Mutations in ATP-binding cassette A3 (ABCA3), a phospholipid transporter critical for surfactant homeostasis in pulmonary alveolar type II epithelial cells (AEC2s), are the most common genetic causes of childhood interstitial lung disease (chILD). Treatments for patients with pathological variants of ABCA3 mutations are limited, in part due to a lack of understanding of disease pathogenesis resulting from an inability to access primary AEC2s from affected children. Here, we report the generation of AEC2s from affected patient induced pluripotent stem cells (iPSCs) carrying homozygous versions of multiple ABCA3 mutations. General anesthesia-a pharmacologically induced reversible state of unconsciousness-enables millions of life-saving procedures. Anesthetics induce unconsciousness in part by impinging upon sexually dimorphic and hormonally sensitive hypothalamic circuits regulating sleep and wakefulness. Thus, we hypothesized that anesthetic sensitivity should be sex-dependent and modulated by sex hormones. Using distinct behavioral measures, we show that at identical brain anesthetic concentrations, female mice are more resistant to volatile anesthetics than males. Anesthetic sensitivity is bidirectionally modulated by testosterone. Castration increases anesthetic resistance. Conversely, testosterone administration acutely increases anesthetic sensitivity. Conversion of testosterone to estradiol by aromatase is partially responsible for this effect. In contrast, …

BACKGROUND: Cardiac metabolic dysfunction is a hallmark of heart failure (HF). Estrogen-related receptors ERRα and ERRγ are essential regulators of cardiac metabolism. Therefore, activation of ERR could be a potential therapeutic intervention for HF. However, in vivo studies demonstrating the potential usefulness of ERR agonist for HF treatment are lacking, because compounds with pharmacokinetics appropriate for in vivo use have not been available.

Protein kinases are frequently dysregulated and/or mutated in cancer and represent essential targets for therapy. Accurate quantification is essential. For breast cancer treatment, the identification and quantification of the protein kinase ERBB2 is critical for therapeutic decisions. While immunohistochemistry (IHC) is the current clinical diagnostic approach, it is only semiquantitative. Mass spectrometry-based proteomics offers quantitative assays that, unlike IHC, can be used to accurately evaluate hundreds of kinases simultaneously. The enrichment of less abundant kinase targets for quantification, along with depletion of interfering proteins, improves sensitivity and thus promotes more effective downstream analyses. AMP-activated protein kinase (AMPK) plays a crucial role in maintaining ATP homeostasis in photoreceptor neurons. AMPK is a heterotrimeric protein consisting of α, β, and γ subunits. The independent functions of the 2 isoforms of the catalytic α subunit, PRKAA1 and PRKAA2, are uncharacterized in specialized neurons, such as photoreceptors. Here, we demonstrate in mice that rod photoreceptors lacking PRKAA2, but not PRKAA1, showed altered levels of cGMP, GTP, and ATP, suggesting isoform-specific regulation of photoreceptor metabolism. Furthermore, PRKAA2-deficient mice displayed visual functional deficits on electroretinography and photoreceptor outer segment structural abnormalities on transmission electron microscopy consistent with neuronal dysfunction, …

Abdominal aortic aneurysms (AAAs) are prevalent with aging, and AAA rupture is associated with increased mortality. There is currently no effective medical therapy to prevent AAA rupture. The monocyte chemoattractant protein (MCP-1)/C-C chemokine receptor type 2 (CCR2) axis critically regulates AAA inflammation, matrix-metalloproteinase (MMP) production, and extracellular matrix (ECM) stability. We therefore hypothesized that a diet intervention that can modulate CCR2 axis may therapeutically impact AAA risk of rupture. Since ketone bodies (KBs) can trigger repair mechanisms in response to inflammation, we evaluated whether systemic ketosis in vivo could reduce CCR2 and AAA progression. BACKGROUND: Pediatric dilated cardiomyopathy often leads to death or cardiac transplantation. We sought to determine whether changes in left ventricular (LV) end-diastolic dimension (LVEDD), LV end-diastolic posterior wall thickness, and LV fractional shortening (LVFS) over time may help predict adverse outcomes.
METHODS AND RESULTS: We studied children up to 18 years old with dilated cardiomyopathy, enrolled between 1990 and 2009 in the Pediatric Cardiomyopathy Registry. Age-related macular degeneration (AMD) is a leading cause of blindness featuring pathogenic neovascularization of the choroidal vasculature (CNV). Although systemic immunity plays a role in AMD, the ocular signals that recruit and activate immune cells remain poorly defined. Using single-cell RNA sequencing, we prospectively profile peripheral blood mononuclear cells from 65 individuals including AMD and controls, which we integrate with existing choroid data. We generate a network of choroid-peripheral immune interactions dysregulated in AMD, including known AMD-relevant gene vascular endothelial growth factor (VEGF) receptor 2. Additionally, we find CYR61 is upregulated in choroidal veins and may signal to circulating monocytes. …

