Introduction:. During the course of orthodontic treatment, enamel decalcification is a common problem. Demineralized enamel, the precursor to caries formation, can be attributed to fixed orthodontic appliances and prolonged exposure to bacterial plaque. Progression to clinically detectable white spot lesions may occur as early as one month following the placement of orthodontic appliances. Such problems have influenced clinicians to search for a solution. Streptococcus mutans (Sm) and Streptococcus sobrinus (Ss) , are considered the major bacterium associated with dental caries. However, recent findings identify Scardovia species (Sc) is associated with early childhood caries, with or without the presence of Sm or Ss. This IRB approved study quantified the presence of Sm and Sc in saliva by real-time quantitative PCR (qPCR) in relation to caries. Saliva samples were collected from 72 preschool children in a high-caries risk community in rural Alabama. Runx2 and its downstream target Specificity Protein 7 (Sp7) are essential for skeletogenesis. In humans, mutations in Sp7 gene are associated with osteogenesis imperfecta. Unlike Runx2, the role of Sp7 in palatogenesis, tooth development or differentiation of ameloblasts and odontoblasts remains unknown. The main goal of this research was to identify the functional requirement of Sp7 transcription factor during tooth development and palatogenesis. Molecular, biochemical, and histological approaches were employed to answer this question. We established Sp7-null mice that are completely void of mineralized tissue, exhibit craniofacial dysmorphogenesis and die shortly after birth. Objectives: The specific aim of this study was to analyze any entrapped debris in implant abutment screw threads from post processed constructs from dental laboratories. Materials and Methods: Forty implant screws were used to collect debris where screws were removed from the implant prosthesis as received from dental laboratories. In all samples, laboratory procedures including fabrication of the custom abutment, definitive prosthesis and finishing/polishing were completed prior to sampling. Monolithic zirconia restorations are popular but removing these crowns is difficult. The purpose of the study is to measure and compare the cutting effectiveness of 10 types of diamond burs through monolithic sintered zirconia. Ten types of diamond burs (n=8) were compared for the cutting effectiveness in Zirconia (CERCON; DeguDent) using a high speed computer controlled cutting device with water spray (5.45L/min). The purpose of this study is to compare the facial morphology of African-American children to Welsh children using three-dimensional imaging. Materials and Methods: Three-dimensional images were captured using a stereo-photogrammatic camera system (3dMDfaceTM). 107 African-American children were imaged with the 3dMDfaceTM system and were compared with 64 Welsh children which were previously imaged using two Minolta Vivid VI900 3D laser scanners arranged in a stereo pair. Custom made abutments are utilized to correct for non-ideal implant positions and can successfully accommodate the morphologic requirements of the overlying crown. Previous studies have demonstrated the essential role of miRNA in many biological processes, including skeletal development and tooth formation. Dlx3, a homeobox gene, is critical for craniofacial patterning and postnatal skeletal development. The purpose of this study is to compare the facial morphologies of an adult African-American population to an adult Caucasian-American population using three-dimensional surface imaging. Materials and Methods: Three-dimensional facial images were captured via a stereo-photogrammetric camera system (3dMDfaceTM system). Pleiotrophin (PTN) is an extracellular matrix-associated growth factor and chemokine expressed in mesodermal and ectodermal cells. It plays an important role in osteoblast recruitment and differentiation. PTN has two well-known physiological receptors, protein tyrosine phosphatase beta/zeta (RPTPRZ1), and N-syndecan. Protein tyrosine phosphatases are a group of enzymes that remove phosphate groups from phosphorylated tyrosine residues on proteins. Real-Time Quantitative Pcr For Evaluation Of Mutans Streptococci And Dental Caries, Chanika Manmontri

Mutans streptococci (MS), e.g., Streptococcus mutans (Sm) and Streptococcus sobrinus (Ss), are associated with dental caries. Quantification of MS has been shown to predict caries risk. A new technology using PCR may streamline quantification compared to traditional culture methods. Purpose: To use real-time quantitative PCR (qPCR) and standard plate counting (SPC) for detection and quantification of levels of Sm, Ss and total streptococci (TS) in clinical samples from high-caries-risk children with no regular access to dental care and to study correlation of these bacterial levels and ratios to caries prevalence.
