Research outputs 2022 to 2026

Delayed recall (DR) impairment is one of the most significant predictive factors in defining the progression to Alzheimer’s disease (AD). Changes in brain functional connectivity (FC) could accompany this decline in the DR performance even in a resting state condition from the preclinical stages to the diagnosis of AD itself, so the characterization of the relationship between the two phenomena has attracted increasing interest. Another aspect to contemplate is the potential moderator role of the APOE genotype in this association, considering the evidence about their implication for the disease. 379 subjects (118 mild cognitive impairment (MCI) and 261 cognitively intact …

Epigenetic Regulation Of Corneal Epithelial Differentiation By Tet2, Yuzuru Sasamoto, Siyuan Wu, Catherine A. A. Lee, Jason Y. Jiang, Bruce R. Ksander, Markus H. Frank, Natasha Y. Frank

Research outputs 2022 to 2026

Epigenetic DNA modification by 5-hydroxymethylcytosine (5hmC), generated by the Ten-eleven translocation (TET) dioxygenases, regulates diverse biological functions in many organ tissues, including the mammalian eye. For example, 5hmC has been shown to be involved in epigenetic regulation of retinal gene expression. However, a functional role of 5hmC in corneal differentiation has not been investigated to date. Here, we examined 5hmC and TET function in the human cornea. We found 5hmC highly expressed in MUC16-positive terminally differentiated cells that also co-expressed the 5hmC-generating enzyme TET2. TET2 knockdown (KD) in cultured corneal epithelial cells led to significant reductions of 5hmC peak distributions …

Editorial: Untangle The Broad Connections And Tight Interactions Between Human Microbiota And Complex Diseases Through Data-Driven Approaches, Liang Wang, Zuo-Bin Zhu, Li Zhang, Jian Li, Qi Zhao

Research outputs 2022 to 2026

It is well-known that microorganisms are ubiquitous in the environment and occupy almost all habitats in animals and humans (Finlay and Clarke, 1999; Rosenberg, 2021). Traditionally, microorganisms are studied as individuals grown in isolation under artificial conditions; however, with the development of experimental techniques and computational methods, microbes are now frequently considered as a functional group in a particular niche and studied at the community level in order to best mimicking the real-world situations (American Academy of Microbiology, 2004). During the study of microbial communities, two terms are commonly used, that is, microbiota and microbiome.

Mindfulness Based Stress Reduction Techniques In Chronic Illness, Anna Celia Gil Feb 2023

Mindfulness Based Stress Reduction Techniques In Chronic Illness, Anna Celia Gil

Electronic Theses and Dissertations

Abstract

Chronic stress and its effect on both physical and mental health have been a topic of increasing interest in the past several decades. There is mounting evidence that recurrent, persistent stress of both a physiologic and psychological nature leads to a dysfunction in the hypothalamic pituitary axis (HPA), the system which helps the body recover and adapt to short term stress. In long term stress, this HPA dysfunction leads to impairment of the immune and neuroendocrine systems resulting in chronic low-grade inflammation. This type of chronic inflammation acts as a precursor to several chronic illnesses such as diabetes, hypertension, …

Analysis Of Electrophysiological Markers And Correlated Components Of Neural Responses To Discourse Coherence, Kurt M. Masiello Feb 2023

Analysis Of Electrophysiological Markers And Correlated Components Of Neural Responses To Discourse Coherence, Kurt M. Masiello

Dissertations, Theses, and Capstone Projects

Constructing meaning from spoken language is invaluable for learning, social interaction, and communication. In clinical populations with developmental disorders of speech comprehension, the severity of disruption can persist and vary from limiting occupational opportunities to lower performance outcomes. Previous research has reported an event-related potential (ERP) neural positivity over right hemisphere lateral anterior sites in response to semantic and discourse processing. Although useful as a marker for clinical populations of autism spectrum disorder (ASD) and developmental language disorder (DLD), little is understood about the dynamics and neural sources of this biological marker. In addition to traditional methods of ERP analysis, …