A collaborative effort from various artists at MRC artWorks located in Kalamazoo, MI contributed to the cover art for the Winter 2024 issue of the Open Journal of Occupational Therapy (OJOT). MRC Industries is a non-profit organization that focuses on supporting individuals with developmental disabilities, cognitive or emotional impairments or illnesses, and people who have experienced traumatic brain injuries. One particular program has become a staple in the Kalamazoo community – MRC artWorks, a professional working art studio for adults with disabilities to learn skills, both physical and emotional, through art instruction and art-making.

Large-scale commercial farmers and small scale “hobby” farmers engage in growing crops for subsistence and survival for themselves, their families, communities, and beyond. Their reverence and passion for farming make them good stewards of both community and world population nourishment. We suggest there may be a collective physical, emotional, and spiritual draw to farming that is interwoven with occupational engagement. Through a personal narrative approach, we reflect on the meaning and the work-focused occupation of farming and specifically how spirituality may be a guiding force that supports engagement despite seemingly insurmountable environmental and personal hardships. Teens and young adults living with autism spectrum disorder (ASD) do not have equal access to sexual education, thus having lower sexual awareness, little knowledge of preventative sexual health care, and having a more difficult time reading sexual body language (Hannah & Stagg, 2016; Holmes et al., 2019; Mogavero & Hsu, 2020). Musicians are a distinct population with a high rate of developing musculoskeletal and neurological disorders affecting their ability to participate in music-based occupations. These injuries negatively affect the musician’s health and well-being in physical, financial, psychosocial, and emotional dimensions. Given music’s complex and integrated role in the musicians’ life, treatment must be multi-dimensional, considering aspects of the person, the type of instrument, environment, context, and social obligations. Background: Upper limb neuromuscular impairments can adversely impact function. This case report investigates the process and outcomes of occupational therapy (OT) for training in the use of the NuroSleeve, a novel research-grade exoskeletal powered orthosis, with a participant with chronic right hemiparesis following a stroke.
Method: The participant engaged in 24 OT sessions using the NuroSleeve over 10 weeks. Therapeutic interventions included neuromuscular reeducation, device management, and engagement in occupation-based activities with training to use the NuroSleeve. The COVID-19 pandemic emotionally and physically impacted students in occupational therapy and occupational therapy assistant programs. College students lost autonomy and access to meaningful occupations and environments. As students returned to campus, they struggled to navigate and adapt to in-person occupations and how they use their time. This study uses a mixed method descriptive research design to understand how the pandemic affected occupational therapy students' time-use and occupational engagement during the transition to in-person learning. Background: Objective Structured Clinical Examinations (OSCEs) are widely used in health programs to assess clinical skills. We present results of a qualitative study investigating occupational therapy students’ perceptions of OSCEs’ impact on their learning and readiness for clinical practice.
Method: Six second and six third year students in the University of Alberta’s Master of Science in Occupational Therapy program were interviewed in separate focus groups. Independent reviewers applied thematic analysis to the focus group transcripts to identify, analyze, and report themes in the data.

Background: Hand therapy clinical rotations are 12 weeks, and one common anecdotal change frequently expressed is an increase in hand strength. Quantifiable measures of change are not documented in the literature. The purpose of this study is to determine if students experience significant increases in strength during a clinical rotation.
Methods: This study is a quasi-experimental, double-blinded, non-randomized sample of convenience. Grip and pinch strength were assessed pre, mid, and post clinical rotation. Demographic data were collected and reported. Paired t-tests were used to compare means between groups. SPSS 27.0 was used for data analysis.

Background: Doctoral students experience decreased well-being during their educational experience. Self-compassion, engagement in meaningful occupations, and occupational balance positively impact well-being in individuals. This study examined the relationships between these constructs in postprofessional occupational science and occupational therapy students.
Method: This quantitative cross-sectional study collected national survey data (N = 113) using the Self-Compassion Scale – Short Form, the Engagement in Meaningful Activities Survey, the Occupational Balance Questionnaire 11, and the 14-item Scales of General Well-Being. Background: Universal design for learning (UDL) is considered best-practice for embracing inclusion for students with disabilities and there is growing evidence of its effectiveness in primary, secondary, and postsecondary education. However, little is known about if and how UDL is being implemented into United States graduate allied health and medical school curriculum as well as evidence of its benefits.

