Clinical-Transcriptional Prioritization Of The Circulating Proteome In Human Heart Failure, Andrew S Perry, Junedh Amrute, Kory J Lavine, Et Al.
Clinical-Transcriptional Prioritization Of The Circulating Proteome In Human Heart Failure, Andrew S Perry, Junedh Amrute, Kory J Lavine, Et Al.
2020-Current year OA Pubs
Given expanding studies in epidemiology and disease-oriented human studies offering hundreds of associations between the human "ome" and disease, prioritizing molecules relevant to disease mechanisms among this growing breadth is important. Here, we link the circulating proteome to human heart failure (HF) propensity (via echocardiographic phenotyping and clinical outcomes) across the lifespan, demonstrating key pathways of fibrosis, inflammation, metabolism, and hypertrophy. We observe a broad array of genes encoding proteins linked to HF phenotypes and outcomes in clinical populations dynamically expressed at a transcriptional level in human myocardium during HF and cardiac recovery (several in a cell-specific fashion). Many identified …

Vglut2-Based Glutamatergic Signaling In Central Noradrenergic Neurons Is Dispensable For Normal Breathing And Chemosensory Reflexes, Yuan Chang, Savannah Lusk, Andersen Chang, Christopher S Ward, Russell S Ray
Faculty, Staff and Students Publications
Central noradrenergic (NA) neurons are key constituents of the respiratory homeostatic network. NA dysfunction is implicated in several developmental respiratory disorders including Congenital Central Hyperventilation Syndrome (CCHS), Sudden Infant Death Syndrome (SIDS), and Rett Syndrome. The current unchallenged paradigm in the field, supported by multiple studies, is that glutamate co-transmission in subsets of central NA neurons plays a role in breathing control. If true, NA-glutamate co-transmission may also be mechanistically important in respiratory disorders. However, the requirement of NA-derived glutamate in breathing has not been directly tested and the extent of glutamate co-transmission in the central NA system remains uncharacterized. …

Myotube Guidance: Shaping Up The Musculoskeletal System, Aaron N Johnson
Myotube Guidance: Shaping Up The Musculoskeletal System, Aaron N Johnson
2020-Current year OA Pubs
Myofibers are highly specialized contractile cells of skeletal muscles, and dysregulation of myofiber morphogenesis is emerging as a contributing cause of myopathies and structural birth defects. Myotubes are the myofiber precursors and undergo a dramatic morphological transition into long bipolar myofibers that are attached to tendons on two ends. Similar to axon growth cones, myotube leading edges navigate toward target cells and form cell-cell connections. The process of myotube guidance connects myotubes with the correct tendons, orients myofiber morphology with the overall body plan, and generates a functional musculoskeletal system. Navigational signaling, addition of mass and volume, and identification of …
Adamts12 Promotes Fibrosis By Restructuring Extracellular Matrix To Enable Activation Of Injury-Responsive Fibroblasts, Konrad Hoeft, Benjamin D. Humphreys, Et Al.
Adamts12 Promotes Fibrosis By Restructuring Extracellular Matrix To Enable Activation Of Injury-Responsive Fibroblasts, Konrad Hoeft, Benjamin D. Humphreys, Et Al.
2020-Current year OA Pubs
Fibrosis represents the uncontrolled replacement of parenchymal tissue with extracellular matrix (ECM) produced by myofibroblasts. While genetic fate-tracing and single-cell RNA-Seq technologies have helped elucidate fibroblast heterogeneity and ontogeny beyond fibroblast to myofibroblast differentiation, newly identified fibroblast populations remain ill defined, with respect to both the molecular cues driving their differentiation and their subsequent role in fibrosis. Using an unbiased approach, we identified the metalloprotease ADAMTS12 as a fibroblast-specific gene that is strongly upregulated during active fibrogenesis in humans and mice. Functional in vivo KO studies in mice confirmed that Adamts12 was critical during fibrogenesis in both heart and kidney. …
Design Considerations For Self-Management Technologies For People Living With Dementia And Informal Carers – Perspectives Of Healthcare Professionals And Charity Workers, Dympna O'Sullivan
Design Considerations For Self-Management Technologies For People Living With Dementia And Informal Carers – Perspectives Of Healthcare Professionals And Charity Workers, Dympna O'Sullivan
Conference papers
Dementia is a neurodegenerative disorder that leads to decline in memory, language, reasoning, and the ability to perform daily activities. It is linked to poorer quality of life for the person with dementia and their informal (unpaid) carers. While early intervention and access to adequate care are critical in slowing dementia's progression and better managing associated symptoms, dementia is frequently only diagnosed at an advanced stage and care is often fragmented. To better understand how to meet the complex needs of persons living with dementia and their informal carers, 10 healthcare professionals and 10 charity workers from relevant community and …

Partial Purification Of Alzheimer’S Amyloid-𝛽 Specific Antibody Using Ammonium Sulfate, Noor Yousaf
Partial Purification Of Alzheimer’S Amyloid-𝛽 Specific Antibody Using Ammonium Sulfate, Noor Yousaf
Undergraduate Research Symposium
Alzheimer's disease (AD) is the most common neurodegenerative disease. The trigger for AD is the accumulation of amyloid-beta protein (Aβ) as senile plaques in the brain. Prior to forming the fiber-like structures found in the plaques, Aβ undergoes an oligomerization process that produces intermediate structures called protofibrils. Substantial data from the Nichols laboratory demonstrated that soluble Aβ protofibrils were highly inflammatory compared to other forms of Aβ. Based on these findings, a serum polyclonal antibody, named Antibody St. Louis or AbSL, was developed to target Aβ protofibrils. A significant challenge with serum antibodies is the presence of many other biological …
