Parasitological studies of archaeological sites can be used to interpret past behavior and living conditions. During the 1980s problem-oriented research into prehistoric- and historical-period parasitism developed and resulted in thefield of archaeoparasitology. Archaeoparasitology attempts to integrate parasite data into archaeological theory and interpretation. Within the last decade, four major archaeoparasitological laboratories emerged. They developed interpretive frameworks that apply parasitological data to a remarkable variety of prehistoric behaviors. Parasite remains can be used to reconstruct aspects of diet, health, and other behaviors such as transhumance and trade. Finally. analysis of the distribution of parasite remains can be used to interpret aspects …

The most remarkable dietary remains recoverable from archaeological contexts are coprolites. Coprolites are desiccated or mineralized feces that are preserved in sheltered and open sites in arid regions, primarily in the New World. These dietary remains are remarkable from several perspectives. They typically contain a variety of macroscopic and microscopic remains that form interrelated data sets for the reconstruction of diets. Because contexts containing coprolites are typified by excellent preservation, the remains coprolites contain tend to be in better states of preservation than dietary remains recovered from nonfecal deposits. Vertebral pathology has long been a useful criterion for anthropologists in the assessment of activity patterns, stress, and general health of extinct peoples. This method of analysis, however, has never been applied to the peoples of the Nebraska Great Plains. This study is the first to concentrate on the indigenous Native Americans of this region, examining the spinal pathology present in the prehistoric and historic skeletal remains. Pathology present in the form of spondylolysis, Schmorl's nodes, osteophytosis (degenerative disc disease), and osteoarthritis (degenerative joint disease), provides evidence to suggest differing activity patterns and levels of stress in Plains groups before …

The Dan Canyon burial was discovered at a time when the philosophy, ethics, and legislation concerning the study of human remains are in a state of flux. A number of important sensitive issues germane to managers, archeologists, and American Indians are discussed in the introduction. The subsequent analysis provides a detailed scientific account of these remains and a glimpse of a segment of a people's past lifeway while remaining sensitive to the wishes of the American Indians.

Nearly 2,400 plant species have been documented ethnographically as having medicinal value among Native Americans. Therefore, it is not surprising that evidence of medicinal plants appears in some of the over 1,000 coprolites analyzed from the southwestern U.S. Three medicinal species identified in pollen analysis of coprolites are discussed. Willow (Salix) is the most common analgesic in the Native American pharmacopoeia. Prehistoric use of this plant is documented in coprolites from Bighorn Cave in the Black Mountains of Arizona and also in a burial from the Mimbres Valley, New Mexico. During 1988 the number of Shigella dysenteriae type 1 infections reported in the United States increased fivefold. To determine if recent isolates from Mexico were related to those that caused epidemics of dysentery worldwide, Southern hybridization analysis was done with Shiga toxin and ribosomal RNA gene probes. Western hemisphere and Eastern Hemisphere strains differed by the size of a single EcoRI fragment carrying the Shiga toxin genes. Three ribosomal DNA (rDNA) patterns were observed, which correlated with the strain;s continental origin for 81 of 83 isolates tested. Measurement of the availability of health care providers in a geographic area is a useful component in assessing access to health care. One of the problems associated with the county provider-to-population ratio as a measure of availability is that patients frequently travel outside their counties of residence for health care, especially those residing in non-metropolitan counties. Thus, in measuring the number of providers per capita, it is important that the geographic unit of analysis be a health service area. We have defined health care service areas for the coterminous United States, based on 1988 Medicare data on travel patterns between …

Four coprolites were excavated with Burial 5 at Ventana Cave. a partially mummified five-year-old child. Two coprolites were granular and dark in color and two were fibrous and light in color. The coprolites are remains of the child's intestinal contents and were submitted for dietary and parasitological analysis. No parasites were found. The fibrous coprolites proved to be remains of highly masticated mesquite pods (Prosopis). The granular coprolites consist of seeds of saguaro cactus (Carnegiea gigantea). Pollen analysis reveals two dietary pollen types, both derived from cactus. No evidence of cultivated plants except for a trace amount of …

