Thirty-two canine tibiae with patellae and patellar tendons were harvested from 17 skeletally mature cadavers. Each tibia was randomly allocated to a construct group: plate and pin (Plate), plate with countersink compression screw (HCS), plate with tension band (TB), or plate with HCS and TB (HCSTB). Samples were loaded by distraction until failure. The stiffness, yield load, and ultimate load were compared between each fixation method. No difference in stiffness of the constructs was detected between groups. Yield load and ultimate load for the HCSTB group was greater than the TB group, the HCS group, and the Plate group. CBLO …

Discomfort in the hocks of horses can cause lameness, resulting in poor performance. Diagnosis by veterinarians typically includes evaluating movement or use of ultrasound or radiography to evaluate tendons, ligaments, or bones. Treatment methods and times vary depending on diagnosis, but often include joint injections, medications, and stall rest. Treatments can be costly both financially and regarding time off from practice or competition. The most common best management practice used to combat diseases in horses is vaccination, which can decrease the incidence or severity of illness. However, the duration of immunity will vary for each vaccination and must be considered for revaccination purposes. Titer concentration, which evaluates antibodies in the blood, may be a helpful way of determining how long an animal has immunity to a disease. Leptospirosis, a zoonotic, bacterial disease, can result in uveitis, potentially leading to blindness, and abortion in mares. Bovine respiratory disease is a multifaceted disease with agent, host, and environmental factors. Stocker calves are at high risk of contracting the disease through many stressors like weaning, transportation, commingling, etc. The purpose of this project was to test if externally observed physical characteristics of calves on arrival at a stocker facility can be used to predict calfhood vaccination status determined based on antibody titer levels. The objective of this thesis was to determine the anticoccidial profile of Eimeria spp. derived from wild turkeys. These drug-sensitive Eimeria spp. have potential to be used as vaccine candidates to control coccidiosis in commercial turkeys. Chapter two consists of a brief literature review focused on coccidiosis in turkeys. Chapter three addresses the methodologies utilized to isolate, recover, and speciate Eimeria recovered from wild turkey feces in the eastern two-thirds of the United States. From this, we successfully obtained single oocyst-derived stocks for E. meleagrimitis, E. dispersa, E. meleagridis, E. gallopavonis, and E. adenoeides. To increase capacity for monitoring and surveillance of tick-borne diseases, publicly available tick distribution and climate change datasets are required to create accurate predictive distribution models. It is difficult, however, to assess model accuracy and utility when using incomplete datasets.  The more recent development of comprehensive tick databases for Europe and availability of climate change scenarios from multiple IPCC Assessment Reports allows for improved modeling efforts. Multiple tick datasets were combined and three climate change projections were compared by predicting current and future distributions of Ixodes ricinus ticks in Europe using the MaxEnt species distribution model. Despite being ecotypes, only body lice are known to transmit bacterial diseases to humans, and it appears that lower humoral and cellular immune responses allow body lice to possess a higher vector competence. To supplement the Government’s effort on rabies control, the Victoria Falls Wildlife Trust and Veterinarians for Animal Welfare Zimbabwe have joined forces to carry out dog rabies vaccinations in rural communities adjacent to the major tourist attraction of The Victoria Falls. Frontal plane slab fractures account for the majority of third carpal bone (C3) fractures in performance horses. Treatment is stabilization with an AO cortical screw. Complications are fragment splitting, fragment spinning, and irritation of dorsal soft tissue structures. A novel, headless, cannulated screw with interlocking threads (the Headless Compression Screw Fastener, HCSF) has been developed to resist multidirectional forces. Simulated C3 slab fractures were created in nine paired equine carpi. HCSF or AO cortical screws were loaded in shear to failure. Twelve pairs of cadaveric pelvic limbs were harvested from skeletally mature dogs. Tibias were randomly assigned pairwise to two study groups: locking buttress screw fixation (LBS) and bone-screw-fastener fixation (BSF). A tibial plateau leveling osteotomy (TPLO) was performed. Each specimen was positioned in a servohydraulic testing machine to simulate physiological orientation and loading at the mid-point of the stance phase at the walk. Cyclic loading was performed for 30,000 cycles at 4Hz with a peak-load of 1000N. Lameness is a major issue in animal welfare and the broiler industry. Bacterial chondronecrosis with osteomyelitis (BCO) is one of the main causes of lameness. Many staphylococcal species, including Staphylococcus agnetis isolate 908, have been isolated from the bones and blood of lame broilers at the University of Arkansas. Staphylococcus agnetis is a coagulase-variable, Gram-positive bacterial species that has been previously associated with subclinical or mild clinical cases of mastitis in dairy cattle. Chronic pancreatitis in dogs is typically managed with a low-fat diet. Human research suggests consuming medium-chain triglycerides (MCT) may lower pancreatic enzyme release compared to consuming long chain fatty acids (LCFA). Biliary diseases are uncommon, potentially fatal causes of acute abdomen in dogs. Little information is present comparing the performance of computed tomography (CT) to ultrasound in identifying canine biliary pathology. Thirty-five client-owned dogs presenting for acute abdomen signs received an abdominal ultrasound and contrast-enhanced abdominal CT. Two authors reviewed the randomized, anonymized CT and ultrasound studies. Twenty-eight dogs had biliary pathology and seven dogs serving as controls had no evidence of biliary disease. The final diagnoses of patients with biliary pathology included cholelithiasis, gallbladder mucoceles, cholangiohepatitis/cholangitis, extrahepatic biliary obstruction, gallbladder wall edema, gallbladder wall mass, and cystic mucosal hyperplasia. Dirofilaria immitis is a common and largely preventable mosquito-borne parasite that causes dirofilariasis, or heartworm disease, in a number of animal species. The necessity of heartworm prevention in the canine patient is undisputed and well-known among veterinary professionals, yet there are still many dogs not protected from this fatal parasite. Heartworm prevention traditionally requires a once-a-month administration of medication; therefore, its efficacy relies on owner compliance. Background: The GI microbiome has not been characterized in dogs being medically managed for congenital portosystemic shunts [CPSS].