From The Neck Up, Christopher J. Smiley Feb 2023

From The Neck Up, Christopher J. Smiley

The Journal of the Michigan Dental Association

This editorial emphasizes integrating behavioral health into dentistry for comprehensive patient care. It highlights evidence-based approaches showing positive outcomes of behavior change techniques in oral hygiene and smoking cessation. However, implementing behavioral change models in clinical dental care faces challenges, such as reimbursement issues and concerns about disrupting the patient-dentist relationship. The editorial suggests enlisting outside resources and fostering collaboration between oral and mental health professionals to address patients' overall health needs. Policy initiatives are seen as a promising avenue for advancing this integration.

Marijuana Liberalization And Public Finance: A Capital Market Perspective On The Passage Of Medical Use Laws, Stephanie F. Cheng, Gus De Franco, Pengkai Lin Feb 2023

Marijuana Liberalization And Public Finance: A Capital Market Perspective On The Passage Of Medical Use Laws, Stephanie F. Cheng, Gus De Franco, Pengkai Lin

Research Collection School Of Accountancy

We find that the staggered passage of state-level laws that legalize marijuana for medical use increases states' borrowing costs by 7–9 basis points. Consistent with economic theory on substance use suggesting that marijuana legalization increases local consumption of the drug (by expanding its availability and reducing its perceived risks), we predict and find that increased consumption represents an important mechanism that explains the higher state bond spreads. We also show that following such laws’ passage, states incur higher marijuana-consumption-related expenditures, including for police, corrections, and public welfare.

Whilst DNA repeat expansions cause numerous heritable human disorders, their origins and underlying pathological mechanisms are often unclear. We collated a dataset comprising 224 human repeat expansions encompassing 203 different genes, and performed a systematic analysis with respect to key topological features at the DNA, RNA and protein levels. Comparison with controls without known pathogenicity and genomic regions lacking repeats, allowed the construction of the first tool to discriminate repeat regions harboring pathogenic repeat expansions (DPREx). At the DNA level, pathogenic repeat expansions exhibited stronger signals for DNA regulatory factors (e.g. H3K4me3, transcription factor-binding sites) in exons, promoters, 5'UTRs and …

Immunotherapy efficacy is limited in melanoma, and combinations of immunotherapies with other modalities have yielded limited improvements but also adverse events requiring cessation of treatment. In addition to ineffective patient stratification, efficacy is impaired by paucity of intratumoral immune cells (itICs); thus, effective strategies to safely increase itICs are needed. We report that dietary administration of l-fucose induces fucosylation and cell surface enrichment of the major histocompatibility complex (MHC)-II protein HLA-DRB1 in melanoma cells, triggering CD4+ T cell-mediated increases in itICs and anti-tumor immunity, enhancing immune checkpoint blockade responses. Melanoma fucosylation and fucosylated HLA-DRB1 associate with intratumoral T cell abundance …

Do We Need Patient-Specific Qa For Adaptively Generated Plans? Retrospective Evaluation Of Delivered Online Adaptive Treatment Plans On Varian Ethos, Xiaodong Zhao, Dennis N Stanley, Carlos E Cardenas, Joseph Harms, Richard A Popple

2020-Current year OA Pubs

BACKGROUND: The clinical introduction of dedicated treatment units for online adaptive radiation therapy (OART) has led to widespread adoption of daily adaptive radiotherapy. OART allows for rapid generation of treatment plans using daily patient anatomy, potentially leading to reduction of treatment margins and increased normal tissue sparing. However, the OART workflow does not allow for measurement of patient-specific quality assurance (PSQA) during treatment delivery sessions and instead relies on secondary dose calculations for verification of adapted plans. It remains unknown if independent dose verification is a sufficient surrogate for PSQA measurements.

PURPOSE: To evaluate the plan quality of previously treated …
