Although they share certain biological properties with nucleic acid based infectious agents, prions, the causative agents of invariably fatal, transmissible neurodegenerative disorders such as bovine spongiform encephalopathy, sheep scrapie, and human Creutzfeldt Jakob disease, propagate by conformational templating of host encoded proteins. Once thought to be unique to these diseases, this mechanism is now recognized as a ubiquitous means of information transfer in biological systems, including other protein misfolding disorders such as those causing Alzheimer's and Parkinson's diseases. To address the poorly understood mechanism by which host prion protein (PrP) primary structures interact with distinct prion conformations to influence pathogenesis, …

Kisspeptin Receptor Agonist (Ftm080) Increased Plasma Concentrations Of Luteinizing Hormone In Anestrous Ewes, Brian K. Whitlock, Kelly A. Chameroy, Rebecca R. Payton, Shinya Oishi, Nobutaka Fujii, Joseph A. Daniel

Brian K Whitlock, PhD, DVM, DACT

Kisspeptin (KP) and the KP receptor (Kiss1r) are integral to central regulation of the gonadotropic-axis. The demonstration that intravenous infusion of KP can stimulate gonadotropin secretion and ovulation in seasonally anestrous female sheep offers a means of manipulating the reproductive axis. However, KP may be of limited clinical use because of the short circulating half-life. Kiss1r agonists with increased half-life and similar efficacy to KP in vitro potentially may provide beneficial applications in breeding management of many species. However, many of these agonists have not been tested in vivo. This study was designed to test and compare the effects of …

The Pharmacologic Inhibition Of Premature Labor, Jennifer Niebyl, D. Blake, J. Johnson, T. King May 2013

The Pharmacologic Inhibition Of Premature Labor, Jennifer Niebyl, D. Blake, J. Johnson, T. King

Jennifer R Niebyl

Oxytocin, elevated estrogen-progesterone ratio, fetal corticosteroids, prostaglandins, catecholamines, and changes in uterine blood flow have all been implicated as triggers of labor. In approximately one-third of cases of threatened premature labor contractions stop spontaneously. Thus placebo-controlled randomized trials of any new drug for inhibition of premature labor are necessary, as the spontaneous cessation of contractions always favors the claimed therapeutic efficacy. Alcohol inhibits the release of endogenous oxytocin and has an additional direct effect on the myometrium. In one study alcohol was more effective than placebo in the postponement of delivery. Isoxsuprine, ritodrine, and terbutaline have also been shown to …

An 8-month-old crossbred ewe, normal upon physical examination, was humanely euthanized for tissue collection. After approximately 3 weeks in tissue culture, fungi began budding out of cells obtained from the choroid plexus. After an additional 3 weeks, budding was observed in kidney cell cultures and eventually in monocyte cultures as well. Serum from the lamb was submitted to the Veterinary Diagnostic Laboratory at Colorado State University for fungal diagnosis and was found negative for Aspergillus, Blastomyces, Coccidioidomycosis and Histoplasmosis. DNA was isolated from fungi collected from tissue culture supernatants and used in a set of pan-fungal PCR assays with …

Turkish Journal of Veterinary & Animal Sciences

The aim of this study was to determine the effects of 2 different transport temperatures on the in vitro maturation of oocytes collected from frozen-thawed sheep ovaries. Sheep ovaries were transferred into saline at temperatures of 4 °C and 32 °C. After the 2 experimental groups (A: fresh cortex, B: frozen-thawed cortex) were formed, each group was divided into 2 subgroups (group A1: 4 °C, group A2: 32 °C [control]; group B1: 4 °C, group B2: 32 °C). The cortexes were dissected into slices 1-3 mm thick and pieces of 0.5 cm2. For groups B1 and B2, 1-2 cortex pieces …

Turkish Journal of Veterinary & Animal Sciences

The aim of this study was to determine the levels of serum selenium in coarse-haired Pomeranian sheep and to evaluate the effect of the oral supplementation of selenized yeast on selected performance parameters in such sheep. The sheep were divided into 2 groups, with 53 ewes forming the experimental group and 34 ewes forming the control group. The concentration of serum selenium in all 87 sheep from the 2 groups was analyzed. The sheep in the experimental group received selenized yeast for the 3 days before and 3 days after mating (1 g preparation/animal daily, with 1 g of preparation …

Turkish Journal of Veterinary & Animal Sciences

Scrapie is a fatal neurodegenerative disease affecting small ruminants that causes economic losses in many countries. It is categorized together with other fatal neurodegenerative diseases as a transmissible spongiform encephalopathy. The aim of this study was to investigate clinical cases and the cause of death in a flock of sheep from Northern Cyprus (Turkish Republic of Northern Cyprus) suspected of having scrapie. Brains from 4 sheep showing clinical signs of classical scrapie, out of a flock of 200 animals, were analyzed by histopathological examination and infective prion enzyme-linked immunosorbent assay (ELISA). The 4 affected sheep were 2-3 years of age …
