Multidimensional Mass Spectrometry Of Chemonic™ Ccg-6 Nonionic Surfactant With Separation By Polarity And Shape, Charles Johnson

Williams Honors College, Honors Research Projects

Chemonic™ CCG-6 surfactant is a water-soluble poly(ethylene glycol) (PEG) conjugated alkyl glyceride emollient. This surfactant exists as a complex mixture of a glycerol cores conjugated with poly(ethylene glycol) branches (PEGylation) that were partially esterified with caprylic (C8) and capric (C10) acids. These may be esterified on one, two, or all three arms of the glyceride. The architecture of the structures in this mixture was studied using multidimensional mass spectrometry (MS). Mass spectrometry was interfaced with ultra-performance liquid chromatography (UPLC) and ion mobility (IM) separation. The mixture was separated by reversed-phase LC, oligomers of the star-branched polymer were separated according to …

Interfacial Corrosion Of Copper And The Formation Of Copper Hydroxychloride, Mary Teague, Shengxi Li, Hongbo Cong

Williams Honors College, Honors Research Projects

Electrical circuitry is an industry, among many others, heavily using the element of copper. Ensuring the mechanical integrity of Cu is crucial, especially in salt environments, for the multifaceted composition of circuits. 4N NaCl solution (equilibrium concentration in ~84% RH) simulated this three-phase system. Rectangular Cu samples were partially immersed in both ambient and continuous lab air sparging atmospheres to understand waterline corrosion of the metal. Open circuit potentials (OCP) were continuously taken during the immersion testing for a maximum of 5 days. A scanning electron microscopy (SEM) equipped with an energy dispersive X-ray spectrometer (EDS), Raman spectroscopy, and 3-D …

Tandem Mass Spectrometry (Ms/Ms) For Determination Of Architecture Of Synthetic Polymers, Shoumik Saha

Williams Honors College, Honors Research Projects

Tandem Mass Spectrometry (MS/MS) was used to determine the architecture of synthetic polymers and they are difluorene-N3, 3-difluorene-N3 and VPOSS-3-difluorene. VPOSS materials usually exhibit electrochemical properties and the research was performed to determine their accurate structure and to provide evidence of their proper synthesis. The compounds were mixed with a matrix solution, DCTB, and a silver salt to enable proper protonation. The solvent mixture was on a MALDI plate and then put into the mass spectrometer. The mass spectrum generated was analyzed and each fragment was identified to structures with the synthetic polymers via ChemDraw. Thus, trial …

Metabolomic Profiling Of Chiari Malformation Type I: Comparison Of Bioinformatic Programs For Untargeted Analysis, Hunter W. Korsmo Jan 2017

Metabolomic Profiling Of Chiari Malformation Type I: Comparison Of Bioinformatic Programs For Untargeted Analysis, Hunter W. Korsmo

Williams Honors College, Honors Research Projects

Chiari Malformation Type I is a neurodegenerative trait that can result from disease or from acquiring. Metabolomic analysis was done on normal pressure hydrocephalous and Chiari CSF samples using LC-MS and multiple bioinformatic programs. After analysis from multiple programs, we were able to analyze the stringency in statistical algorithms done by each program and determined qualities that are shared between programs that offer multiple details. We identified dysregulation in glucuronated metabolites in CSF of Chiari versus NPH. Using LC-MS, we established the experimental MS/MS of glucuronic acid in attempt to identify similarities in mass-to-charge features primarily identified. We could not …

3d Printing Applications Within Spectrophotometry, Eric J. Stanczyk Jan 2016

3d Printing Applications Within Spectrophotometry, Eric J. Stanczyk

Williams Honors College, Honors Research Projects

Ultraviolet-visible spectroscopy is a tool used throughout the field of chemistry and in chemical labs across the world. Spectrophotometers are a core technology in analytical chemistry, and are used to obtain accurate data on solution concentration. Unfortunately spectrophotometers can be difficult to obtain due to their high cost and low availability outside of a laboratory; this is especially true in either high school or lower grades where buying a spectrophotometer could be considered unreasonable due to price. There are ways to obtain cheaper spectrophotometers, but they can have a high initial cost or low overall quality. This paper will discuss …

Mutagenesis-Based Active-Site Characterization Of The Diabetes Drug Target Mitoneet, Lisa Mickley Jan 2016

Mutagenesis-Based Active-Site Characterization Of The Diabetes Drug Target Mitoneet, Lisa Mickley

Williams Honors College, Honors Research Projects

This project investigates the cluster stability of an outer mitochondrial membrane protein mitoNEET under mutational stress and drug binding analysis. MitoNEET is a part of a class of proteins that coordinate an Fe-S molecule into its tertiary structure. This protein is believed to facilitate the oxidative capacity of the electron transport chain by reversible loss of its coordinated Fe-S molecule. The research of this paper uses site directed protein mutagenesis to selectively alter the three cysteine, one histidine cluster coordination into more common coordination patterns such as two cysteine, two histidine or four cysteine. The ultimate goal is to optimize …

Determination Of The Structure Of Sphingomyelin By Tandem Mass Spectrometry And Matrix - Assisted Laser Desorption/Ionization, Antonio Ganios

Williams Honors College, Honors Research Projects

Mass spectrometry is a method that uses masses to determine the structure of a compound. This is completed by first charging the compound until it is a cation or anion, and then its molecular weight can be determined by measuring a mass to charge ratio. The focus of this study was determination of sphingomyelin’s structure. The purpose of this was that sphingomyelin has been shown to play a critical role in many biological pathways and being able to identify this lipid could prove to be beneficial for preventative medicine. Utilizing electrospray ionization (ESI) Quadruple-Time-of-flight (Q-TOF) mass spectrometry on a Waters …

Characterization Of Noncovalent Complexes Of Polyelectrolytes By Mass Spectrometry, Anthony J. Marino Jan 2015

Characterization Of Noncovalent Complexes Of Polyelectrolytes By Mass Spectrometry, Anthony J. Marino

Williams Honors College, Honors Research Projects

Various ions of poly-L-lysine and polystyrene sulfonic acid were successfully observed first within a matrix-assisted laser desorption/ionization mass spectrometer and subsequently in an electrospray ionization mass spectrometer. The successful ionization of these polymers allowed for studies to be performed on the noncovalent interactions between the two. Noncovalent interactions were effectively studied on both instruments, but further quantified using tandem mass spectrometry found within the MALDI system and ion-mobility mass spectrometry found within the electrospray ionization system. As a result of employing the use of ion-mobility, various different stoichiometries and charges of polyelectrolyte complexes could be distinguished.
