Current Opinion article considers the diverging perspectives of two academics on the trainability and role of perceptual-cognitive abilities in sports performance, specifically applied to agility and sidestepping. This work uses a moderated dialogue approach between these two authors, each representing differing viewpoints: one advocating for the role of perceptual-motor skills through representative learning environments and another emphasising physical resilience. Background: Neutrophil gelatinase-associated lipocalin (NGAL) is present in secondary granules of neutrophils and it is a relatively newly recognized marker of kidney diseases. The fibrinogen-to-albumin ratio (FAR) is a marker of inflammation but its diagnostic value has not been determined in sickle cell disease patients with kidney diseases. PACT (encoded by the PRKRA gene) is a double-stranded RNA binding protein with defined antiviral defense and cytoplasmic RNA-induced silencing actions in mammals. We previously described a further role for PACT as a modulator of nuclear receptor (NR)-regulated gene expression. Here, we investigated the role of PACT in prostate cancer (PCa) using a loss-of-function approach. The effects of a 21-day live high-train low (LHTL) on hemoglobin mass (Hbmass) and iron demand including the erythropoietin (EPO)-erythroferrone (ERFE)-hepcidin axis and routine iron markers were investigated. Fifteen female endurance athletes completed either 21-day LHTL in normobaric hypoxia (2500 m, ~18 h·day⁻¹) (INT, n = 8) or lived and trained in normoxia (CON, n = 7). Hbmass and resting blood were collected before and after the intervention. An additional blood sample was collected on Day 6 for INT. 21-day LHTL increased Hbmass 3.8% in INT (p < 0.001). EPO increased 35.6% from Day 0 to Day 6 in INT (p = 0.037) and then decreased 42.4% from Day 6 to Day 21 (p = 0.019). In INT, no changes were detected in ERFE or hepcidin, and from the routine iron markers only serum transferrin receptor increased from Day 0 to Day 21 (13.9%, p = 0.013). In CON, no changes were detected in Hbmass or iron markers. In INT, Hbmass and ferritin were positively associated (Day 0 to Day 6, p = 0.005). Thus, hepcidin and ERFE may not provide additional information regarding iron demand following 6- or 21-day LHTL compared to routine iron markers. The relationship between Hbmass and ferritin indicates that adequate ferritin levels are needed during hypoxia to support hematological adaptations.

Introduction: Dietary nitrate, through conversion to nitric oxide, which supports vascular and nervous system function, may lower dementia risk but may also form neurodegenerative N-nitrosamines, depending on the nitrate source. Methods: We investigated associations between source-specific nitrate and nitrite intake and incident and early-onset dementia (< 65 years) in 54,804 dementia-free participants from the Danish Diet, Cancer and Health Cohort Study over ∼27 years. Nitrate and nitrite intakes were derived from food frequency questionnaires and nitrate and nitrite databases. Results: Higher plant-sourced nitrate intake was non-linearly associated with lower rates of incident dementia (fifth vs first quintile hazard ratio 95% confidence interval: 0.90 [0.83, 0.98]), while increased risk was seen for higher intakes of animal-sourced, additive-permitted meat-sourced, and tap water-sourced nitrate. Similar associations were seen for source-specific nitrite intake and were more pronounced for early-onset dementia. No clear effect modification was observed. Discussion: These findings highlight the importance of nitrate source in dementia risk and warrant further investigation. Highlights: Plant nitrate is associated with a lower risk of incident and early-onset dementia. Animal and tap water nitrate are associated with an increased risk of dementia. Encouraging consumption of plant-based nitrate sources may lower risk of dementia.

Objective Chronically elevated circulating lipocalin-2 (LCN2) levels are implicated in poor energy regulation, increased cardio-metabolic disease risk, and poor physical function. Exercise is known to improve these factors, but whether LCN2 is modifiable by exercise is not clear.
