Benzofuran Derivatives Substitute For The Discriminative Stimulus Effects Of 3,4methylenedioxymethamphetamine (Mdma) In Male Sprague-Dawley Rats, Candace Johnson

Masters Theses

4-Methylenedioxymethamphetamine (MDMA) is currently under evaluation in phase III clinical trials as medication-assisted therapy for post-traumatic stress disorder (PTSD) and is expected to be approved by the FDA for clinical use in the near future. MDMA is also a popular abused substance with risks for cardiovascular toxicity and neurotoxicity, particularly when misused at higher doses. Characterization of the behavioral and neurochemical effects of novel psychoactive substances is an essential step in the development of safer alternative therapeutic agents. Drug discrimination is a preclinical behavioral assay with pharmacological specificity for characterizing in vivo drug actions in the central nervous system. This …

Organocatalyzed Synthesis Of Epoxides From Chalcones Utilizing Amino Acids, Sabrina N. Kegeler Apr 2018

Organocatalyzed Synthesis Of Epoxides From Chalcones Utilizing Amino Acids, Sabrina N. Kegeler

Masters Theses

The epoxide functional group is important throughout the chemical and pharmaceutical industries, as well as in nature. In the chemical industry, epoxides are present in resins and fragrances. In the pharmaceutical industry, epoxide-containing compounds are used as intermediates in the manufacturing of drugs. In nature, many natural products contain epoxide groups and are used for medicinal purposes, and for models to create synthetic molecules.

One approach to epoxide synthesis involves the use of an alkene precursor, a base, and an oxidizing agent. This is where my investigations began. The first step was to optimize the epoxidation reaction, examining substrate scope, …

The Oncolytic Potential Of Tanapoxvirus To Treat Retinoblastoma, John D. Christie Dec 2016

The Oncolytic Potential Of Tanapoxvirus To Treat Retinoblastoma, John D. Christie

Masters Theses

Retinoblastoma is the leading cause of intraocular malignancy in children under 2 years of age. Current treatments, while generally effective in eliminating the tumor, have many negative side effects. Oncolytic virotherapy is the use of either wild type or recombinant viruses, to destroy tumor cells. There are currently several viruses being engineered as potential therapeutics for a wide range of cancers. They offer tumor specific alternative to current therapies with lesser toxicity. This study looked at the oncolytic efficacy of multiple tanapoxviruses (TPV) recombinants in an in vitro retinoblastoma. Here we have tested recombinants of TPV for their ability to …

Behavioral Sensitization Following Concurrent Exposure To 4-Methylmethcathinone (4-Mmc) And 3,4-Methylenedioxymethamphetamine (Mdma) In Male Sprague-Dawley Rats, Trent Bullock Apr 2016

Behavioral Sensitization Following Concurrent Exposure To 4-Methylmethcathinone (4-Mmc) And 3,4-Methylenedioxymethamphetamine (Mdma) In Male Sprague-Dawley Rats, Trent Bullock

Honors Theses

Recreational use of a new class of stimulant drugs known as synthetic cathinones is a recent public health concern. Although the Drug Enforcement Administration placed several of the most common of these substances permanently on schedule 1, their use is still prevalent as they remain low cost, accessible, and potent. Concomitant use of cathinone derivatives with other psychostimulant drugs is commonly reported by recreational users. Despite the prevalence of synthetic cathinone abuse, there is currently a paucity of scientific research regarding the behavioral and neurochemical effects of these drugs in mixtures with other drugs of abuse. The behavioral sensitization paradigm …

The Effects Of Aging And Estrogen Treatment During Aging On The Rat Epididymis, Kathryn Davis Apr 2016

The Effects Of Aging And Estrogen Treatment During Aging On The Rat Epididymis, Kathryn Davis

Honors Theses

Previous studies from our lab have demonstrated that a normal estrogen environment in the testis contributes to maintaining spermatogenesis in adult rats and that estrogen treatment can attenuate the age-associated decline in sperm production. The purpose of this study was to determine if the estrogen environment of the epididymis is altered with age and what effects estrogen treatment may have on the epididymis during aging. The study compared untreated rats at 15 months of age to 18 month old rats that were treated with either vehicle, or estradiol- valerate, once every third day from 15 to 18 months. Mean concentrations …

