optimality conditions, we transform the optimal control problem into one with control constraints and inequality-constrained trajectories by defining some functions. The properties of these functions are then discussed and optimality conditions for optimal control problem are also given. Finally, an improved simplex method …

So a proper feeding rate is required during the process. Taking the concentration of 1, 3-PD at the terminal time as the performance index and the feeding rate of glycerol as the control function, we propose an optimal control model subject to a nonlinear dynamical system and constraints of continuous state and non-stationary control. A computational approach is constructed to seek the solution of the above model in two aspects. On the one hand we …

Some properties of the controlled switching system are obtained. Subsequently, we prove the existence of optimal control. In order to deduce the optimality conditions, we transcribe the optimal switching control model into an equivalent one with fixed switching instants and parameters. Finally, the optimality conditions of the equivalent problem are investigated …
