My dissertation research has focused on the development of ambient ionization – mass spectrometry (MS) for clinical measurements, specifically intrasurgical cancer detection. The molecular differences between normal and cancerous tissue were detected via direct tissue analysis in vitro by touch spray ionization (TS) or by analyzing sectioned or smeared tissue using desorption electrospray ionization (DESI). The physical form of the tissue, e.g. in vitro sampling, sectioned, or smeared, was inconsequential in differentiating normal from cancerous tissue; however, the spectra acquired by TS and DESI differed due to differences in ionization processes. Information privacy issues have plagued the world of electronic media since its inception. This research focused mainly on factors that increase or decrease perceived patient control over personal health information (CTL) in the presence of context-specific concerns. Control agency theory was used for the paper's theoretical contributions. Personal and proxy control agencies acted as the independent variables, and context-specific concerns for information privacy (CFIP) were used as the moderator between proxy control agency, healthcare provider, and CTL. Standard curves for a Perkin Elmer TriCarb 2800 liquid scintillation detector (LSC) and a Ludlum 3030p Passivated Implanted Planar Silicon detector have been developed and utilized for studying the dose distribution of depleted uranium (DU) within zebrafish. The DU source was crystallized uranyl nitrate (N2O8U•6H2O) solution, normally used for staining in electron microscopy with a manufactured average specific activity of 0.3 uCi/g. Zebrafish, both larvae and adults, were exposed to three different mass concentrations, dissected, dissolved and counted using an LSC. Glioma is a common type of primary brain tumor that represents 28% of all brain tumors and 80% of malignant tumors. According to a recent study by the Centers for Disease Control and Prevention (CDC), gliomas account for 53%, 35% and 29% of all brain tumors (68%, 74% and 81% of malignant brain tumors) among children (aged 0-14), teenagers (aged 15-19) and young adults, respectively. Gliomas are often diagnosed through radiological imaging and histopathology. The electrochemical method of redox cycling was exploited to achieve new discoveries in neurotransmitter detection and to advance its suitability toward in vivo use. Redox cycling has advantages in signal amplification, selectivity of species based on their electrochemical reaction mechanisms, and limited or no background subtraction. Distinction of dopamine from norepinephrine in a mixture with an electrochemical method at unmodified electrodes was demonstrated for the first time in vitro. This ability resulted from a series of fundamental studies of redox cycling behavior of the catecholamines (dopamine, norepinephrine and epinephrine) using different electrode configurations. The purpose of this study was to evaluate the effects of sodium hypochlorite on the organic and inorganic composition of enamel. Background: With the advent of enamel bonding for orthodontic appliances in the late 1970s, it has been shown that traditional phosphoric acid etching affects the inorganic portion of the enamel.1, 2 In an attempt to enhance the acid etching pattern and, furthermore, the bond strength, additional pretreatment techniques that target the organic components of the enamel biofilm have been proposed. This thesis presents numerical simulation of terahertz (THz) wave interaction with breast cancer tumor tissue sections. The obtained results are expressed in THz images of heterogeneous material that mimics the excised breast cancer tissue sections. The finite-element software package ANSYS High Frequency Structural Simulator (HFSS) was used in this work. Fibrosis around the implanted medical devices is a severe problem that can plague long-term device reliability. Activation of macrophage phenotype (macrophage polarization) has emerged as a new and possible means for reducing fibrosis in the fields of biomaterials and regenerative medicine. Macrophages are phagocytic cells that respond to microenvironmental cues that direct their phenotype. Evidence suggests that high vitamin D status (marked by serum 25-hydroxyvitamin D, 25(OH)2 D) is associated with a decreased risk of breast cancer. It has been established that 1,25-dihydroxyvitamin D (1,25(OH) 2 D) can alter glycolysis and the Krebs cycle of breast cancer cells (Jiang et al., 2010; Zheng et al., 2013) but little information is available on 1,25(OH) 2 D's alterations of lipid metabolism in breast cancer cells. Synthetic cannabinoids (SC's) began to gain popularity around the world in 2009. Since then, many of the compounds have been outlawed and methods developed to detect them and their metabolites using mass spectrometry. Our work investigated the possibility of developing a colorimetric presumptive test. The SC JWH-019 was synthesized and its ketone targeted as a possible reaction site. Many SC's contain ketones and thus a reaction at this site would be applicable to many of the compounds. Histatin 5 is a naturally occurring human salivary antifungal peptide and the first line of defense against infections of the oral cavity. This work tested the hypothesis that oxidation of methionine 388 in thrombomodulin is higher in cigarette smokers, and thus a likely contributor towards the hypercoagulable state in