The major current focus on treating rheumatoid arthritis is to put an end to long-term treatments and instead, specifically block widespread immunosuppression by developing antigen-specific tolerance, while also permitting an intact immune response toward other antigens to occur. There have been promising preclinical findings regarding adoptive Treg cells immunotherapy with a critically responsible function in the prevention of autoimmunity, tissue repair and regeneration, which make them an attractive candidate to develop effective therapeutic approaches to achieve this interesting concept in many human immune-mediated diseases, such as rheumatoid arthritis. Ex vivo or invivo manipulation protocols are not only utilized to correct …

Treatment Options For Obstructive Sleep Apnea, Shao-Chih Hsieh, Szu-Ting Chou, Yu-Chuan Tseng

Taiwanese Journal of Orthodontics

Obstructive sleep apnea (OSA) is sleep-disordered breathing caused by the collapse of the upper airway. Because anatomical abnormalities of the narrow upper airway are undoubtedly the most critical cause of OSA, the available treatment methods for OSA mostly focus on these anatomical problems. The collapse of the respiratory tract is associated with different levels of upper airway obstruction. The obstruction of the airway can be divided into three parts: the nasal cavity, retropalatal region, and retrolingual region. This article summarizes some common OSA treatment methods, their underlying theories, and their effect on the upper airway.

Mir-365 (Microrna): Potnetial Biomarker In Oral Squamous Cell Carcinoma Exosomes And Extracellular Vesicles, Jeffery Coon, Karl Kingsley, Katherine M. Howard

Dental Medicine Faculty Research

Introduction: miR-365 is a non-coding microRNA that regulates transcription and has been demonstrated to promote oncogenesis and metastasis in some cancers, while suppressing these effects in others. Many microRNAs are produced and then exported extracellularly in exosomes, which are small extracellular vesicles ranging from 30 to 100 nm that are found in eukaryotic fluids and facilitate many cellular functions. Exosomes and extracellular vesicles are produced by many cell types, including oral cancer cells—although no study to date has evaluated miR-365 and oral cancer exosomes or extracellular vesicles. Based on this information, our research question was to evaluate whether oral cancers …

Cleft palate (CP) is the second most common congenital birth defect. The etiology of CP is complicated, with involvement of various genetic and environmental factors. To investigate the gene regulatory mechanisms, we designed a powerful regulatory analytical approach to identify the conserved regulatory networks in humans and mice, from which we identified critical microRNAs (miRNAs), target genes and regulatory motifs (miRNA-TF-gene) related to CP. Using our manually curated genes and miRNAs with evidence in CP in humans and mice, we constructed miRNA and transcription factor (TF) co-regulation networks for both humans and mice. A consensus regulatory loop (miR17/miR20a-FOXE1-PDGFRA) and eight …

Plant And Fungal Products That Extend Lifespan In Caenorhabditis Elegans, Jan Martel, Cheng Yeu Wu, Hsin Hsin Peng, Yun Fei Ko, Hung Chi Yang, John D. Young, David M. Ojcius

All Dugoni School of Dentistry Faculty Articles

The nematode Caenorhabditis elegans is a useful model to study aging due to its short lifespan, ease of manipulation, and available genetic tools. Several molecules and extracts derived from plants and fungi extend the lifespan of C. elegans by modulating aging-related pathways that are conserved in more complex organisms. Modulation of aging pathways leads to activation of autophagy, mitochondrial biogenesis and expression of antioxidant and detoxifying enzymes in a manner similar to caloric restriction. Low and moderate concentrations of plant and fungal molecules usually extend lifespan, while high concentrations are detrimental, consistent with a lifespan-modulating mechanism involving hormesis. We review …

Antibacterial Efficacy Of Novel Eastern Medicine-Inspired Toothpastes Compared To Commercial Formulations, Joshua S. Goldfaden Jul 2020

Antibacterial Efficacy Of Novel Eastern Medicine-Inspired Toothpastes Compared To Commercial Formulations, Joshua S. Goldfaden

Pacific Journal of Health

Toothpaste is an oral health care agent that dates back to the 5th Century B.C. in East Asia. Following the conception of dentistry in the 7th Century B.C. and the field’s subsequent growth, the embodiment of toothpaste has evolved from containing natural substances such as tea leaves to abrasive chemicals, particularly: fluoride, triclosan, and sodium lauryl sulfate (SLS). Regarding how each respective reagent functions, fluoride forms a complex with calcium ions in the teeth to prevent erosion of the protective enamel coating. This ion also disrupts metabolic activities of Streptococcus mutans (S. mutans), which are the …

Activation of the nod-like receptor 3 (NLRP3) inflammasomes is crucial for immune defense, but improper and excessive activation causes inflammatory diseases. We previously reported that Cbl plays a pivotal role in suppressing NLRP3 inflammasome activation by inhibiting Pyk2-mediated apoptosis-associated speck-like protein containing a CARD (ASC) oligomerization. Here, we showed that Cbl dampened NLRP3 inflammasome activation by inhibiting glycolysis, as demonstrated with Cbl knockout cells and treatment with the Cbl inhibitor hydrocotarnine. We revealed that the inhibition of Cbl promoted caspase-1 cleavage and interleukin (IL)-1β secretion through a glycolysis-dependent mechanism. Inhibiting Cbl increased cellular glucose uptake, glycolytic capacity, and mitochondrial oxidative …