Background: Universal Design for Learning (UDL), an educational framework that describes flexible approaches to teaching and learning, can be used to address problems related to limitations in health literacy in health care settings.
Methods: This exploratory observational study, using a web-based online survey, was undertaken to discern how occupational therapists use the principles of UDL in educating their clients and to determine if differences exist between degree type or practice settings and UDL implementation.

Background: Occupational Adaptation occurs when a person develops a response to meet an occupational challenge. This process is disrupted when a person experiences a life altering event, such as stroke. Stroke self-management programs equip persons with stroke with education and skills to manage the daily tasks associated with their condition. Few studies have explored the adaptive process survivors experience as they seek to effectively use the tools provided through stroke self-management programs. Background: Adult functional literacy ensures adequate and safe engagement in daily activities. It is assessed through the Inventory of Reading Occupations-Adult (IRO-A). The instrument underwent translation with cultural adaptation and content validation to ensure relevance to the Filipino context.
Method: The translation and cultural adaptation of the IRO-A to Filipino (Fil IRO-A) was guided by the process proposed by two international guidelines for cross-cultural adaptations that involves (a) forward translation and synthesis, (b) back translation, and (c) pre-panel review of the adaptation to the Filipino context. Background: Sensory processing behaviors, the temperament characteristic for effortful control, and executive function promote self-regulation, activity engagement, and problem-solving. This study examined inter-relationships between executive function, effortful control, and sensory processing in school-aged children between 7 and 10.11 years of age.
Method: Descriptive correlation research design was used to examine relationships of outcomes from three caregiver-reported, standardized questionnaires of behaviors related to sensory processing (Sensory Profile-2), effortful control (Temperament in Middle Childhood Questionnaire) and executive function (Behavior Rating Inventory of Executive Function-2) in daily activities (N = 19).

Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease characterized by motor neuron loss. Importantly, non-neuronal cell types such as astrocytes also play significant roles in disease pathogenesis. However, mechanisms of astrocyte contribution to ALS remain incompletely understood. Astrocyte involvement suggests that transcellular signaling may play a role in disease. We examined contribution of transmembrane signaling molecule ephrinB2 to ALS pathogenesis, in particular its role in driving motor neuron damage by spinal cord astrocytes. In symptomatic SOD1G93A mice (a well-established ALS model), ephrinB2 expression was dramatically increased in ventral horn astrocytes. OBJECTIVES: Galectin-3 is a beta-galactoside-binding lectin and is a marker of cardiovascular disease (CVD) in the general population. It may also play a role in joint inflammation. We asked whether serum galectin-3 is a useful marker of subclinical vascular disease in patients with rheumatoid arthritis (RA).
METHODS: RA patients without clinical CVD underwent assessment of coronary artery calcium (CAC) score, aortic inflammation (using 18Fluorodeoxyglucose positron emission-computed tomography [FDG PET/CT]), and myocardial flow reserve (MFR). Increased popular discourse about societal inequities highlights the importance of critical consciousness (CC) as a developmental competency for youth. Peer dialogue has been implicit to theories of CC, but we know little about how critical reflection, action, and sociopolitical efficacy inform interpersonal mechanisms through which social networks are selected in developmentally salient contexts. We used social network analysis methods to explore the role of CC components in how late adolescents cultivated their peer affiliation and friendship networks within a specific context of a college living and learning community. BACKGROUND: QiDiTangShen granules (QDTS), a traditional Chinese medicine (TCM) compound prescription, have remarkable efficacy in diabetic nephropathy (DN) patients, and their pharmacological mechanism needs further exploration.
METHODS: According to the active ingredients and targets of the QDTS in the TCMSP database, the network pharmacology of QDTS was investigated. The potential active ingredients were chosen based on the oral bioavailability and the drug similarity index. Low human papillomavirus (HPV) vaccine uptake is a key barrier to cervical cancer elimination. We aimed to evaluate the health impact and cost-effectiveness of introducing different HPV vaccines into immunization programs and scaling up the screening program in Guangdong. We used a dynamic compartmental model to estimate the impact of intervention strategies during 2023-2100. We implemented the incremental cost-effectiveness ratio (ICER) in costs per averted disability-adjusted life year (DALY) as an indicator to assess the effectiveness of the intervention. Our objective was to evaluate the association of antioxidant intake and the inflammatory potential of the diet with functional decline in older men. A diet history questionnaire was used to collect dietary intake data from men aged ≥ 75 years (n 794) participating in the Concord Health and Aging in Men Project cohort study. Intake of vitamins A, C, E and Zn were compared with the Australian Nutrient Reference Values to determine adequacy. The Energy-adjusted Dietary Inflammatory Index (E-DIITM) was used to assess the inflammatory potential of the diet. Physical performance data were collected via handgrip strength and walking speed …