smokers. Thrombomodulin, a protein cofactor found on endothelial cell surfaces, regulates the activity of thrombin. Thrombin bound to thrombomodulin no longer converts fibrinogen to fibrin, but instead activates Protein C which, in turn, stops the coagulation cascade by inactivation of clotting factors. The oxidation of methionine 388 of thrombomodulin has been shown in vitro to dramatically decrease the anticoagulant cofactor activity of thrombomodulin. A transparent nanofluidic system with embedded sensing electrodes was designed and fabricated by integrating Atomic Force Microscopy (AFM) nanolithography, Focused Ion Beam (FIB) milling and metal deposition, and standard microfabrication processing. The fabrication process started with the evaporation of chrome/gold (Cr/Au) onto a Pyrex 7740 wafer followed by photolithography and wet etching of the microchannels. The wafer was patterned a second time to form Au microelectrodes with 15-45 micrometer separation gaps in the nanochannel region. Poly-N-substituted glycines (peptoids) are a very versatile family of synthetic molecules that can be customized for any number of applications. In this study, we chose to use peptoids as a foundation for sandwich ELISA microarray analysis with a long term goal of creating an early detection device for complex diseases such as cancer. The peptoids were designed to self-assemble into microspheres to be used in coatings on the surface of the microarray substrates to increase the surface area available for antibody attachment. In seeking of effective prevention programs to improve physical activities, we want to examine the factors related to physical activities improvement in Alliance for a Healthy Border, a chronic disease prevention program with pre-post-post evaluations through 12 federally qualified community health centers serving primarily Hispanics in communities along the U.S.- Mexico border. A proof-of-concept microfluidic device combined with heparin-immobilized magnetic beads was created to concentrate cytokine proteins collected from microdialysis samples. Cytokines are known to be related to several diseases such as cancer, and Parkinson's diseases, so to be able to develop more effective diseases treatments their interactions have to be well understood. Amine-functionalized polystyrene and carboxyl-functionalized magnetic microspheres of ~6.0 ìm in diameter were used to immobilize heparin. Introduction: Orthodontic tooth movement requires the precise coordination of a multitude of biological factors in order for proper bone remodeling to occur. Cytokines, such as Interleukin-6 (IL-6), play a critical role in the communication necessary for both bone resorption and bone apposition. IL-6 reacts with receptor proteins, such as soluble IL-6 receptor (sIL-6R), on target cells in order to transmit signals important to both osteoclast and osteoblast activities. Characterizing the genetic contributors to complex disease traits will inevitably require consideration of haplotypic phase, the specific alignment of alleles on a single homologous chromosome. In population based studies, however, phase is generally unobservable as standard genotyping techniques provide investigators only with data on unphased genotypes. Several statistical methods have been described for estimating haplotype frequencies and their association with a trait in the context of phase ambiguity. These methods are limited, however, to diploid populations in which individuals have exactly two homologous chromosomes each and are thus not suitable for more general infectious disease settings. The classroom environment can be an acoustically difficult atmosphere for students to learn effectively, sometimes due in part to poor acoustical properties. Noise and reverberation have a substantial influence on room acoustics and subsequently intelligibility of speech. The American Speech-Language-Hearing Association (ASHA, 1995) developed minimal standards for noise and reverberation in a classroom for the purpose of providing an adequate listening environment. A lack of adherence to these standards may have undesirable consequences, which may lead to poor academic performance.

The effects of a physiologically balanced mixture of amino acids on the synthesis of proteins has been investigated. The roles of leucine and tryptophan, both implicated in the regulation of protein synthesis, were also studied. The balance of amino acids is important under specific rate-limiting conditions; the physiological balance "protects" the protein synthesizing system from the stressed condition of leucine limitation. Leucine is an important regulator of protein synthesis and the tryptophan effect on translation is dependent on the concentration of leucine. Thus tryptophan is a secondary regulator. The potential of propylene glycol in water as a vehicle for topical delivery of methotrexate has been examined. The genus Brunfelsia (Fam. Solannceae) comprises some 40 species of shrubs native to South Africa and West Indies. Brunfelsia hopeana (common name: Manaca root) and related species are recognized in folk medicine in South America as being useful as diuretic, antirheumatic, antisyphilitic agents and as narcotic hallucinogens. In view of the interesting folkloric uses and a paucity of chemical and pharmacological investigation of B. hopeana, a systematic phytochemical and pharmacological investigation on B. hopeana was undertaken in our laboratory.