Evolutionary Convergence Of The Caffeine Biosynthetic Pathway In Chocolate Followed Duplication Of A Constrained Ancestral Enzyme, Andrew J. O'Donnell Jun 2015

Evolutionary Convergence Of The Caffeine Biosynthetic Pathway In Chocolate Followed Duplication Of A Constrained Ancestral Enzyme, Andrew J. O'Donnell

Masters Theses

Caffeine biosynthesis is widely distributed in flowering plants and requires three consecutive methylation steps of xanthine alkaloids. Genes that have previously been reported to participate in the multi-step pathway in Coffea sp. (coffee) and Camellia sinensis (tea) encode members of the SABATH family of methyltransferases. Two genes highly expressed in fruits of Theobroma cacao (cacao) are orthologous to the caffeine genes in tea and appear to have diversified following gene duplication. Biochemical characterization of the enzymes (XMTs) encoded by these genes strongly suggest an unprecedented major pathway to theobromine, a precursor to caffeine. These findings imply that caffeine biosynthesis evolved …

The Effects Of Adderall On Maternal And Fetal Health, Alyson Brady Apr 2015

The Effects Of Adderall On Maternal And Fetal Health, Alyson Brady

Honors Theses

Information available for woman prescribed Adderall to treat ADHD and the effects that it may have on her fetus is limited. However, Adderall and methamphetamine have similar pharmacological and addictive qualities. This paper is concerned with whether there is an adequate approach to treatment that balances the needs of the mother and the fetus. With Adderall use becoming more prevalent, it is essential that society is aware of the physiology and pharmacology of this drug, as well as, the need for more research on this subject to be produced.

Stability Of Eukaryotic Elongation Factor One Alpha Protein In The Cell, Jennifer Watry Apr 2007

Stability Of Eukaryotic Elongation Factor One Alpha Protein In The Cell, Jennifer Watry

Honors Theses

Eukaryotic Elongation Factor One Alpha (eEF1A) is an abundant protein found in every cell of every organism and is involved in functions concerning protein synthesis and degradation, the cytoskeleton, and signaling. How these functions are regulated is not yet understood for this protein. Regulation of the protein may be controlled by transcription of the genes encoding the protein or the stability and modification of the protein. The goal of this project was to determine the stability of eEf1A proteins in Arabidopsis thaliana. Protein stability was determined by treating tissues with cycloheximide, an inhibitor of protein synthesis. Gel electrophoresis and …

Effects Of Zinc On Glucose-6-Phosphate Dehydrogenase In Buffalo Rat Liver Cells, Bryan G. Sauer Apr 1998

Effects Of Zinc On Glucose-6-Phosphate Dehydrogenase In Buffalo Rat Liver Cells, Bryan G. Sauer

Honors Theses

In recent years, interest has surfaced in the use of insulin-mimicking agents for diabetic therapies. Zinc has been shown to have insulin-like effects in vivo and in vitro but has never been studied for its ability to mimic insulin in the regulation of carbohydrate and fatty acid metabolism. Therefore, the author studied the effects of zinc on glucose-6-phosphate dehydrogenase (G6PDH), an enzyme stimulated by insulin that is imperative in both carbohydrate and fatty acid metabolism. Because G6PDH is highly regulated in liver cells, a stable rat liver cell line, Buffalo Rat Liver (BRL), was used. Endogenous G6PDH activity and G6PDH …

A Clinical Trials Evaluation Of A Double-Blinded Protocol To Assess The Therapeutic Effectiveness Of Stimulant Medication Prescribed For Children Diagnosed Adhd, Ann V. Thompson

Dissertations

This clinical trials investigation developed and evaluated a protocol for determining the therapeutic effectiveness of medication prescribed for children diagnosed ADHD. The protocol included two separate baseline periods of two week durations, each of which were followed by four randomly assigned probes of one week duration, consisting either of Methylphenidate or placebo capsules. Baseline probes were unblinded, but medication and placebo probe trials were double-blinded. Therapeutic effect, expectancies, and integrity of the double-blind control were assessed through a battery of rating scales and forms completed by the parent, teacher and child at the end of each protocol week, including baseline …

Sds Non-Acrylamide Polymeric Gel-Filled Capillary Electrophoresis For Molecular Size Separation Of Protein, Devon Andres Aug 1993

