New strategies for surface modification have been explored to promote osseointegration and osteogenesis, which are crucial factors for the success of titanium dental implants. Coating the titanium surface with mRNA encoding therapeutic growth factors may achieve these objectives. In our pilot study, we employed green fluorescent protein (GFP) mRNA as a model. Our investigation focused on determining whether titanium disks coated with GFP mRNA encapsulated with lipid nanoparticles and collagen Type I (GFP mRNA-Col) could enhance transfection in human bone marrow mesenchymal stem cells (hBMSCs), in comparison to titanium disks coated with GFP mRNA alone, without collagen. The objective of this study was to investigate the effect of local inflammation on platelet derived growth factor-BB (PDGF-BB) expression after gingival injection of PDGF-BB mRNA in lipid nanoparticles (LNPs) in ligature-induced rat periodontitis and to examine the dose-dependent effect of gingival delivery of mRNA. 3-0 silk was placed around maxillary left second molar for two weeks and removed, while the corresponding molar on the right was non-ligated. Nucleoside-modified messenger RNA (mRNA) technology can be a promising novel platform in regenerative medicine. The aims of this study were to evaluate whether pseudouridine modified mRNA encoding vascular endothelial growth factor (VEGF) could induce VEGF production in human periodontal ligament cells (PDLCs) and this translated protein function by promoting in vivo blood vessel formation using chorioallantoic membrane (CAM) assay. MSCs exert their immunomodulatory effects on various immune cells by cell-cell contact and cytokines secretion. Our previous study has demonstrated that supracrestal gingival connective tissue-derived mesenchymal stem cells (SG-MSCs) were recognized to be a good candidate for periodontal regeneration. Objective: To investigate and compare the characteristics of human mesenchymal stem cells (MSCs) among 3 different stem cell sources in periodontium; periodontal ligament (PDL) tissue, marginal gingival tissue (MG), and supra-crestal gingival connective tissue (SG). Materials and Methods: PDL-MSCs, MG-MSCs, and SG-MSCs were obtained from 4 subjects with healthy periodontal condition. The characteristics of 3 types of MSCs were compared in terms of an expression of MSCs markers, colony-forming ability (CFA), cell proliferation, alkaline phosphatase (ALP) activity, osteogenic and adipogenic potential. Previous studies suggest that periodontitis may be associated with an increasing risk of hypertension. However, data from large epidemiological studies are limited. The aim of this cross-sectional study was to examine the association between periodontitis and hypertension using data from a cohort study in a Thai population. The study population comprised of 1,378 employees of the Electricity Generating Authority of Thailand (EGAT), aged 53 to 73 years old, were categorized into no/mild, moderate, or severe periodontitis groups. Despite the important role of tissue - resident memory T (TRM) cells in immunity and pathology of mucosal tissues, the study of tissue-resident memory T cells in periodontitis tissues is limited. Previous investigation in our laboratory identified the localization of TRM (CD103+ T cells) in periodontitis tissue. However, their function is not known.
