The purpose of this project was to study the antibacterial effect of the short exposure of 5.25% NaOCl and 17% EDTA on eight strains of Enterococcus faecalis in extracted human teeth. The crowns of 240 single rooted teeth were removed and the canals were instrumented to ISO 0.36. The canals were irrigated with 5.25% NaOCl and 17% EDTA, autoclaved and separated into eight groups of 30 teeth consisting of 20 experimental teeth, 5 positive and 5 negative controls. Each group was inoculated with one of eight different strains of E. faecalis in BHl broth and incubated for four weeks. The …

The purpose of this investigation was to determine the antimicrobial effect of Biopure MTAD (a mixture of a tetracycline isomer, an acid, and a detergent) as a final irrigant on eight strains of Enterococcus faecalis (E. faecalis) in contaminated root canals of extracted human teeth. In addition, the Minimum Inhibitory Concentration (MIC) and Minimum Lethal Concentration (MLC) of MTAD were also measured on the same strains of bacteria. Two-hundred and forty extracted human teeth were instrumented using 1.3% NaOCl followed by 17% EDTA to remove the smear layer. The teeth were randomly contaminated with one of eight strains of E. …

While previous studies have shown that presence of bacteria in systemic circulation can cause infective endocarditis, there is no information on the specific amount of bacteria necessary to cause this condition. The purpose of this study was to establish the minimum level of circulating bacteria that will cause colonization of damaged heart valves in rats. Fifty-two Sprague-Dawley rats were anesthetized and their heart valves were damaged using an established protocol. A clinical isolate of Enterococcus faecalis was transformed with a plasmid bearing the luxF and Chloramphenicol cassettes to label the bacteria with the light-emitting protein. Bacteria have been shown to play an important role in the development of pulp and periradicular disease. The source of the infective bacteria is most often the oral cavity. Previous studies have detected bacteria in teeth that are structurally intact that seemingly have no communication intraorally. Anachoresis has been proposed as a mechanism for localizing circulating bacteria to such teeth. In several previous bacteriologic anachoresis studies, bacteria have been found to localize in inflamed dental tissues. The criticism most commonly associated with these investigations has been the potential contribution of contaminant bacteria resulting in false positive findings.

A possible cause for root canal failure is the persistence of bacteria that have colonized dentinal tubules. To reduce this risk and, if possible, to also shorten the time consuming endodontic therapy, new equipment and materials are constantly being introduced. The aim of the present investigation was to study the effect of disinfection of dentinal tubules by intracanal laser irradiation using an in vitro model. The groups irradiated by various lasers were compared with calcium hydroxide, a material frequently used as intracanal medication between appointments. The pathogenesis of periapical lesions is the interaction of the oral flora with the existing host defenses. The role of bacteria has been established in endodontic disease but the contribution of the host defenses in periapical lesion formation is less clear. The purpose of this study was to compare periapical lesion development in immunosuppressed rats with that in normal rats. Thirty-five, eight-week old Sprague-Dawley rats were equally divided into seven groups. Groups 1, 2, and 3 were given weekly injections of Cytoxan, an immunosuppressive agent. The 5-lipoxygenase metabolites of arachidonic acid such as leukotrienes (LT) B4 and C4, are potent mediators of inflammation. The purpose of this investigation was to determine the concentrations of LTB4 and LTC4 in human periradicular lesions. Specimens from asymptomatic and symptomatic patients were obtained and immediately frozen in liquid nitrogen. Periradicular tissues from unerupted third molars as well as chronically inflamed gingival tissues were also obtained, frozen and were used as negative and positive controls respectively. Endodontic (root canal) infections are generally considered to be caused by the invasion of microorganisms which destroy pulp tissue and extend to the periapical region. No definitive study has identified a specific bacterium or group of bacteria as etiological agents. We quantitively measured antibodies present in the serum of 30 normal individuals and patients with either chronic (n = 30) or acute (n = 30) periapical lesions with the indirect fluorescent antibody (IFA) and enzyme-linked immunosorbent assay (ELISA) for IgG and IgM against 11 oral microorganisms routinely isolated from such lesions. The Lasting Antimicrobial Qualities Of Three Root Canal Sealing Materials : Grossman's Sealer, Rickert's Sealer, And N2 Permanent, Lee R. Ingersoll

An in vitro study using polyethylene tubes filled with Grossman's sealer, Rickert's sealer, and N2 permanent was conducted over a six month period in an effort to determine their lasting antimicrobial properties against eight different microorganisms.

From the study it was observed that N2 permanent possesses a long-lasting antimicrobial effect, but this effect appears to diminish greatly with time. Further investigation is needed to determine whether or not the effect is actually permanent in nature. The N2 permanent was also shown to have broader antimicrobial effectiveness against the eight microorganisms tested. N2 permanent produced zones of inhibition against five of …