Sds Non-Acrylamide Polymeric Gel-Filled Capillary Electrophoresis For Molecular Size Separation Of Protein, Devon Andres

Honors Theses

Sodium dodecyl sulfide (SDS) non-acrylamide gel-filled capillary columns are a new technology being used for analysis and separation of biotechnology-derived proteins. This research was to compare this new technology to the current methods of SDS polyacrylamide gel electrophoresis (SDS-PAGE) and high-performance size-exclusion chromatography (HPSEC). The molecular mass of four different recombinant proteins were determined by two commercialized SDS non-acrylamide gel-filled capillary columns, SDS-PAGE, and HPSEC. The data obtained showed that the SDS non-acrylamide gel-filled capillary columns were compatible with the SDS-PAGE technique for molecular mass determination. HPSEC was shown to be unreliable for molecular weight determination. SDS non-acrylamide gel-filled capillary …

The Effects Of Strain Differences On The Binding Of Concavalian A To Mouse Sperm, Lisa A. Mckay Jun 1986

The Effects Of Strain Differences On The Binding Of Concavalian A To Mouse Sperm, Lisa A. Mckay

Honors Theses

The current study, lectins were used as genetic probes to determine the phenotype of the sperm from different strains of mice. Lectins are proteins of non-immune origin, which bind to specific carbohydrates (McCoy, 1986). Being that virtually all cell membranes contain carbohydrate components, lectins have been used to identify all types of cells. In this study, a biotinylated lectin Concavalian A (Con A) derived from Canavalia ensiformis, was used and binding was detected using an avidin-biotin peroxidase (ABP) complex technique. The object of the current research was to 1) determine if mouse strain differences have an effect on lectin binding, …

The Survival Of Mesophilic And Thermophilic Bacteria Subjected To Uv Irradiation, Sajida A. Shaikh Aug 1985

The Survival Of Mesophilic And Thermophilic Bacteria Subjected To Uv Irradiation, Sajida A. Shaikh

Masters Theses

Survival curves were determined for the mesophile, Bacillus licheniformis and the thermophile, Bacillus stearothermophilus after subjecting the cells to ultraviolet irradiation. Cells were grown at 37°C for the mesophile and at 55°C for the thermophile.

At low UV exposure times, the thermophile was more resistant to UV irradiation (had a better survival rate) than was the case for the mesophile. At higher exposure times, the situation was reversed, the mesophile having a greater survival rate than the thermophile.

The effect of changes in the temperature during irradiation was also determined. Thermophiles were greatly affected by changes in temperature while the …

A Quantitative Study Of The Distinctive Proteins Of The Eosinophil: A Comparison Of Normals And Eosinophilic Patients, James M. Olson Dec 1984

A Quantitative Study Of The Distinctive Proteins Of The Eosinophil: A Comparison Of Normals And Eosinophilic Patients, James M. Olson

Honors Theses

Eosinophils have long been associated with parasitic and allergic diseases. Methods of procurement and purification of eosinophils in the last ten years have allowed intensive investigation into their components, properties, functions, and mechanisms (Gleich et al, 1982). The goals of this project are to separate eosinophils in blood samples taken from patients with eosinophilia and from normals based on density over distontinuous Metrizamide gradients; to determine quantitatively the levels of the three distinctive proteins of the eosinophil, specifically MBP, EDN, and ECP; and to compare the levels of these proteins found in eosinophils of patients with eosinophilia to the amounts …

A Comparative Consideration Of Lipoprotein Distribution And Stress Response In The Mallard Duck And Man, Janice Mackay Apr 1972

A Comparative Consideration Of Lipoprotein Distribution And Stress Response In The Mallard Duck And Man, Janice Mackay

Honors Theses

Lipid is associated with all of the major serum protein fractions of both the mallard duck and man. however, whereas the ρ lipoproteins predominate in man with little lipo-albumin evident, according to available literature, in ducks the lipo-albumin constitutes more than half of the serum lipoproteins, and a substantial lipo-prealbumin fraction is evident. Most studies of lipo-proteins are carried out on human serum primarily for the purpose of diagnosis of abnormalities. The stress response in many animals includes a rise in serum α-globulin levels, but its physiological basis is still speculative. While a consideration of the changes in lipoprotein distribution …
