Formulation Development Of A Polymer-Drug Matrix With A Controlled Release Profile For The Treatment Of Glaucoma, Eric W. Tsoi
Formulation Development Of A Polymer-Drug Matrix With A Controlled Release Profile For The Treatment Of Glaucoma, Eric W. Tsoi
Master's Theses
Glaucoma is the leading cause of blindness in the United States accounting for 9-12% of all cases of blindness. Currently, the front line treatment for glaucoma are prostaglandins that may have to be taken up to several times a day. Even with proper treatment, roughly 11% of the patients using the treatment are non-compliant and lose their vision. In this project, ForSight Laboratories has developed a pharmaceutical drug delivering implant with the capability of sustaining long-term release of a prostaglandin as a new way to treat the condition. This project reports the product development of a polymer drug matrix with …

To control the rate of release of methylprednisolone (MP) in lysosomes, new dextran-MP conjugates with peptide linkers were synthesized and characterized. Methylprednisolone succinate (MPS) was attached to dextran 25 kDa using linkers with 1-5 Gly residues. The release characteristics of the conjugates in pH 4.0 and 7.4 buffers, blood, liver lysosomes, and various lysosomal proteinases were determined using a size-exclusion and/or a newly developed reversed-phase HPLC method capable of simultaneous quantitation of MP, MPS, and all five possible MPS-peptidyl intermediates. We synthesized conjugates with >= 90% purity and 6.9-9.5% (w/w) degree of MP substitution. The conjugates were stable at pH …

Polymeric foams seem to have potential for use in many research areas such as cell seeding and macromolecule or drug loading. Three factors were studied to evaluate the efficiency of foams as drug carrier and releasing agents. Effect of polymeric properties was examined by using two types of polymers (PHBV 7 and PHBV 22) differing only in their valerate (copolymer) content. The other factor studied was the effect of loading extent on release kinetics and rate. The results showed an unexpected inverse proportionality between drug loading and release rate. They, however, were in accordance with foam physical properties such as …
