Vulvovaginal candidiasis (VVC), caused by is characterized by aberrant inflammation by polymorphonuclear neutrophils (PMNs) in the vaginal lumen. Data from the established murine model shows that despite potent antifungal properties, PMNs fail to clear due to local heparan sulfate that inhibits the interaction between PMNs and , resulting in chronic vaginal immunopathology. To understand the role of neutrophil extracellular traps (NETs) in defense against at the vaginal mucosa, we investigated the NET-forming capacity of PMNs in chronic VVC-susceptible (CVVC-S/C3H) and -resistant (CVVC-R/CD-1) mouse strains. Immunofluorescence revealed the formation of NETs (release of DNA with PMN-derived antimicrobial proteins) in PMN cocultures …

Recruitment of polymorphonuclear neutrophils (PMNs) into the vaginal lumen is the hallmark of an acute immunopathologic inflammatory response during vulvovaginal candidiasis (VVC) caused by Candida albicans. Recurrent VVC (RVVC) remains a chronic health burden in affected women worldwide despite the use of antifungal therapy. Based on the role leukotrienes (LTs) play in promoting inflammation, leukotriene receptor antagonists (LTRAs) targeted for LTB4 (etalocib) or LTC4, LTD4, and LTE4 (zafirlukast or montelukast) have been shown to reduce inflammation of epithelial tissues. An open-label pilot study using long-term regimens of zafirlukast in women with RVVC indicated the potential for some relief from recurrent …

Introduction: The causative factors of dental caries have been studied for many years, however, preventing the formation of dental caries remains an elusive task. Microorganisms have been the focal point of numerous studies as a prime cariogenic factor. Streptococcus mutans and Candida albicans have been frequently co-isolated from dental biofilm in patients with early childhood caries (ECC) and studies have demonstrated the synergistic effects of these two microorganisms in promoting tooth demineralization. Acid and glucan production, which are two virulence factors produced by S. mutans that mediate biofilm formation and tooth decay, have been shown to be promoted during co-colonization …

Mass Spectrometric Analyses Of Surface Proteins From Candida Albicans After Treatment With Histatin 5, Shirley Fan

Electronic Thesis and Dissertation Repository

Current drug therapies have lost their effectiveness in controlling the fungal species known as C. albicans; therefore, the need for newer drugs becomes crucial. Histatin 5 (Hst 5), a naturally occurring peptide found in the oral cavity, has demonstrated strong effects on controlling candidiasis. Here, we have proposed that the adhesion of cells through surface proteins is an important process. To allow the analysis of surface proteins, the concept of on-whole cell tryptic digestions were developed. This on-target approach allows for isolation of the peptides of interest, while greatly reducing digestion times.

To study the effects of Histatin 5 …

Candida albicans is an opportunistic pathogen that is present in the normal flora in a majority of individuals. One key factor in C. albicans virulence is the ability to change its morphology from yeast to an elongated or hyphal form. The regulation of this morphogenesis relies in part upon quorum sensing (QS) molecules. C. albicans often exists as part of a mixed culture alongside other microbes and is influenced by their presence as well as the presence of QS molecules that they produce. In this study, a library of diploid homozygous transcriptional regulator knockout (TRKO) mutants were screened to identify …

The oral cavity harbors more than 700 microbial species and is one of the most complex ecosystems ever described. While the majority of these microbes are considered commensal, some of them are responsible for oral infectious diseases such as dental caries, periodontitis, halitosis and stomatitis. The advancement of modern science has greatly furthered our understanding of oral microbes and their roles in host health and disease. It has also led to the development of new tools for early detection, effective treatment, and prevention of oral microbial infections. This perspective provides a general understanding of oral microbiology, and its clinical relationship …
