Surgical patients can be discharged to a variety of facilities which vary widely in intensity of care. Postoperative readmissions have been found to be more strongly associated with post-discharge events than pre-discharge complications, indicating the importance of discharge destination. We sought to evaluate the association between discharge destination and 30-day outcomes. A retrospective cohort study was conducted using the American College of Surgeons National Surgical Quality Improvement Program (ACS-NSQIP) database. Patients were dichotomized based on discharge destination: home versus non-home. The main outcome of interest was 30-day unplanned readmission. The secondary outcomes included post-discharge pulmonary, infectious, thromboembolic, and bleeding complications, …

Technologies capable of programmable translation activation offer strategies to develop therapeutics for diseases caused by insufficient gene expression. Here, we present "translation-activating RNAs" (taRNAs), a bifunctional RNA-based molecular technology that binds to a specific mRNA of interest and directly upregulates its translation. taRNAs are constructed from a variety of viral or mammalian RNA internal ribosome entry sites (IRESs) and upregulate translation for a suite of target mRNAs. We minimize the taRNA scaffold to 94 nucleotides, identify two translation initiation factor proteins responsible for taRNA activity, and validate the technology by amplifying SYNGAP1 expression, a haploinsufficiency disease target, in patient-derived cells. …

Intrinsically disordered regions (IDRs) represent a large percentage of overall nuclear protein content. The prevailing dogma is that IDRs engage in non-specific interactions because they are poorly constrained by evolutionary selection. Here, we demonstrate that condensate formation and heterotypic interactions are distinct and separable features of an IDR within the ARID1A/B subunits of the mSWI/SNF chromatin remodeler, cBAF, and establish distinct "sequence grammars" underlying each contribution. Condensation is driven by uniformly distributed tyrosine residues, and partner interactions are mediated by non-random blocks rich in alanine, glycine, and glutamine residues. These features concentrate a specific cBAF protein-protein interaction network and are …

Topics in Exercise Science and Kinesiology Volume 4: Issue 1, Article 15, 2023. In volleyball, the approach run serves as a strategy to execute a spike from an elevated striking position. Nevertheless, the influence of the step number of the approach run on the jumping kinematics in spike jumps is unclear. Therefore, this investigation aims to clarify the impact of varying step numbers within the approach run on jumping kinematics. Seven female volleyball athletes were tasked with executing spike jumps, employing 2 and 3-step approach runs. These trials were recorded using high-speed cameras and digitized, and then the kinematics of …

An Investigation Of Mitigation Measures On The Spread Of Covid-19 In A College Classroom Using Agent-Based Modeling, Saharsh Talwar, Anne E. Yust
Spora: A Journal of Biomathematics
In this manuscript, we describe the process of using agent-based modeling in NetLogo to create a simulation of COVID-19 spread in a traditional college classroom. The model allows for an evaluation of different preventative measures implemented by the University of Pittsburgh, including the cohort classroom attendance model, mask and vaccine mandates, contact tracing, and classroom sanitation. Through the use of the model's interactive interface, the impact of adjusting specific measures by the institution could be visualized, providing a valuable tool for combating diseases that spread through droplet transmission.

Background
- Considerable stigma exists toward persons experiencing homelessness (PEH)—a vulnerable population at greater risk for health disparities.1, 2
- Many healthcare students begin their programs without interacting with PEH and with preconceived notions influenced by societal stigma.
- Interprofessional education programs, such as the Enhancing Services for People Experiencing Homelessness program below, have incorporated experiential learning into curricula to address student biases and develop understanding and compassion.
- Educating students to understand issues that PEH face can help mitigate healthcare disparities.
- Evaluating the effectiveness of these interprofessional educational experiences requires valid assessment of the attitudes, interest, and confidence …

Chemotherapy remains a primary treatment for younger AML patients, though many relapse. Data from our group have shown that highly phosphorylated S6 in blasts may predict response to sirolimus given with chemotherapy. We report the results of a phase I study of this combination in newly diagnosed AML and the pharmacodynamic analysis of pS6 before and after treatment. Subjects received sirolimus (12 mg on day 1, 4 mg daily, days 2-10), then idarubicin and cytarabine (days 4-10). Response was assessed at hematologic recovery or by day 42 using a modified IWG criteria. Fifty-five patients received sirolimus. Toxicity was similar to …
