Pigs challenged with hypoglycosylated variants of Porcine Reproductive and Respiratory Syndrome Virus (PRRSV) major surface protein GP5 exhibited increased production of PRRSV-neutralizing antibodies relative to the levels of neutralizing antibodies produced by pigs immunized with wild type (wt) or glycosylated GP5. This invention provides for methods of obtaining improved immune responses in pigs to PRRSV, compositions useful for obtaining the improved immune responses as well as isolated polynucleotides that encode hypoglycosylated variants of PRRSV major surface protein GP5.

The nucleocapsid protein (N) of vesicular stomatitis virus and other rhabdoviruses plays a central role in the assembly and template functions of the viral N-RNA complex. The crystal structure of the viral N-RNA complex suggests that the central region of the N protein interacts with the viral RNA. Sequence alignment of rhabdovirus N proteins revealed several highly conserved regions, one of which spanned residues 282 to 291 (GLSSKSPYSS) in the central region of the molecule. Alanine-scanning mutagenesis of this region suggested that replacement of the tyrosine residue at position 289 (Y289) with alanine resulted in an N-RNA template that is …

A recombinant vesicular stomatitis virus (VSV-PeGFP-M-MmRFP) encoding enhanced green fluorescent protein fused in frame with P (PeGFP) in place of P and a fusion matrix protein (monomeric red fluorescent protein fused in frame at the carboxy terminus of M [MmRFP]) at the G-L gene junction, in addition to wild-type (wt) M protein in its normal location, was recovered, but the MmRFP was not incorporated into the virions. Subsequently, we generated recombinant viruses (VSV-PeGFP-∆M-Mtc and VSV-∆M-Mtc) encoding Mprotein with a carboxy-terminal tetracysteine tag (Mtc) in place of the M protein. A potentially painful experience may modify normal behavioural responses. To gauge the importance of pain relative to predation or social status, we presented competing stimuli, a predator cue or an unfamiliar social group, to two groups of noxiously treated rainbow trout, Oncorhynchus mykiss. In the predator cue experiment, fish were classified as bold or shy. Noxiously stimulated fish did not show antipredator responses, suggesting that pain is the imperative. In the social status experiment, noxiously stimulated fish held individually and undisturbed showed an increase in respiration rate and plasma cortisol. Newby and Stevens’ (2008) paper ‘‘The effects of the acetic acid ‘pain’ test on feeding, swimming, and respiratory responses of rainbow trout (Oncorhynchus mykiss)’’ examines the effects of a noxious stimulus on the behaviour of trout in an attempt to replicate research conducted in my laboratory (Sneddon, 2003a; Sneddon, 2007; Sneddon et al., 2003a,b; Reilly et al., 2008). However, the authors used a different protocol to the one already published and I would like to respond to some of their points of discussion to provide scientific explanations for their results using data from my laboratory. Feeding individualized rations for lactating dairy cattle in the commercial herd based on dietary needs is a popular issue. This has been a debating topic since evolution of the Total Mixed Ration (TMR). Retinal ganglion cells that express the photopigment melanopsin are intrinsically photosensitive (ipRGCs) and exhibit robust synaptically driven ON-responses to light, yet they will continue to depolarize in response to light when all synaptic input from rod and cone photoreceptors is removed. The light-evoked increase in firing of classical ganglion cells is determined by synaptic input from ON-bipolar cells in the proximal sublamina of the inner plexiform layer. OFF-bipolar cells synapse with ganglion cell dendrites in the distal sublamina of the inner plexiform layer. We developed a method for somatic cell nuclear transfer in zebrafish using laser-ablated metaphase II eggs as recipients, the micropyle for transfer of the nucleus and an egg activation protocol after nuclear reconstruction. We produced clones from cells of both embryonic and adult origins, although the latter did not give rise to live adult clones.

