Development Of An Enzyme Immunoassay For The Determination Of Ovine Growth Hormone In Plasma, Behi̇ç Serpek, Seyfullah Hali̇loğlu

A competitive enzyme immunoassay for the determination of ovine growth hormone (oGH) was developed. The assay is based on (anti-rabbit IgG) goat IgG-coated microtitreplates, anti-oGH from rabbits, biotinyl-oGH and streptavidin-horseradish peroxidase. The assay is done directly with 40 ml of plasma and the calibration graph (90% relative binding at 0.38 ± 0.02 ng ml -1 and 50% relative binding at 3.80 ± 0.18 ng ml -1 ) is prepared in plasma without measurable endogenous oGH. Recovery in different plasma samples with added oGH amounted to 96.78 - 100.03 %, and all variabilities were < 10.44. The assay shows no significant binding of oFSH, oLH, oTSH and oPRL. The assay detects high and low plasma oGH levels within the physiological variation as well as changes in plasma oGH after stimulation with growth hormone releasing factor. Hence, it offers a reliable alternative radioimmunoassay. However, a reference method providing evidence of the identity of exogenous oGH will be required for proving illegal treatment with oGH and for forensic purposes.

The present study was conducted to determine whether the mushroom Pleurotus florida has potential to increase the digestibility and feeding value of wheat straw in with urea treatment. In the trial the effects of inoculation of P. florida, and 2% urea treatment were examined in a 2 by 2 factorial design. The mushroom was inoculated to urea supplemented or unsupplemented wheat straw by solid-state fermentation. Following inoculation, on the 20th, 40th, 60th and 80th days straw samples were taken and analysed for nutrient content and tested for digestibility on sheep using the nylon bag technique. One hundred pneumonic lungs from calves were examined by an immunoper oxidase technique using avidin-biotin- per oxidase complex method in formalin-fixed, paraffin-embedded sections for Haemophilus somnus. In the histological examination of tissue sections, strong positive reactions against H. somnus were detected in 14% of the lungs. H. somnus antigens were observed at the surface and/or within the epithelial cells, macrophages, necrotic alveolar walls, alveolar -bronchial/br onchiolar exudates, fibrins, distended interlobular septa and perivascular areas. In addition, dense antigen accumulations were detected in the necrotic macrophages and neutrophils.

The objective of this study is to compare meat yield and chemical composition of blue crabs and sand carbs. The ratio of crude protein (18.93%), dry matter (22.43%) and crude ash (2.34%) in the meat taken from male individuals were found to be higher in sand crabs than in blue crabs, but lipid levels were not statistically different for male individuals of the two species (p>0.05). In this study, the qualitative and quantitative aspects of aerobic bacterial flora of the skin, gills, intestine, liver, kidney and spleen of rainbow trout in relation to pond water in two different fish farms were analysed. The bacterial load of pondwater in Seniçbey Farm was 10 2 -10 5 cfu/ml. It was determined that the total viable counts of aerobic microorganisms of pond water in this farm were higher than these in Aksu Farm (10 2 -10 4 cfu/ml). This study was conducted to investigate the levels calcium, magnesium, sodium and potassium in the seminal plasma of bulls, semen volume, sperm motility, sperm concentration and the correlation between these parametres. Five Holstein bulls constituted the material of this study. Twenty ejaculates were collected from each bull and the spermatological characteristics of these ejaculates were determined. Then, seminal plasma was separated from 3ml semen from each ejaculate by centrifugation at 1500g for 20 min at 4 °C. The effects of 0%, 10%, 20% and 30% dried poultry excreta supplementation on rumen fermentation and blood parameters in Akkaraman lambs were investigated in the control group, group I, group II and group III respectively. Six different doses of PGF2a were injected into uterine tissue samples obtained from three groups using an isolated-organ bath. The response of the uterine muscles to PGF2a was recorded. This research was carried out to determine the amount of concentrate and hay consumed during the milk feeding period and the weaning age of Brown Swiss and Holstein calves. Twenty-five Brown Swiss and twenty Holstein Friesian calves were used. Calves reached 500 g and 1000 g/daily calf starter feed consumption at 36.5 ± 1.8 and 53.8 ± 1.2 days-old respectively. A feeding trial was conducted to assess the effects of supplementing the diets of commercial-size rainbow trout (Oncorhynchus mykiss) with red pepper as a natural carotenoid source and synthetic astaxanthin. Diets without carotenoid supplemented (control) and containing 3% or 6% red pepper and 0.05% "Carophyll Pink 8%" were given to groups 1-4, respectively. Rainbow trout, with weight of groups between 470 and 483 g, were fed four different diets for 45 days. In this research, the effects of early weaning on the growth rate and milk production characteristics of Brown Swiss cattle in later years were investigated. The calves used in the study were weaned at 5, 7 and 9 weeks of age, and the weights obtained at 6, 9, 12, 15 and 18 months of age were not significantly affected by the age of weaning. Modified tryptic soy broth, lauryl sulfate tryptose broth, modified EC broth, sorbitol MacConkey agar and Fluorocult E. coli O157 agar were tested for the recovery of 3 strains of Escherichia coliO157:H7 against E. colitype 1 and Citrobacter freundii as common competitive microflora of raw meat products. Two-stepped trials showed that modified EC broth and lauryl sulfate tryptose broth were superior to modified tryptic soy broth as selective enrichment media while sorbitol MacConkey agar was clearly better than Fluorocult E. coli O157 agar as a selective agar medium. Protein bands of hydatid fluids of sheep liver were determined by SDS-PAGE and Western blotting, and this was followed by the determination of specific bands of hydatid cysts for sheep and humans. For diagnostic purposes, 30 positive and 20 negative sheep sera, 1 commercially obtained non-infected sheep serum, and 10 posi- tive and 10 negative human sera were used in this experiment. The effect of halothane anesthesia in different doses and a period of two weeks on glucose-6-phosphate dehydrogenase (G6PDH) activity of mouse liver were investigated. The mean and SD of G6PDH for liver of normal mice were 0.036±0.017 U/mg protein and 1.756±0.814 U/g liver, respectively. Although there was histopathological damage in the liver, liver weights in treated mice increased significantly compared to control animals. Increasing the dosage of halothane inhalation induced liver G6PDH enzyme activity (U/g liver) by 84%, 42% and 4%, showing an inverse relation with the dose of halothane (0.25, 0.50, 1.00 cc twice daily for two weeks). The Monthly Variations In The Blood Glucose Level Of Capoeta Capoeta Capoeta (G.Ldenstaedt, 1772) Living In Aras River, Seyi̇t Aydin, Ayhan Yildirim, Orhan Erdoğan