Objectives: To characterize the fecal microbiome in a population of dogs being medically managed for CPSS.

Animals: 27 client-owned dogs.

Methods: Prospective cohort study enrollment of fecal samples was performed with follow-up data collected retrospectively. The overall fecal dysbiosis index [DI] and individual bacterial abundances were determined using real-time qPCR. However, few studies have investigated the in vivo immune response to LTA, and none of the in vivo studies done have been performed in birds. Opioid drugs have the potential of provide local analgesia in inflamed joints. To date, morphine is the only opioid that has been tested for intra-articular (IA) administration in horses. Having an alternative drug, other than morphine, could widen the therapeutic options, particularly in cases of drug shortages or inaccessibility to specific drugs. The work presented in this dissertation reports the cytotoxic effects of buprenorphine on cultured equine chondrocytes, and the pharmacokinetic, pharmacodynamic and analgesic effects, of IA administered buprenorphine in horses with experimentally induced synovitis.

The canine lungs are well suited for ventilation, gas exchange, and gas transport. In diseased states, changes within bronchi and pulmonary parenchyma alter the blood-gas interface. Diagnostic workups to characterize pulmonary disease may include blood gas analysis, bronchoalveolar lavage, tracheobronchoscopy, radiography and/or computed tomography. Surgical pulmonary diseases include cysts, bullae, and blebs; congenital anomalies; consolidation or abscessation; bronchiectasis; lacerations; lung lobe torsions; and neoplasia. Surgical options may include laceration repair, partial or total lung lobectomy, and pneumonectomy. These options are most commonly utilized in cases that fail medical management. Tick-borne diseases threaten the health of humans, animals, and ecosystems, but surveillance efforts are lacking in some regions. Throughout 2019, 2020, and 2021, we conducted tick surveys aimed at determining the status of the black-legged tick (Ixodes scapularis), the American dog tick (Dermacentor variabilis) and the lone star tick (Amblyoma americanum) in eastern South Dakota. Each year, ticks were collected using a flagging method and identified in the lab. Post-traumatic stress disorder (PTSD) is associated with elevated allostatic load, nearly double the risk for metabolic syndrome, reduced hippocampal volume, and contextual memory processing deficits. Emerging evidence suggests that these stress effects may predispose individuals to the development of PTSD, and there is a known relationship between chronic stress and metabolic dysfunction. In this work, we utilized two rat models of PTSD to explore these connections. Bone resorptive diseases affect humans and horses, alike. Examples of these diseases include osteoporosis, Paget’s disease, and osteogenesis imperfecta in humans, and navicular disease in the horse (Mitchell et al., 2019; Suva et al., 2021). Bisphosphonates are used to treat these diseases, and may remain in the bone for several years after administration, demonstrating the need for newer drug testing methods.

With an increasing senior horse population (< 15 years old), alternatives to the use of non-steroidal anti-inflammatory drugs (NSAIDs) such as phenylbutazone, firocoxib, and flunixin meglumine, are desired as these common anti-inflammatory treatments have negative side effects. Because senior horses may have chronic low-grade inflammation, termed inflamm-aging, they are a natural model to investigate the efficacy of CBD on inflammatory responses as well as other health parameters. Given this interest, and due to the lack of research conducted on CBD in the horse, the overall objective of this dissertation work was to conduct both basic and applied research investigating the effects of CBD on equine immune function and health. Pancreatic beta-cell replacement is recognized for feasible type 1 diabetes (T1D) treatment. However, in post-transplantation, the autoimmune destruction incidentally attacks the activity and survival of beta-cells are reported in animal and human. To address these concerns, the generation of immortalized, biocompatible beta-cells, and the engraftment platform are insightfully investigated. The stepwise chemical process was used for in vitro Insulin-producing cells (IPCs) production from mouse gingival fibroblast-induced pluripotent stem cells (mGF-iPSCs). The real-time qRT-PCR, glucose stimulation C-peptide/Insulin secretion, immunostaining, and visible cell methods were examined during IPC differentiation.