This research was designed to investigate whether there would be a difference in mean exhaled carbon monoxide levels and mean number of cigarettes smoked for two randomized groups of smokers.Smoking cessation therapy was manipulated in smokers with a chronic, productive cough (COPD) and those who were asymptomatic (normal). Individual therapy was compared to self-help therapy.The main purpose of this study was an attempt to define the smoking cessation therapy that worked best for each of these two groups.With the information gained from this research, it was hoped that educating different types of smokers (those with and without lung disease) about …

A rapid diagnostic method employing a polymerase chain reaction procedure (PCR) was used to identify Shigella and enteroinvasive Escherichia Coli. This procedure amplified a region of the invasive-associated locus (ial) from a crude DNA extract of feces. A synthetic 21-base oligonucleotide corresponding to the ial gene sequence was shown to specifically hybridize only with enteroinvasive E. Coli (EIEC) strains and Shigella species. Upon PCR amplification, a 320-base pair fragment was generated in DNA extracted from feces reconstituted with EIEC or Shigella flexneri but not in DNA from 70 normal stools lacking these organisms and could be readily …

Mortality surveys and death certificate studies have suggested an association between leukemia and farming. To investigate whether exposure to carcinogens in an agricultural setting is related to risk of leukemia, the authors conducted a population-based case-control inter view study of 578 white men with leukemia and 1245 controls living in Iowa and Minnesota. Consistent with recent mortality studies, there were slight, but significant, elevations in risk for all leukemia (odds ratio (OR) 1.2) and chronic lymphocytic leukemia (OR 1.4) for farmers compared to nonfarmers. The South Texas area, Region 3 of the Southwestern Division, U.S. Army Corps of Engineers, is synthesized from archeological and bioarcheological perspectives. Three distinct geographic units within Region 3 are treated in detail: Central Texas Plateau Prairie, South Texas Plains, and Lower Pecos Canyonlands. More than 11,000 years of human adaptation are chronicled for this area, stretching from the Gulf of Mexico to the Rio Grande along the border with northeastern Mexico. In 1987, 93 Escherichia coli 0157:H7 isolates were collected during routine surveillance for this pathogen in the state of Washington. Toxin genotypes and plasmid profiles were correlated with the clinical sequelae of illness in 88 of the 93 patients from whom these strains were isolated. Thirteen plasmid patterns were observed among the 88 tested isolates; four patterns accounted for 82(7o of the isolates. Genetic probing for Shiga-like toxins (SLT) I and II demonstrated the presence of both genes in 67 (76%), SLT I alone in three (3%), and SLT II alone in 18 (20%). The skeletal remains of an adult male associated with desiccated tissue and a coprolite were recovered from an apen-air midden deposit at the NAN Ranch Ruin (LA15049)) a large Mimbres site in Grant County) New Mexico. The find dates to about A.C. 1000-1100. Identifiable macroscopic elements in the caprolite consist offi nely fragmented corn and tiny seed fragments of an unknown plant. High amounts of willow (Salix) and mustard (Brassicaceae) pollen may indicate the ingestion of medicinal plants to combat a deteriorating health condition. One of the main problems encountered in the review of the bioarcheology of Region 3 has been the limited number of sites where human skeletal material has been adequately recovered and analyzed. In the preceding chapter it was documented that less than 30% of the burials recovered from recorded sites have been reported in published literature. It was further estimated that of the 323 sites with burials, no more than 80 sites have published detailed bioarcheological reports on the burials recovered. Only 50 of these 80 reports provide individual descriptions of each burial which facilitate subsequent analyses and evaluation.

The structural genes for Shiga toxin, designated stx A and stx B, were cloned from Shigella dysenteriae type 1 3818T, and a nucleotide sequence analysis was performed. Both stx A and stx B were present on a single transcriptional unit, with stx A preceding stx B. The molecular weight calculated for the processed A subunit was 32,225, while the molecular weight of the processed B subunit was 7,691. Comparison of the nucleotide sequences for Shiga toxin and Shiga-like toxin I (SLT-I) from Escherichia coli revealed that the genes for Shiga toxin and SLT-I were greater than 99% homologous; three nucleotide …