Prevalence Of Intestinal Parasite Infection In Symptomatic And Asymptomatic Dogs In Southwestern West Virginia: The Potential Impact On Human Health, Tashina Marie Savilla

Most dog owners are unaware of asymptomatic infection and of the possibility of contracting zoonotic parasites from their dogs. We hypothesized that parasite infection is: associated with one or more symptoms; independent of gender and age; and independent of anthelmintic usage. Stool samples were collected from 231 dogs in Kanawha County, West Virginia, and were examined by simple fecal flotation. Parasitic prevalence was found to be 23% for Ancylostoma caninum, 8% for Trichuris vulpis, 7% for Toxocara canis, 4% for Isospora species, and 32.5% overall. There was no significant relationship between infection and gender, nor between infection and symptom for …

The testes and the spermatic cord of raccoons (Procyon lotor, kits to adult breeders; n = 48) were examined. Segmental arteritis confined to the extratesticular portions of the testicular artery was present in raccoons of all ages. The arterial changes were seen in laboratory-confined experimental and control animals as well as in wild-caught raccoons. The lesions consisted of proliferative endarteritis with presence of inflammatory cells within the intima, media, and the adventitial regions of most affected vessels. Some aspects of the proliferative arterial lesions were reminiscent of systemic necrotizing vasculitis (polyarteritis nodosa), an immunologically mediated condition of animals and humans. …

The neuroinvasive property of several alpha-herpesviruses underlies an uncommon infectious process that includes the establishment of life-long latent infections in sensory neurons of the peripheral nervous system. Several herpesvirus proteins are required for replication and dissemination within the nervous system, indicating that exploiting the nervous system as a niche for productive infection requires a specialized set of functions encoded by the virus. Whether initial entry into the nervous system from peripheral tissues also requires specialized viral functions is not known. Bovine herpesvirus 1 (BoHV-1) infected cell protein 0 (bICP0) is an important transcriptional regulatory protein that stimulates productive infection. In transient transfection assays, bICP0 also inhibits interferon dependent transcription. bICP0 can induce degradation of interferon stimulatory factor 3 (IRF3), a cellular transcription factor that is crucial for activating beta interferon (IFN-β) promoter activity. Recent studies also concluded that interactions between bICP0 and IRF7 inhibit trans-activation of IFN-β promoter activity. The issue of food safety is a growing concern to Irish agriculture and to the food industry. In the last twenty years, many novel pathogens, which can cause concern to the food industry, have been identified. Organisms such as Verocytotoxigenic Escherichia coli (VTEC) cause hemorrhagic colitis and Haemolytic Uremic Syndrome (HUS), which is the major cause of acute renal failure in children in many countries, and it’s presence in food is a cause of growing concern. The prevalence, intensity, and pathology of a Myxobolus sp. affecting the kidneys of cultured Diplodus puntazzo C. during the period 2003-2005 are described. The study included data collected from the largest farm in southern Greece where a high rate of mortality due to myxosporean infection occurred. Sharp snout seabream reared in sea cages in southern Greece underwent microbiological, parasitological, and histological examinations, while measurement and identification of parasites was performed immediately after sampling. Experiments were conducted to compare the efficacy of montmorillonite (M) and montmorillonite nanocomposite (MN) to reduce the toxicity of aflatoxin (AF) in broiler chicks from 0 to 42 days of age. A total of 240 chicks were assigned to 6 dietary treatment groups (0 g of adsorbents and 0 mg of AF/kg feed, 3 g of M/kg feed, 3 g of MN/kg feed, 0.11 mg of AF/kg feed, 0.11 mg of AF plus 3 g of M/kg feed, and 0.11 mg of AF plus 3 g of MN/kg feed). Two stocking densities (20 and 40 fry/80-l tank) with 4 feeding rates (2.5%, 5.0%, 7.5%, and 10.0% body weight/day) were used to determine the optimum feeding level for himri barbel fry during a 60-day culture period. Water quality parameters (temperature, dissolved oxygen, pH, and total ammonia nitrogen) were monitored during the experiment. This study was conducted to determine some properties of set and stirred type yoghurts containing 0.5% and 1% garlic (Allium sativum L.) during storage period of 28 days at 4±1 °C. Some microbiological, physical, chemical, and sensory properties of yoghurts were determined in days 1, 7, 14, 21, and 28 of the storage period. Coliform bacteria were not detected in all samples during the storage period ( 0.05) and sensory scores of yoghurts decreased during the storage period.