In this study, the monthly variation in the blood glucose level of Capoeta capoeta capoeta caught from Aras between August 1995 and June 1996 was determined. The average blood glucose level was determined to be 112.04±5.19 mg/dl in males and 106.32±9.68 mg/dl in females and 110.06±4.19 mg/dl in the population. The blood glucose level was highest (141.07±13.67 mg/dl) in April, and lowest (69.14±9.75 mg/dl) in September. It was deter- mined that the level of blood glucose was related to temperature and reproduction.

Toxoplasma gondii pseudocysts in cattle are sources of infection for humans. Therefore, cattle toxoplasmosis is a very important factor in public health. The aim of the present study was to determine the seroprevalence of cattle toxoplasmosis. A total of 122 cattle from different age groups were selected in Elazığ and environs. Blood samples were taken and sera were collected. Serum samples were kept at -20^0 C until further examination. The values of toxoplasmosis seroprevalence were determined by SF test for anti-Toxoplasma gondii antibodies. Antibodies were detected in 86 out of 122 cattle sera (70.49%). In the first step, semen samples were diluted with Tris+Citric acid+Fructose+Egg yolk and Sadium citrate+Egg yolk diluents and were frozen in liquid nitrogen vapour. In the second step, 120 Merino ewes were synchronised with sponges, which contained flourogestone acetate, for 14 days. PMSG was injected into the ewes on the withdrawal day. In this study mutagenic effects of some chemicals used in Veterinary Medicine and agricultural fields was investigated by Somatic Mutation and Recombination Test (SMART). SMART test was to observe the chemical effects on wing phenotype of the trans- heterozygote flies carrying marker gene. Lethal doses of the chemicals used were determined. A positive correlation was observed between total mutation and the number of wings having mutations. In addition, the observed mutations were classified according to the size and the type of the mutations per wing. In this study, the heart structures of 30 English racing horses with different weights were examined by M-mode echocardiography. Horses were separated into 3 groups of 10 horses. The aim of this study was the determination of the shelf life of marinated fish balls. The anchovies were boiled, and after bones and heads were removed they were mixed with spice and fried. Then, they were placed in brine and stored at +4^0 C (± 1). The sensory analyses pH, TVB-N and TMA-N analyses were performed every 15 days during the storage. The results showed that this product was excellent quality for 60 days, fair quality for 105 days, and was marketable until the 120^th day. The spoilage was determined after the 120 ^th day.

Multiple regression analysis was utililised to estimate the relationship between somatic cell counts (SCC) and milk yield using the data of 1687 quarter milk samples collected weekly from 20 cows at the Dairy Unit of A.U. Veterinary Faculty's Research Farm between May and October 1998. The findings of this study supported the results of similar previous studies on that there is a negative relationship between SCC and milk yield, the relationship between milk yield and lnSCC (natural logarithmic form) is not linear.