Examination of coprolites excavated from archaeological sites in the Americas demonstrates excellent preservation of helminth eggs and, in some cases, larvae. To gain an understanding of helminth parasitism in prehistory on the Colorado Plateau of Arizona, New Mexico, and Utah, 319 coprolites from 5 archaeological sites were analyzed. Helminth eggs and larvae were recovered after the coprolites were rehydrated, screened, and sedimented. At a sixth site, soils excavated from 5 rooms used as latrine areas were processed with palynological techniques. The results indicate that all but 1 of the prehistoric populations examined were infected with intestinal worms. The helminths implicated …

Escherichia coli 0157:H7 strain 933 contains two distinct toxin-converting phages (933J and 933W). The biologic activities and antigenic relationship between the toxins produced by 933J and 933W lysogens of E. coli K-12, as well as the homology of the genes that encode the two toxins, were examined in this study. The 933J and 933W toxins, like Shiga toxin produced by Shigella dysenteriae type 1, were cytotoxic for the same cell lines, caused paralysis and death in mice, and caused fluid accumulation in rabbit ileal segments. The cytotoxic activity of 933J toxin for HeLa cells was neutralized by anti-Shiga toxin, whereas …

Several helminth parasites have been recovered from archaeological sites on the Colorado Plateau. These include Ascaris, Trichuris, Enterobius, Moniliformis, and tapeworms in the families Hymenolepidae and Taeniidae. The recovery of these animals demonstrates that cross-temporal: comparison of parasitism between different prehistoric lifestyles is possible. Such comparison might elucidate the effects of changing lifestyle and subsistance pattern on human parasitism.

Three monoclonal antibodies, designated MAb 16E6, MAb 13C4, and MAb 19G8, were produced which recognize Shiga-like toxin (SLT) from Escherichia coli. All three monoclonal antibodies neutralized the cytotoxicity of E. coli SLT and were able to immunoprecipitate intact labeled toxin with Staphylococcus aureus protein A. The three antibodies were of the Gl heavy and kappa light chain classes. MAb 16E6 bound to the B subunit of SLT in Western blots and also neutralized the lethality of the toxin for mice and the enterotoxicity of the toxin in ligated rabbit ileal loops. Effect Of Membrane Filtration Of Antimalarial Drug Solutions On In Vitro Activity Against Plasmodium Falciparum, J. Kevin Baird, C. Lambros

Antimalarial activities ofchloroquine, mefloquine, amodiaquine, and quinine in vitro against Plasmodium falciparum were diminished as a consequence ofmembranefiltration. Filtered drug solutions gave ID50 values up to 25-fold greater than those of non-filtered (ethanol-sterilized) drug solutions. Loss ofactivity byfiltration was overcome by increasing the drug concentration prior to filtration. Water solutions filtered through Millex-GSfilter units consistently showed an absorbance maximum at 277 nm, accompanied by a lesser peak at 225 nm. Water filtrates from Nucleopore and Millex-G Vfilters showed no absorbance at 277 nm and only slight absorbance was evidentfor the Gelmanfilter unit. The composition of phosphate metabolites and the intracellular pH in erythrocytes from a patient with hereditary pyrimidine-5'-nucleotidase deficiency were examined using 31P NMR spectroscopy. Several resonances were identified in spectra from intact cells and from extracts. The 2,3-bisphosphoglycerate line intensities were normal but the NTP resonances were about twice normal due to the presence of millimolar quantities of pyrimidine phosphates. Several intense resonances were also observed in the diphosphodiester region of the spectrum. One compound contributing to these lines has been identified as cytidine diphosphocholine. Assessing the microbiological water quality of impoundments and the potential for waterborne disease outbreaks is a difficult task when using traditional sampling programs. Problems associated with using fecal coliform bacteria as indicators of human pathogen presence complicates assessments of future water quality in preimpoundment areas. Reliable determination of future and present microbiological water quality requires knowledge of how the chemical, physical, and biological characteristics of the watershed and impoundment interrelate to influence microbial indicator and pathogen densities. Accurate estimates of microbial indicator and pathogen densities, obtainable by using the enumeration methods and their modifications suggested in this report, will allow …

Multi-individual cremation deposits found in the upper Santa Cruz River drainage were previously interpreted as the result of hypothesized cultural contact in the area. A review of 50 cremation deposits for which detailed analyses are available indicates that mulii-individual cremation deposits are the result of incomplete gleaning practices and have doubtful cultural significance. Hypotheses are generated from the present data which account for the apparent variation in cremation practices within the study area.
